Sympathetic Nervous System Reactivity In Women Following Preeclamptic Pregnancies, Nina Stachenfeld, Charlotte Usseleman, Cheryl Leone, Michael Paidas

Yale Day of Data

Women who have had preeclamptic pregnancies are at risk for life-long cardiovascular disease. However, the factors contributing to this risk have yet to be established. Sympathetic nervous system dysregulation has been proposed to contribute to cardiovascular dysfunction during preeclamptic pregnancies. Therefore, we examined muscle sympathetic nerve activity (MSNA) at baseline and during a chemoreflex stimulus in women 6-24 months postpartum following a preeclamptic pregnancy (PE; n=6, age 28±2 y, BMI 27±3 kg/m2, 17±4 months postpartum). We hypothesized that MSNA responses to apnea would be greater in PE relative to control subjects, that is, women 6-24 months following a healthy pregnancy …

Phylogenetic Estimates Of Hiv-1 Gp120 Indel Rates Across The Group M Subtypes, John Palmer, Art Poon Jun 2019

Phylogenetic Estimates Of Hiv-1 Gp120 Indel Rates Across The Group M Subtypes, John Palmer, Art Poon

Western Research Forum

Insertions and deletions (indels) in the HIV-1 envelope glycoprotein gp120 play a significant role in the evolution of HIV pathogenesis and transmission fitness. While substitution rates in HIV-1 are well characterized by phylogenetic models, there is a lack of quantitative measures of indel rates in HIV-1. Here we use a dated-tip phylogenetic analysis of gp120 sequences to estimate indel rates for 7 subtypes and CRFs of HIV-1 group M.

We obtained and processed 26,359 HIV-1 gp120 sequences from the Los Alamos National Laboratory HIV Sequence database. After filtering these sequences, we extracted the conserved and variable regions from the remaining …

Mouse Performance On A Novel Touchscreen Continuous Performance Task Is Dependent On Signaling In The Prelimbic Cortex, Tyler D. Dexter, Daniel Palmer, Amy C. Reichelt, Anita Taksokhan, Lisa M. Saksida, Tim J. Bussey

Western Research Forum

Attention is the cognitive processing that facilitates the ability to target and attend to relevant environmental stimuli, while filtering out irrelevant or distracting stimuli. Control over selective attention is theorized to be dependent on organized neural communication that stems from the medial prefrontal cortex (mPFC). To evaluate selective and sustained attention, mice were trained on the novel touchscreen rodent continuous performance task (rCPT), a task designed to emulate the human CPT. In the rodent version, images are continuously presented on a touchscreen, where mice have been trained to selectively respond to one image type while suppressing responses to all others. …

Cell-Free Dna Release During Programmed Cell Death In Ischemia Reperfusion Injury, Alexander Dionne, Anthony M. Jevnikar, Zhu-Xu Zhang Jun 2019

Cell-Free Dna Release During Programmed Cell Death In Ischemia Reperfusion Injury, Alexander Dionne, Anthony M. Jevnikar, Zhu-Xu Zhang

Western Research Forum

Transplantation is invariably associated with acute allograft injury caused by ischemia reperfusion injury (IRI). This injury causes cells of the allograft to undergo various forms of programmed cell death including apoptosis and necroptosis. During programmed cell death, immunogenic molecules are released from cells, one of which is cell-free DNA (cfDNA). We hypothesize that cfDNA is released by microvascular endothelial cells (MVECs) during programmed cell death of IRI and that cfDNA acts as both a biomarker for cellular injury as well as a biologically active molecule capable of amplifying inflammation and organ injury.

Our results indicate that cfDNA is released by …

Exercise To Combat Neurocognitive Decline In Older Adults, Joyla Furlano Jun 2019

Exercise To Combat Neurocognitive Decline In Older Adults, Joyla Furlano

Western Research Forum

Background: Older adults with type 2 diabetes (T2D) experience cognitive decline and neural atrophy, and therefore are at high risk for developing dementia. Consequently, older adults at-risk for developing T2D (i.e., overweight or pre-diabetic individuals) are at higher risk for cognitive decline, and intervening at this point may prevent or delay the onset of such decline. One promising lifestyle intervention that may improve neurocognitive function is exercise. For example, 6 months of aerobic training improves cognitive function in overweight or pre-diabetic older adults, but research has not examined whether resistance training (RT) can produce comparable results in this population.

