Genome Sequence Of Lactobacillus Pentosus Kca1: Vaginal Isolate From A Healthy Premenopausal Woman, Kingsley Anukam

The vaginal microbiota, in particular Lactobacillus species, play an important role in female health through modulation of immunity, countering pathogens and maintaining a pH below 4.7. We report the isolation and genome sequence of Lactobacillus pentosus strain KCA1 (formally known as L. plantarum) from the vagina of a healthy Nigerian woman. The genome was sequenced using Illumina GA II technology. The resulting 16,920,226 paired-end reads were assembled with the Velvet tool. Contigs were annotated using the RAST server, and manually curated. A comparative analysis with the available genomes of L. pentosus IG1 and L. plantarum WCFS1 showed that over 15% …

Key Points • Alleviating the problems of maternal and child health in countries with large malnourished and/or HIV-infected populations, will require multidimensional approaches including holistic and pharmaceutical interventions. • Urogenital infections have a major role in preterm labor and the well-being of newborns. • Probiotics show potential in reducing the risk of recurrence of urinary tract infection (UTI). • There is some evidence to suggest that probiotics can have a role to play in bacterial vaginosis (BV) and maternal health. • The role of probiotics in a number of newborn, and childhood diseases is reviewed in this chapter.

Background: Bacterial vaginosis (BV) is a condition with diverse etiology. This condition predisposes women to increased susceptibility to sexually transmitted diseases, including human immunodeficiency virus (HIV) infections and preterm birth. The diagnostic methods currently adopted in the evaluation of patient samples for BV are arguably Amsel criteria, and Nugent score that require microscopy and expert interpretation. These two methods are still subjective. OBJECTIVE: The objective of this study was to determine whether Lactobacillus species found in African women differ substantially to those of Caucasian decent, described in previous studies. The vaginal microbiota play an important role in female health, and when the naturally dominant lactobacilli are displaced resulting in bacterial vaginosis (BV), the host is more at risk of acquiring sexually transmitted diseases, including HIV. METHODS: Vaginal samples were collected from 241 healthy, pre-menopausal Nigerian women, which were then Gram stained for Nugent scoring. Lactobacilli are ubiquitous in nature and in humans they play a very significant role in the general health maintenance of the host. Identification of Lactobacilli has previously been based on cultutre-dependent methods and recently molecular techniques involving gene sequencing are now the ‘gold standard’. Scarce information exists in Africa on the real identity of Lactobacillus species, albeit phylogenetic distances among the species present in the human vagina. 185 vaginal swabs were collected from healthy premenopausal women (18-48 years). The recent papers by Nishijima et al 1 and Morelli et al.2 not only confirmed our previous studies that orally administered lactobacilli can confer benefits on vaginal health, but began to uncover new concepts of disease prevention. Each day, over 7,000 women acquire HIV, a disease whose prevalence is significantly raised by absence of lactobacilli from the vagina3. In addition, over 23,000 babies (conservative 7% estimate) are born premature each day, and although it’s difficult to know how many are associated with bacterial vaginosis (BV), it could represent over 2,000 babies per day. The potential use of probiotics in restoring the urogenital and gastrointestinal health has received tremendous interest in the last decade, while few safety concerns are still being debated. In order to demonstrate the safety of the probiotic strains, Lactobacillus rhamnosus GR-1 and Lactobacillus fermentum RC-14, we designed a study in an animal model to investigate the effects of feeding probiotics on the haematological parameters of rats. The rats were fed daily with 109CFU/ml of the probiotic strains orally with an oro-gastric tube for 21 days.
