Regulation Of Learning And Memory By The Drosophila Melanogaster Swi/Snf Complex, Max H. Stone

The SWI/SNF complex is a highly-conserved ATP-dependent chromatin remodelling complex that is important in the etiology of intellectual disability (ID). I systematically investigated the overall and adult-specific roles of each of the 15 Drosophila melanogaster SWI/SNF complex components in memory. Flies with RNAi-mediated knockdown of individual SWI/SNF genes in the mushroom body (MB) were tested for short- and long-term memory impairment using courtship conditioning. Knockdown of several SWI/SNF genes through development, including brahma, Bap60, Snr1, and e(y)3, caused loss of memory. Adult-specific knockdown of SWI/SNF genes caused some loss of memory phenotypes, indicating an acute role in adult MB activity. …

Gene Expression Profiling In The Larval Fat Body Of Desiccation Selected Drosophila Melanogaster, Adriana Michelle Charles

Drosophila melanogaster selected for resistance to desiccation (no food or water) display slower development and a higher body mass compared to fed controls due to an extended third larval instar. I hypothesize that desiccation selected D. melanogaster larvae will have a different gene expression profile compared to fed controls. Separate populations of D. melanogaster were subjected to desiccation (no food or water), starvation (no food) until 80-85% mortality, for 75 generations. mRNA from the larval fat body was collected at 88 hours, 96 hours, 112 hours and 120 hours post-hatching. In Vivo Characterization Of The Drosophila Mrna 3’-End Processing Core Cleavage Complex: Poly(A) Mrna & Histone Mrna 3’-End Processing, Daniel Michalski

A core cleavage complex (CCC) consisting of CPSF73, CPSF100 and Symplekin is required for co-transcriptional 3’ end processing of all metazoan pre-mRNAs, yet little is known about the in vivo molecular interactions within this complex. The CCC is a component of two distinct complexes, the cleavage/polyadenylation complex and the complex that processes non-polyadenylated histone pre-mRNAs. RNAi-depletion of CCC factors in Drosophila culture cells causes reduction of CCC processing activity on histone mRNAs, resulting in read through transcription. In contrast, RNAi-depletion of factors only required for histone mRNA processing allows use of downstream cryptic polyadenylation signals to produce polyadenylated histone mRNAs. …

Cryptochromes are evolutionarily conserved blue-light absorbing flavoproteins which participate in many important cellular processes including in entrainment of the circadian clock in plants, Drosophila and humans. Drosophila melanogaster cryptochrome (DmCry) absorbs light through a flavin (FAD) cofactor that undergoes photoreduction to the anionic radical (FAD) redox state both in vitro and in vivo. However, recent efforts to link this photoconversion to the initiation of a biological response have remained controversial. Recent studies have linked neuropsychiatric disorders to older parents. These disorders often include changes in social behaviours like the social spacing between neighbouring individuals, which can be modeled in organisms such as Drosophila melanogaster. I investigated the effects of aging on the social space between neighbouring D. melanogaster and how aging impacts the next generation. To achieve this, I used the social space assay and found that individuals become less social with age and that this effect is passed on to the first generation only. Cold exposure appears to activate aspects of the insect immune system; however, the functional significance of the relationship between cold and immunity is unclear. Insect success at low temperatures is shaped in part by interactions with biotic stressors, such as pathogens, thus it is important to understand how and why immunity might be activated by cold. Here we explore which components of the immune system are activated, and whether those components differ among different kinds of cold exposure. Amyotrophic lateral sclerosis (ALS) is the most common adult onset motor neurodegenerative disease. The cause of the disease remains obscure, and as such there is no effective treatment or cure. Amyotrophic lateral sclerosis and other neurodegenerative diseases are frequently characterized by dysfunction of the RNA-binding protein, TDP-43. Using model systems to understand the mechanisms underlying TDP-43 dysfunction should accelerate identification of therapeutic targets. A recent report has shown that motor defects caused by the deletion of the Drosophila TDP-43 ortholog, tbph, are not driven by changes in the physiology at the neuromuscular junction. We have developed an undergraduate laboratory to allow detection and localization of proteins in the compound eye of Drosophila melanogaster, a.k.a fruit fly. This lab was a part of the undergraduate curriculum of the cell biology laboratory course aimed to demonstrate the use of Western Blotting technique to study protein localization in the adult eye of Drosophila. Lipocalin genes NLaz, GLaz, and Karl are evolutionarily conserved genes in Drosophila melanogaster. There are studies on lipocalin gene expression differences under diverse diet conditions, but these studies have focused mainly on age-dependent expression profiles of these genes. The main aim of our study is to determine lipocalin expression in the developmental period by nutritional manipulation with an isofemale-based design. Three larval developmental periods have been researched under normal and restricted diets. We found significant differences between lines during their developmental time-related lipocalin expression.