Selenomethionine Protects Mutant Tau N27a Cell From Oxidative Stress And Decreases Phosphorylation Of Tau, Madelin Locicero May 2019

Selenomethionine Protects Mutant Tau N27a Cell From Oxidative Stress And Decreases Phosphorylation Of Tau, Madelin Locicero

Celebration of Learning

Alzheimer’s Disease (AD) is neurodegenerative disease characterized by loss of cells and aggregated tau in the hippocampus. Hyperphosphorylated tau causes the misfolding of tau proteins (aggregated tau), which are responsible for proper neuronal activity. Selenium-methionine has been known to decrease the amount of phosphorylated tau within mutant and wild type 3xTg- AD mice. However, selenium can be toxic to cell depending on the type of cells and the concentration of selenium. The purpose of this study is to determine the effect selenium-methionine has on decreasing hyperphosphorylation of tau, protecting cells from oxidative stress, and decreasing the amount of aggregated tau …

Repetitive Mild Traumatic Brain Injury Impairs Performance In A Rodent Assay Of Cognitive Flexibility, Christopher P. Knapp, Doug P. Fox, Ramesh Raghupathi, Laura L. Giacometti, Stan B. Floresco, Barry D. Waterhouse, Rachel L. Navarra

Rowan-Virtua Research Day

Mild traumatic brain injury (mTBI) occurs in almost 80% of the 3 million reported cases of TBI-related emergency department visits each year in the United States. The majority of mTBIs, sometimes classified as concussions, are due to sports-related activities and typically occur repeatedly over the course of an athlete’s career. mTBI symptoms are generally classified as either somatic or neuropsychiatric/cognitive in nature and include impairments in prefrontal cortex mediated functions, including attention, memory, processing speed, reaction times, problem solving, and cognitive flexibility. To date, there remains a major gap in our understanding of the behavioral manifestations, underlying neurobiology, and treatment …

Amiodarone Induced Epididymo-Orchitis, Yaser Khalid D.O., Neethi Dasu D.O., Sravan Panuganti D.O., Stephen Daly D.O.

Rowan-Virtua Research Day

Amiodarone has a well-established and extensive side effect profile: pulmonary fibrosis, thyroid toxicity, corneal deposits, skin discoloration. However, in some rare instance, epididymitis/orchitis is a side effect of amiodarone. Amiodarone toxicity was first studied in (1, 2) Symptoms range from testicular pain to swelling and erythema (1,2) The mechanism of how this toxicity occurs is unknown. In this case report, we will discuss the case of an elderly patient who developed epididymitis and orchitis after several years of tolerating amiodarone without any adverse events. Our patient underwent a full workup with testicular ultrasound, evaluation by urology and cardiology specialists. Then …

A Mathematical Investigation On Tumor-Immune Dynamics: The Impact Of Vaccines On The Immune Response, Jonathan Quinonez, Neethi Dasu, Mahboobi Qureshi

Rowan-Virtua Research Day

Mathematical models analyzing tumor-immune interactions provide a framework by which to address specific scenarios in regard to tumor-immune dynamics. Important aspects of tumor-immune surveillance to consider is the elimination of tumor cells from a host’s cell-mediated immunity as well as the implications of vaccines derived from synthetic antigen. In present studies, our mathematical model examined the role of synthetic antigen to the strength of the immune system. The constructed model takes into account accepted knowledge of immune function as well as prior work done by de Pillis et al. All equations describing tumor-immune growth, antigen presentation, immune response, and interaction …

A Rare Manifestation Of A Bleeding Tubulovillous Duodenal Polyp Presenting As An Upper Gastrointestinal Hemorrhage, Neethi Dasu, Yaser Khalid, Herman Suga, Michael Itidiare, Richard Walters

Rowan-Virtua Research Day

• Duodenal polyps are a group of polyps that are fairly uncommon to find on endoscopic evaluation (1).

