Maternal obesity increases fetal adiposity which may adversely affect metabolic health of the offspring. Choline regulates lipid metabolism and thus may influence adiposity. This study investigates the effect of maternal choline supplementation on fetal adiposity in a mouse model of maternal obesity. C57BL/6J mice were fed either a high-fat (HF) diet or a control (NF) diet and received either 25 mM choline supplemented (CS) or control untreated (CO) drinking water for 6 weeks before timed-mating and throughout gestation. At embryonic day 17.5, HF feeding led to higher (p < 0.05) percent total body fat in fetuses from the HFCO group, while the choline supplemented HFCS group did not show significant difference versus the NFCO group. Similarly, HF feeding led to higher (p < 0.05) hepatic triglyceride accumulation in the HFCO but not the HFCS fetuses. mRNA levels of lipogenic genes such as Acc1, Fads1, and Elovl5, as well as the transcription factor Srebp1c that favors lipogenesis were downregulated (p < 0.05) by maternal choline supplementation in the HFCS group, which may serve as a mechanism to reduce fat accumulation in the fetal liver during maternal HF feeding. In summary, maternal choline supplementation improves indices of fetal adiposity in obese dams at late gestation.

While the FDA approval of targeted and immune therapies in metastatic melanoma (MM) have dramatically improved outcomes in this disease, de novo and/or acquired resistance can limit the clinical benefit of these agents. The IGF-1/PI3K/AKT pathway has been implicated in resistance to both targeted and immune therapy. The IGF-1/PI3K/AKT pathway has also been shown to play a key role in the pathogenesis of obesity in other malignancies. To date, the impact of energy balance on clinical outcomes and therapeutic response in MM has not been studied. A diagnosis of Metabolic Syndrome (MSX) requires patients to present with three or more of the following symptoms: elevated fasting blood glucose levels, elevated serum triglyceride levels, low serum HDL levels, elevated blood pressure, and truncal obesity. This pathology shares many similarities with Cushing’s Syndrome (CS) but a diagnosis of CS requires hypercortisolemia. The shared circuitries of depression and the stress response, regulated by the Hypothalamic-Pituitary-Adrenal (HPA) axis, implies that major depression may reflect dysregulations of the HPA axis. Previous studies find that that melancholic depression is accompanied by hyperactive HPA activity, anxiety, insomnia etc. Adversely, patients with atypical depression present CRH deficiency accompanied by lethargy and fatigue. According to the corticosteroid receptor hypothesis, HPA axis dysregulation that causes these variable phenotypic depressive moods may be attributed to defective GR expression or function. In recent years, dietary fat has been shown to be capable of activing taste receptor cells in the tongue. Fatty acids (FAs), which act as the chemical cue and are found in dietary fat, activate a cellular signaling pathway that results in a unique signal being sent to the brain that is then interpreted as the taste of fat. Obesity has become a serious health issue in the world (Ogden 2012), and its correlation to the human gut microbiota has been studied extensively since 2007 (Cani 2007). In 2013, researchers identified Enterobacter cloacae B29 from an obese patient’s gut and demonstrated its causal relationship to the host’s obese symptoms( Fei & Zhao 2013). However, there have not been many methods developed to alleviate obesity symptoms through eliminating pathogenic endotoxin-producing bacteria (such as E. cloacae B29), from the host’s gut flora. Obesity in women is increased by the loss of circulating estrogen after menopause. Shift work, which disrupts circadian rhythms, also increases the risk for obesity. It is not known whether ovarian hormones interact with the circadian system to protect females from obesity. During high-fat feeding, male C57BL/6J mice develop profound obesity and disruption of daily rhythms. Since C57BL/6J female mice did not develop diet-induced obesity (during 8 weeks of high-fat feeding), we first determined if daily rhythms in female mice were resistant to disruption from high-fat diet. Obesity is one of the most widespread nutritional diseases in developed societies and it is considered a cardiovascular disease risk factor. The aim of the present work was to evaluate how a high-fat diet may influence the chronobiology of glucose and albumin circadian rhythms.
