Replacement Of Fetal Calf Serum With Synthetic Serum Substitute In The In Vitro Maturation Medium: Effects On Maturation, Fertilization And Subsequent Development Of Cattle Oocytes In Vitro, Hakan Sağirkaya, Mehmet Yağmur, Zekari̇ya Nur, Mustafa Kemal Soylu

Turkish Journal of Veterinary & Animal Sciences

The aim of this study was to investigate the possibility of using synthetic serum substitute (SSS) instead of fetal calf serum (FCS) in maturation medium to stimulate in vitro maturation (IVM), fertilization (IVF) and subsequent development of bovine oocytes. Ovaries were obtained from a local slaughterhouse. Selected oocytes were matured in tissue culture medium 199 (M-199) supplemented with 2 mM glutamine + 0.25 mM Na-pyruvate + 0.5 µg/ml of FSH + 5 µg/ml of LH + 25 µg/ml of gentamycin and 10% FCS or 10% SSS (FCS and SSS groups, respectively) for 22 h. Matured oocytes were fertilized in vitro …

The Effects Of Various Bsa Levels In Different Media On Development In In Vitro Culture Of Mouse Embryos, Mi̇that Evecen, Serhat Pabucçuoğlu, Serhat Alkan, İ. Kamuran İleri̇

Turkish Journal of Veterinary & Animal Sciences

Various BSA (bovine serum albumin) levels and media effects on development of 2-cell stage mouse embryos in in vitro culture were investigated in this 2 stage study. In the first stage, 1400 2-celled mouse embryos were cultured in 2 various BSA-bearing culture media for 96 h and directly observed microscopically. In the second stage, 1024 2-celled mouse embryos cultured for the same period and cell numbers (nuclei of cells) were determined by staining techniques. Embryos were cultured in both stages in M 16 and Whitten's media containing 0 (control), 0.3, 1, 3, 9, 18 and 36 mg/ml of BSA. The …

Production Of Chimeric Cattle Embryos By Reaggregation Of Blastomeres Obtained From Day 4 Bovine Embryos, Hakan Sağirkaya Jan 2004

Production Of Chimeric Cattle Embryos By Reaggregation Of Blastomeres Obtained From Day 4 Bovine Embryos, Hakan Sağirkaya

Turkish Journal of Veterinary & Animal Sciences

The aim of this study was to investigate the possibility of producing chimeric bovine embryos by reaggregating blastomeres of disaggregated day 4 bovine embryos. Oocytes were obtained from a slaughterhouse. Selected oocytes were matured, fertilized and cultured in vitro. Fertilization day was taken as day 0. Blastomeres of day 4 embryos were disaggregated and then pooled together. Chimeric embryos were randomly constructed by reaggregating blastomeres in empty zonae pellucidae. Non-chimeric handled control embryos were produced by transferring blastomeres from each embryo into their own empty zonae pellucidae. A total of 79 day 4 cattle embryos were used for disaggregation of …