• They are histologically classified according to mucin phenotype into intestinal and gastric subtypes; the intestinal-type polyps are morphologically subdivided into tubular and tubulovillous adenomas (2)

• We present a case of a 76-year-old male with recurrent hematemesis who was found to have an intestinal-type pedunculated tubulovillous adenoma (TVA) in the descending duodenum • An isolated occurrence of non-ampullary sporadic duodenal adenomas (SDA)’s are a rare finding and presentation as an upper GI hemorrhage is extremely uncommon (3)

• Furthermore, our patient’s …

Exploration Of Prostate Cancer Cells: The Significance Of Active Herv, Dakota Shepherd Apr 2019

Exploration Of Prostate Cancer Cells: The Significance Of Active Herv, Dakota Shepherd

Student Scholar Showcase

The second most common cancer in men is prostate cancer. Prostate cancer is traditionally diagnosed by a digital rectal exam. Blood tests can also be used to test for PSA (prostate-specific antigen). These two methods can be used together but can sometimes provide both false negative and false positive results. A new method of testing for prostate cancer could prove to be beneficial. Previous studies have shown that active HERV sequences have been identified in some cancers, including prostate cancer. We hypothesize that the presence of active HERV in prostate cancer lines is significant. In this study, rabbit polyclonal antibodies …

For the purpose of this project, desired outcomes include correct compression rate and depth according to American Heart Association (AHA) guidelines, return of spontaneous circulation (ROSC) in the patient, long term survival of patients who achieve ROSC, and prevention of serious physical damage related to chest compressions. Data from multiple research studies comparing outcomes of mechanical compression devices and manual chest compression was reviewed …

Induction Of Lipid Bodies In Leishmania Infantum-Infected C57bl/6 Macrophages Of Male Versus Female Origin, Victor A. Soupene, Ryan D. Lockard, Mary E. Wilson, Nilda E. Rodriguez Apr 2019

Induction Of Lipid Bodies In Leishmania Infantum-Infected C57bl/6 Macrophages Of Male Versus Female Origin, Victor A. Soupene, Ryan D. Lockard, Mary E. Wilson, Nilda E. Rodriguez

Research in the Capitol

Worldwide, 12 million people are infected with Leishmania spp. parasites. Leishmania are transmitted by the bite of sand flies. The parasite is internalized by immune cells where they develop, reproduce, and spread to new immune cells causing disease. Epidemiological studies have shown that males are more likely to develop disease. We recently demonstrated that Leishmania infantum survive better in immune cells of male origin than in cells derived from females. We also showed that L. infantum infection is characterized by fat accumulation. Additional experiments have shown that infected immune cells from males accumulate more fat than cells from females. We …

The Effect Of Sepsis On Cerebral Microvascular Blood Flow, Laura Mawdsley, Mamadou Diop, Chris G. Ellis Mar 2019

The Effect Of Sepsis On Cerebral Microvascular Blood Flow, Laura Mawdsley, Mamadou Diop, Chris G. Ellis

Western Research Forum

Background: Sepsis is a dysregulated host response to infection that affects 18 000 000 people worldwide, and over 325 000 000 dollars are spent treating sepsis in Canada every year. One of the symptoms of severe sepsis is an altered mental state, which is accompanied with a measured decrease in oxygen levels in the skeletal muscle microvasculature. It is hypothesized that his altered mental state is due to a lack of oxygenated blood reaching the brain.

Hypothesis: After the onset of sepsis, microvascular cerebral blood flow and oxygen levels in the blood will decrease. Onset of decreased blood flow in …

Aortoduodenal Fistula Forms From Primary Aortic Stump Graft In A Two-Time Multi-Visceral Transplant Patient With Presentation Of Gastrointestinal Bleed And Bowel Perforation: A Case Report, Brielle Corrente

Graduate Student Research Symposium

Usually not diagnosed until open laparotomy, aortoduodenalfistulas (ADF) are one of the rarest complications of intestinal transplant surgery. With an incidence rate of only 0.04% at autopsy and only 250 documented cases since the early 1800’s, aortoduodenal fistulas are the most deadly complications of intestinal transplantation with a mortality rate of 100% without surgical intervention. A 39 year old, two-time multi-visceral transplant African American female patient suffered from a primary aortoduodenal fistula formation in a primary modified multi-visceral transplant aortic stump graft site. With emergency open laparotomy repair, revascularization of the secondary multi-visceral transplant was performed, saving the life of …

Updates On Epigenetic Alterations To Brca1: Chemosensitivities, Haley Blum Mar 2019

