Appraisal Of Atp1b1 And Gstm3 Proteins As Freezability Factors In Buffalo Ejaculated Spermatozoa, Marvin Bryan Segundo Salinas, Thuchadaporn Chaikhun-Marcou, Phongsakorn Chuammitri, Anucha Sathanawongs

The Thai Journal of Veterinary Medicine

Sperm cryopreservation intensifies the use of assisted reproductive technologies in food animals. However, variations in freezability confront frozen-thawed semen applications. Hence, this groundwork aimed to identify the fundamental differences between high (HF) and low freezability (LF) ejaculated spermatozoa in swamp buffaloes, which were limitedly described for this agricultural species. Twelve (12) different samples of bubaline semen were collected, cryopreserved and categorized into freezability phenotypes using post-thaw sperm motility as the basis. Sperm kinematics and functional plasma membrane integrity were evaluated in HF and LF samples using computer-assisted sperm analysis (CASA) and hypo-osmotic swelling test, respectively. The relative concentrations of some …

Supplementation Of Extracellular Adenosine 5'-Triphosphate Improves Sperm Post-Thawed Quality In Thai Native Chickens, Phakjira Sanguansook, Junpen Suwimonteerabutr, Kaywalee Chatdarong

The Thai Journal of Veterinary Medicine

Thai native chicken meat is a high-quality protein source of unique flavor. Genetic preservation and breeding programs are beneficial to maintain purebred populations. Aiming at improving post-thawed sperm quality, this study is the first to apply extracellular adenosine 5'-triphosphate (ATPe) in freezing extender to preserve Thai native chicken sperm. Eight semen samples from eight Thai native chickens (n = 8) were included. Initial semen evaluation and sperm size measurement were performed. Beltsville poultry semen extender supplemented with 0 and 30 mM ATPe was served as a control and treatment group, respectively. After thawing, motility and motion characteristics were evaluated by …

The Thai Journal of Veterinary Medicine

Astragalus polysaccharides (APS) have strong antioxidant effects and may thus be useful for the preservation of semen at low temperaturesin vitro. However, the effects of APS on the quality of frozen-thawed Tibetan boar sperm have yet to be clarified. Here, the effects of APS on the quality and levels of genomic DNA methylation in Tibetan boar spermatozoawere assessed. Tibetan boar semen was cryopreserved in high-density tubules in freezing extender supplemented with 0, 0.2, 0.4, 0.6, or 0.8 g·L-1 APS to determine the optimal concentration of APS. Spermgenomic DNAmethylation levels of the three groups (Fresh grouprepresents freshspermatozoa, No add grouprepresents unsupplemented …

The Thai Journal of Veterinary Medicine

The objective of the present study was to compare the efficacy of three diluents on post-thaw motility, viability, and fertility of frozen chicken semen. Semen from Thai native cocks (Pradu Hang Dam; n=24) was collected, pooled, and randomly allocated into three groups, then diluted at a ratio of 1:3 (v:v) with Schramm, EK, and polyvinylpyrrolidone (PVP)-based medium. The diluted semen samples were cooled down to 5ºC and then mixed with Dimethylformamide (DMF) (6% v/v). Semen straws were subjected to cryopreservation using liquid nitrogen vapor method. For cryopreserved sperm quality assessment, the frozen semen straws were thawed, and motility was assessed …

Effects Of Cryopreservation And Leukodepletion On Morphological Index Of Red Blood Cells In Dogs, Deniz Aktaran Bala, Ibrahim Akyazi

The Thai Journal of Veterinary Medicine

Preservation of blood components for transfusion is a long-standing phenomenon. However, studies of cryopreservation of leukodepleted canine erythrocytes are quite limited and no studies are available with respect to morphological evaluation of erythrocytes during the cryopreservation process. The aim of this study was to determine morphological changes caused by cryopreservation, storage and leukocyte filtration. Healthy adult dogs meeting the criteria for blood transfusion were used in this study. Packed red blood cells (pRBCs) were obtained from each dog. The samples were divided into two groups. Leukocyte filtration was performed on one group of pRBCs. The other group received no filtration. …

Advanced Biotechnologies For Wildlife Fertility Preservation, Pierre Comizzoli Dec 2016

Advanced Biotechnologies For Wildlife Fertility Preservation, Pierre Comizzoli

The Thai Journal of Veterinary Medicine

Reproductive biotechnologies are critical tools for saving and maintaining endangered species. Some successes have been reported with the use and integration of artificial insemination (with fresh or frozen-thawed semen) in conservation programs. However, not a single species is currently managed through oocyte freezing or embryo-based technologies. This is primarily due to the lack of knowledge of species biology, as well as inadequate facilities, space, expertise, and funding needed for their successful application. More fundamental studies of animal reproductive biology as well as more fertility preservation options are needed with all parties involved (reproductive technologists, zoo biologists and conservationists) adopting parallel …

In the experiment, semen was either cooled to 5ºC after dilution or diluted with a pre-cooled diluents before cooling to 5ºC. Semen freezing was performed in liquid nitrogen vapor at -135ºC and -35ºC, with 6% DMF, and compared for sperm qualities. After thawing, it was measured for total sperm motility and progressive motile by CASA. …

The Thai Journal of Veterinary Medicine

Semen cryopreservation alters sperm structure and biochemical properties, and subsequently impairs fertilizing ability. A combination of two different sugar molecules revealed an enhanced beneficial effect on frozen-thawed spermatozoa of various species. However, the report in ram has been scarce. The present study investigated the effect of different combinations of sugar supplementation on post-thawed ram semen qualities in a cryopreservative extender in the presence of a single sugar or a combination of sugars. Data showed that a combination of sucrose and trehalose significantly improved cryopreserved sperm motility, viability, longevity and acrosome integrity. Fertility rates following laparoscopic artificial insemination (LAI) using frozen-thawed …

The laparoscopic artificial insemination was performed to test the fertilizing ability of frozen-thawed semen. The spermatozoa cryopreserved with 10% glycerol tended to have a higher percentage of motility, progressive motility, spermatozoa with intact acrosome …
