Functional 3-D Cellulose & Nitrocellulose Paper-Based, Multiplex Diagnostic Platforms Without Coupling Agents, Mackenzie Elizabeth Tageson

The purpose of this thesis was to demonstrate device functionality of 3-D paper-based, multiplex platforms, µPADs, without the use of coupling agents between layers. Previously, these platforms were fabricated with double-sided tape and cellulose powder to try to augment proper fluid routing, but difficulties with this method occurred. An acrylic housing unit with strategically placed pressure tabs was designed to aid horizontal and vertical fluid routing through the platform, thus eliminating the inconsistencies associated with coupling agents. Channel characterization studies, a COMSOLTM simulation, and development time studies were performed to aid device design and demonstrate device functionality.

Currently, there is weak evidence on the effectiveness of different rehabilitation regimens following distal radius fracture (DRF). This thesis evaluated sensory and vascular effects of exercise, thermal and ultrasound interventions that can be used in the mobilization phase (cast removal) after DRF.

Adolescence is a complex transitional period in human development, composing physical maturation, cognitive and social behavioral changes. The objective of this study is to investigate sex differences in white matter development and the associations between intelligence and white matter microstructure in the adolescent brain using diffusion tensor imaging (DTI) and tract-based spatial statistics (TBSS). In a cohort of 16 typically-developing adolescents aged 13 to 17 years, longitudinal DTI data were recorded from each subject at two time points that were one year apart. Two brain regions with established roles in reading are the posterior middle temporal gyrus and the posterior fusiform gyrus. Lesion studies have also suggested that the region located between them, the posterior inferior temporal gyrus (pITG), plays a central role in word recognition. However, these lesion results could reflect disconnection effects since neuroimaging studies have not reported consistent lexicality effects in pITG. Here we tested whether these reported pITG lesion effects are due to disconnection effects or not using parallel ERP/fMRI studies. Pharmacological challenge imaging has mapped, but rarely quantified, the sensitivity of a biological system to a given drug. We describe a novel method called rapid quantitative pharmacodynamic imaging. This method combines pharmacokinetic-pharmacodynamic modeling, repeated small doses of a challenge drug over a short time scale, and functional imaging to rapidly provide quantitative estimates of drug sensitivity including EC50 (the concentration of drug that produces half the maximum possible effect). We first test the method with simulated data, assuming a typical sigmoidal dose-response curve and assuming imperfect imaging that includes artifactual baseline signal drift and random error. Several detection methods have been explored for photoacoustic and ultrasound imaging of biological tissues. Piezoelectric transducers are commonly used, which require contact with the sample to be imaged and have limiting bandwidth characteristics. Interferometry detection exhibits improved bandwidth characteristics and resolution, yet generally require complicated optics and the incorporation of a contacting reflective medium. In this paper, we report the use of a noncontact photoacoustic imaging system that does not require the use of a reflective layer. A simple, robust technique known as gas-coupled laser acoustic detection is used, which has previously been applied to evaluation of composite materials. This …

Objective: This paper presents continued research toward the development of a knowledge-based system for the diagnosis of human toxic exposures. In particular, this research focuses on the challenging task of diagnosing exposures to multiple toxins. Although only 10% of toxic exposures in the United States involve multiple toxins, multiple exposures account for more than half of all toxin-related fatalities. Using simple medical mathematics, we seek to produce a practical decision support system capable of supplying useful information to aid in the diagnosis of complex cases involving multiple unknown substances.