Updates On Epigenetic Alterations To Brca1: Chemosensitivities, Haley Blum

UNO Student Research and Creative Activity Fair

Breast cancer 1, early onset (BRCA1) is a human tumor suppressor gene encoding the BRCA1 protein that maintains genomic integrity. Molecular events may contribute to the loss of BRCA1 function, contributing to site specific tumorigenesis. Loss of BRCA1 function may arise from mutation or epigenetic events. Germline mutations of BRCA1 have been thoroughly implicated in heritable breast and ovarian cancers. More recently, sporadic breast cancer has been shown to be driven by epigenetic alterations to the BRCA1 promoter region, specifically methylation. Breast carcinomas that present with BRCA1 promoter methylation have been associated with triple negative breast cancers, as well …

The Effect Of Arm Swing On Countermovement Vertical Jump Performance, Arash Mohammadzadeh Gonabadi Mar 2019

The Effect Of Arm Swing On Countermovement Vertical Jump Performance, Arash Mohammadzadeh Gonabadi

UNO Student Research and Creative Activity Fair

Vertical jumping is one of the popular ways to evaluate ankle-knee efficiency in athletic population. Arm swing can play a crucial role in enhancing vertical jump performance. This study aimed to address the differences in kinetic and kinematic parameters during countermovement jump motion with arm swing (AS) and no arm swing (NAS). We used OpenSim to examine the efficacy of AS in reducing the impulse applied to the body and changes in range of lower limb joint angles at landing instant. We calculated the maximum vertical peak of the ground reaction force and impulse generated at landing in two different …

Estimating Variations In Metabolic Cost Within The Stride Cycle During Level And Uphill Walking, Arash Mohammadzadeh Gonabadi Mar 2019

Estimating Variations In Metabolic Cost Within The Stride Cycle During Level And Uphill Walking, Arash Mohammadzadeh Gonabadi

UNO Student Research and Creative Activity Fair

Indirect calorimetry provides the average cost of a stride cycle and prevents from identifying which part of the gait cycle causes increased metabolic cost in patients, however, recent simulation methods allow estimating the time profile of metabolic cost within the stride cycle. In this study, we compare the estimations of the time profile of the metabolic cost of two simulation methods for level and uphill walking. We used kinematic, kinetic and electromyography data from level and uphill walking (one participant) to estimate the time profiles of metabolic cost using the muscle-level metabolic model of Umberger using electromyography and kinematic data …

An Investigation Of Stimulants: Reviewing Their Effects On The Brain, Including Memory And Microglia, Michael Douchey Mar 2019

An Investigation Of Stimulants: Reviewing Their Effects On The Brain, Including Memory And Microglia, Michael Douchey

UNO Student Research and Creative Activity Fair

Stimulants are a class of drugs that have a variety of effects on the brain, including changes in receptor activity in the brain areas responsible for- memory, attention, emotion, motor control, and the reward pathway. Stimulants may be generally classified by their availability. For example, prescription stimulants, which include drugs such as methylphenidate (Concerta™, Ritalin™) and mixed amphetamine salts (Adderall™), must be prescribed by medical professionals. Another regulated stimulant, albeit one available without a prescription, is nicotine; a highly addictive chemical that is age-restricted by the U.S. federal government. Nicotine is found in tobacco products, and in tobacco-free alternatives including …

Impacts Of Dietary Nitrate On Endothelial Function, Arterial Stiffness, And Systemic Vascular Pressure In Peripheral Arterial Disease, Elizabeth J. Pekas, Steven D. Scott, Ronald J. Headid Iii, Michael D. Shukis, Jeonghwa Shin, Jiwon Song, Song-Young Park

UNO Student Research and Creative Activity Fair

Peripheral arterial disease (PAD) is the manifestation of atherosclerotic plaque in the larger arteries of the legs, which results in impaired blood flow to the lower extremities. Markers of vascular health, including endothelial function and arterial stiffness, have been shown to be attenuated in PAD.

Purpose: We sought to examine the effects of a nitrate supplement (body mass-normalized dosage of beetroot juice) on vascular function measurements such as endothelial function, arterial stiffness, and central and peripheral blood pressure in patients with PAD. We hypothesized that acute intake of a nitrate supplement would improve vascular function.

Methods: PAD patients (stage II-III, …
