Ultrastructural Changes In Porcine Oocytes After Ultrarapid Freezing, Adisorn Adirekthaworn, Kriengyot Sajjarengpong, Siripen Vethchagarun, Wimon Pothiwong, Mongkol Techakumphu

The Thai Journal of Veterinary Medicine

The objective of the present study was to investigate the ultrastructural changes seen in frozen porcine oocytes at the germinal vesicle and during in vitro maturation when using an ultrarapid freezing technique. The morphological changes were assessed by transmission electron microscopy (TEM). Immature oocytes were collected from pig ovaries at a local slaughter house in Nakorn Prathom province. Some oocytes were matured in vitro for 44 hours at 39oC, 5% CO2 with high humidity. In the pig, sperm transport is regulated, to facilitate normal fertilization of the ovulated oocytes, by the oviduct which contains a particular segment, the sperm reservoir. The main function of the sperm reservoir is to ensure that stored spermatozoa have maintained viability and fertilizing capacity. The process of sperm capacitation is delayed in the sperm reservoir, during the lengthy pre-ovulatory estrus by interactions between the spermatozoa, the oviduct epithelium and secretion products. Twenty, 3-week-old, crossbred pigs from a herd free from porcine reproductive and respiratory syndrome virus (PRRSV) were inoculated intranasally and intramusculary (1 ml each 102.5 TCID50/ml) with Thai PRRSV isolates of the US genotype (01NP1) (n=9) or the EU genotype (02SB3) (n=8). Three pigs were used as negative controls. Three infected and one control group pig were euthanatized 5, 9 or 15 days postinoculation (DPI) to detect and to compare the location and the amount of PRRSV-antigen present. Semi-quantitative analysis of the interleukin 10 (IL-10) in bronchoalveolar lavage cells (BALC), from pigs challenged with porcine reproductive and respiratory syndrome virus (PRRSV), was undertaken in this study. Twenty-one, three weeks old, crossbred pigs from a PRRSV-free commercial farm were separated into three groups, a control, non-infected group (3 pigs/group), and 2 groups (9 pigs/group), the first inoculated with a Thai-isolated European (O2SB3) strain and the second a Thai-isolated, North American (O1NP1) PRRSV strain. The objective of the present study was to investigate survival rates, growth rates, weaning weights and the effect of colostrum in low birth weight piglets. The study was made on 288 piglets with a birth weight below 1 kg. After the individual birth weights were measured, the piglets were divided into 2 treatment groups, a control group (the piglets received colostrums after birth from their dams according to the usual management practice) and a colostrum supplemented group (piglets received an extra 20 ml of colostrum within 12 hrs of birth). Infusion Of Pentobarbital And Fentanyl Combination For Long-Term Anaesthesia In Pigs, Nuh Kiliç, Wolf Erhardt

The cardiopulmonary and clinical effects of the infusion of pentobarbital sodium in combination with fentanyl for long-term anaesthesia in 9 pigs were investigated. Pigs were premedicated with i.m. atropine at a dose of 0.05 mg/kg and azaperon at a dose of 2 mg/kg. General anaesthesia was induced with i.v. thiopental sodium at a dose of 3.3 mg/kg. After induction of anaesthesia, the pigs were intubated and supplied with oxygen during anaesthesia. Anaesthesia was maintained with an i.v. infusion of pentobarbital-sodium at a dose of 45 mg/kg/h and fentanyl at a dose of 0.026 mg/kg/h. Arterial blood pressure, blood gas analyses, …