Osteoporosis is identified by reduced bone mass, decreased bone quality and altered bone micro-architecture. These traits are quantitative and also highly regulated by genetic and environmental factors. As a powerful tool for collaborative analysis of quantitative traits and gene function, BXD recombinant inbred (RI) lines mice have been used to identify genetic effects for bone density.
For the first time, we investigated the bone properties of BXD RI mice by analyzing femur and tibia and compared their phenotypes of different compartments. The purpose of this study was to design, synthesize and develop novel transporter targeting agents for image-guided therapy and drug delivery. Two novel agents, N4-guanine (N4amG) and glycopeptide (GP) were synthesized for tumor cell proliferation assessment and cancer theranostic platform, respectively. N4amG and GP were synthesized and radiolabeled with 99mTc and 68Ga. The chemical and radiochemical purities as well as radiochemical stabilities of radiolabeled N4amG and GP were tested. In vitro stability assessment showed both 99mTc-N4amG and 99mTc-GP were stable up to 6 hours, whereas 68Ga-GP was stable up to 2 hours. Cell culture studies …

Approximately 2 to 3 million people in the United States suffer from chronic wounds, which are defined as wounds that do not heal in 30 days time; an estimated $25 billion per year is spent on their treatment in the United States. In our work, we focused on treating chronic wounds with bacterial infections using hyperbaric and topical oxygen therapies.
We used a mathematical model describing the interaction between bacteria, neutrophils and oxygen. Optimal control theory was then employed to study oxygen treatment strategies with the mathematical model. Existence of a solution was shown for both therapies. Uniqueness was also …

In this study we focused on direct comparison between the spatial distributions of activation detected by functional magnetic resonance imaging (fMRI) and localization of sources detected by magnetoencephalography (MEG) during identical language tasks. We examined the spatial concordance between MEG and fMRI results in 16 adolescents performing a three-phase verb generation task that involves repeating the auditorily presented concrete noun and generating verbs either overtly or covertly in response to the auditorily presented noun. Objetivo: encontrar una relación entre los síntomas y signos hallados al valorar la función lagrimal y la superficie ocular con pruebas diagnósticas de ojo seco, en un grupo de sujetos colombianos y españoles aparentemente sanos. Método: un total de cien sujetos sanos, cincuenta de población colombiana y cincuenta de población española sin historia previa de lentes de contacto se reclutaron y evaluaron. A patient specific image planning system (IPS) was developed that can be used to assist in kV imaging technique selection during localization for radiotherapy. The IPS algorithm performs a divergent ray-trace through a three dimensional computed tomography (CT) data set. Energy-specific attenuation through each voxel of the CT data set is calculated and imaging detector response is integrated into the algorithm to determine the absolute values of pixel intensity and image contrast. Phantom testing demonstrated that image contrast resulting from under exposure, over exposure as well as a contrast plateau can be predicted by use of a prospective image planning …

La cuantificación de una ametropía se hace por medio de técnicas de refracción. Objetivo: determinar la fiabilidad de la retinoscopia dinámica monocular (RDM) de Merchán en la medición de los defectos refractivos. Métodos: en un estudio descriptivo de concordancia se evaluaron 110 sujetos (220 ojos) entre los dieciocho y los treinta años de edad con la RDM, la retinoscopia estática (RE), el cilindro cruzado de Jackson (JCC) y el neto de la RDM obtenido compensando el LAG de la retinoscopia de Nott (RDMc), con medición independiente por parte de dos evaluadores en dos sesiones diferentes. Objetivo: determinar los valores de referencia de amplitud de acomodación mediante la medición objetiva dinámica de la amplitud de acomodación (MODAA) en personas preprésbitas emétropes de 35 a 40 años de edad, examinadas en la Clínica de Optometría de la Universidad de La Salle. Metodología: se incluyeron cincuenta pacientes emétropes, agudeza visual de lejos y de cerca con cada ojo igual o mejor a 20/30, binocularidad normal. Objetivo: conocer los factores asociados a cada uno de los síntomas del síndrome de la visión del computador (CVS) y analizar sus principales causas. Métodos: revisión bibliográfica integrativa en las plataformas SciELO, Lilacs y Pubmed de los últimos treinta años. Resultados: fueron seleccionados 32 artículos que cumplieron los criterios de inclusión, en los cuales se encontró la categorización del CVS en diferentes tipos de síntomas, cada uno asociado a diferentes diagnósticos visuales u oculares, que incluían alteraciones vergenciales, acomodativas o alteraciones de la superficie ocular como ojo seco.
