Clinical Research Challenges: Insight From A Pilot Study At An Academic Healthcare Center, Gretchen E. Maurer Dec 2013

Clinical Research Challenges: Insight From A Pilot Study At An Academic Healthcare Center, Gretchen E. Maurer

PCOM Biomedical Studies Student Scholarship

An investigational clinical research study was conducted at an academic healthcare center evaluating memantine as an adjunct to opioid therapy for treatment of chronic low back pain. The N-Methyl-D-Aspartate (NMDA) receptor is located in pain signaling neuronal synapses of the central nervous system. The receptor binds the excitatory neurotransmitter glutamate in addition to NMDA, to increase the magnitude of the perception of pain. Memantine (Namenda©) is a highly tolerated NMDA receptor antagonist which is currently prescribed in the treatment of Alzheimer’s disease. The purpose of the non-randomized pilot study without placebo was to evaluate the use of adding memantine as …

Comparison Of Electrogustometrically Determined Taste Threshold And Phenylthiocarbamide Sensitivity Between Non-Diabetic Subjects With First Degree Relatives With Type 2 Diabetes And Non-Diabetic Subjects Without Type 2 Diabetic First Degree Relatives, Matthew E. Joseph Do

PCOM Biomedical Studies Student Scholarship

Diabetes is a systemic degenerative disease, having adverse effects on many different organ systems. Nerve conduction specifically related to taste is one of the affected functions and can lead to altered taste perceptions and taste thresholds. Electrogustometry was used in this study to evaluate taste thresholds, and taste sensitivity was tested using phenylthiocarbamide-impregnated testing strips. Healthy non-diabetic subjects were divided into two experimental groups: One with a first degree relative with Type 2 diabetes and the other matched controls having no family history of Type 2 diabetes. The hypotheses: 1). There is a significant difference in taste threshold values and …

Resveratrol Induced Apoptosis In A Human Adenosquamous Carcinoma Cell Line (Cal-27 Cells), Saquib A. Siddiqi Oct 2013

Resveratrol Induced Apoptosis In A Human Adenosquamous Carcinoma Cell Line (Cal-27 Cells), Saquib A. Siddiqi

PCOM Biomedical Studies Student Scholarship

Radiotherapy and surgery are the two principal modalities in the treatment of head and neck cancers, and both therapies can result in severe adverse effects and ultimately lower the quality of life. It is of paramount importance to develop reagents that target the cancer cell specifically without affecting the normal non-cancer cells. Using the tongue cancer cell line Cal 27 as a model system, we dissected the molecular mechanism of the resveratrol-induced cancer cell apoptosis. After demonstrating that resveratrol induces the cancer cell apoptosis in a dose- and time-dependent manner, a systemic apoptosis protein array was conducted to identify the …

Regulation Of Alpha Synuclein Following Chronic Methamphetamine Administration In Guinea Pigs: Correlation With Memory And Synaptic Plasticity, Adnouse Blanc Jan 2013

Regulation Of Alpha Synuclein Following Chronic Methamphetamine Administration In Guinea Pigs: Correlation With Memory And Synaptic Plasticity, Adnouse Blanc

PCOM Biomedical Studies Student Scholarship

Methamphetamine (METH) is a highly addictive drug of abuse that has a severe impact on neuronal changes in the brain including modulations of plasticity, cognitive dysfunction, as well as memory impairment. These changes can be seen as modifications in the expression of biochemical markers associated with synaptic plasticity. One such marker associated with memory impairment is alpha synuclein (α-syn). Alteration of α-syn expression has been linked to memory impairment in patients with Alzheimer’s disease (AD) and Parkinson’s disease (PD). Here we assess the effect of chronic METH treatment in correlation to cognitive functions.

Twenty-nine guinea pigs (male, 150-250 g) were …

Molecular Mechanisms Related To Endotoxemia In Primary Human Cardiomyocytes In Culture, Atijah J. Collins Jan 2013

Molecular Mechanisms Related To Endotoxemia In Primary Human Cardiomyocytes In Culture, Atijah J. Collins

PCOM Biomedical Studies Student Scholarship

Cardiovascular disease (CVD) is the leading cause of death in the United States and has been for the past 80 years. Development of novel therapeutic agents to address the large number of CVD deaths requires an in depth understanding of the structural and functional properties of human cardiomyocytes. Over the last few years we have been developing an in vitro paradigm to assess molecular cardiodynamics in Primary Human Cardiomyocyte in culture (PHCC). We tested the hypothesis whether endotoxemia would exhibit a marked decrease in contractile proteins and cause apoptosis in PHCC. In the current series of experiments, we induced endotoxemia …

Cardioprotective Effects Of Mitochondrial-Targeted Antioxidants In Myocardial Ischemia/Reperfusion (I/R) Injury, Regina Ondrasik Jan 2013

Cardioprotective Effects Of Mitochondrial-Targeted Antioxidants In Myocardial Ischemia/Reperfusion (I/R) Injury, Regina Ondrasik

PCOM Biomedical Studies Student Scholarship

Reactive oxygen species (ROS) generated during myocardial I/R contribute to post-reperfused cardiac contractile dysfunction. Damaged cardiomyocyte mitochondria are major sites of excess ROS generation during reperfusion. We hypothesized that reducing mitochondrial ROS formation should attenuate myocardial I/R injury and thereby improve function of isolated perfused rat hearts subjected to I(30min)/R(45min) compared to untreated I/R hearts. Mitoquinone (MitoQ, MW=579g/mol; complexed with cyclodextrin (MW=1135g/mol) to improve water solubility, total MW=1714g/mol), a coenzyme Q derivative, and SS-31 (Szeto-Schiller) peptide ((D-Arg)-Dmt-Lys-Phe-Amide, MW=639g/mol, Genemed Synthesis, Inc., San Antonio, TX), an alternating cationic-aromatic peptide, are selective mitochondrial ROS inhibitors which significantly improved post-reperfused cardiac function compared …
