Infertility is a widespread condition that does not always have a known cause, and for which we often do not have a cure. One potential cause of infertility is defects in gametogenesis, or the formation of sperm and egg. During gametogenesis in most organisms, the developing sperm and egg are connected to each other or to supporting cells through intercellular bridges, allowing transfer of materials between cells. Defects in these connections can lead to infertility. The developing fruit fly egg is an excellent model system to study intercellular bridges, or ring canals. Homeoviscous adaptation in membranes and changes in energy storage lipids such as triacylglycerols (TAGs) are the two main themes of adaptation to environmental conditions. Basal patterns of lipid compositional profile of Drosophila melanogaster along a latitudinal gradient in the eastern coast of Australia indicate variations of lipidome associated with climatic patterns. Lipidome data were collected for eight locations along the eastern Australian cline for both sexes in D. melanogaster and D. simulans. During embryogenesis, primordial germ cells (PGCs) and somatic gonadal precursor cells (SGPs) migrate and coalesce to form the early gonad. A failure of the PGCs and SGPs to form a gonad with the proper architecture not only affects germ cell development, but can also lead to infertility. Therefore, it is critical to identify the molecular mechanisms that function within both the PGCs and SGPs to promote gonad morphogenesis. We have characterized the phenotypes of two genes, longitudinals lacking (lola) and ribbon (rib), that are required for the coalescence and compaction of the embryonic gonad in Drosophila melanogaster. rib …

Hedgehog (Hh) signaling regulates multiple aspects of metazoan development and tissue homeostasis, and is constitutively active in numerous cancers. We identified Ubr3, an E3 ubiquitin ligase, as a novel, positive regulator of Hh signaling in Drosophila and vertebrates. Hh signaling regulates the Ubr3-mediated poly-ubiquitination and degradation of Cos2, a central component of Hh signaling. In developing Drosophila eye discs, loss of ubr3 leads to a delayed differentiation of photoreceptors and a reduction in Hh signaling. In zebrafish, loss of Ubr3 causes a decrease in Shh signaling in the developing eyes, somites, and sensory neurons. However, not all tissues that require …

This paper examines the effects of the chemical preservative bisphenol-A on the developing nervous systems of Drosophila melanogaster, or fruit fly, larvae. This study examines the effects of bisphenol-A using both behavioral as well as morphological paradigms. It was determined that bisphenol-A significantly increased the amount of time larvae spent in seeking behavior, the number of peristaltic contractions, and the distance traveled in a linear fashion. In the morphological analysis, indications of increased dendritic area in experimental larvae existed, but there was not enough data to determine significance. For future research, it is recommended that more data be gathered to …

Parsing out what makes some individuals more reproductively successful than others is a key pursuit in evolutionary biology. While reproductive success can ultimately be defined as the number of offspring produced over an individual’s lifetime, there are many selective episodes that shape this outcome. Because the majority of animals have multiple mates, achieving matings is but one influence on reproductive success. After copulation occurs, sperm from multiple males compete within the female reproductive tract to fertilize eggs, while females morphologically or behaviorally bias fertilization to preferred males, further shaping reproductive success. By examining genetic pathways in D. melanogaster, a better understanding of the homologous regulatory mechanisms in humans can be utilized to further enhance knowledge of the roles of microRNA within development. This study utilizes the UAS-Gal4 system in order to produce a mutant phenotype capable of being visually studied and analyzed, focusing on the developmental pathway of the wing in D. melanogaster. Dissections of the wandering third instar larvae yielded wing disc tissue expressing the downregulation of loquacious and CG17386.

The important pair-rule segmentation gene fushi tarazu (ftz) encodes a homeodomain (HD)-containing protein involved in the establishment of even-numbered parasegments during embryonic development. The D. melanogaster ftz is a derived homeotic selector (Hox) gene which lost its homeotic function during the evolution of arthropods. Genetic analyses have shown that FTZ has two distinct activities required during development: HD-dependent and HD-independent FTZ activities. The aim of this study was to test the interaction of the two FTZ activities proposed by Hyduk and Percival-Smith (1996), by generating site-specific mutant ftz alleles for intragenic complementation. Wnt/β-catenin signaling controls intestinal stem cell (ISC) proliferation, and is aberrantly activated in colorectal cancer. Inhibitors of the ADP-ribose polymerase Tankyrase (Tnks) have become lead therapeutic candidates for Wnt-driven cancers, following the recent discovery that Tnks targets Axin, a negative regulator of Wnt signaling, for proteolysis. Initial reports indicated that Tnks is important for Wnt pathway activation in cultured human cell lines. However, the requirement for Tnks in physiological settings has been less clear, as subsequent studies in mice, fish and flies suggested that Tnks was either entirely dispensable for Wnt-dependent processes in vivo, or alternatively, had tissue-specific roles. We describe a new method to measure the activation energy for unbinding (enthalpy ΔH*u and free energy ΔG*u) of a strongly-bound membrane-associated protein from a lipid membrane. It is based on measuring the rate of release of a liposome-bound protein during centrifugation on a sucrose gradient as a function of time and temperature. The method is used to determine ΔH*u and ΔG*u for the soluble dengue virus envelope protein (sE) strongly bound to 80:20 POPC:POPG liposomes at pH 5.5. Optogenetics is a revolutionary technique that enables noninvasive activation of electrically excitable cells. In mammals, heart rate has traditionally been modulated with pharmacological agents or direct stimulation of cardiac tissue with electrodes. However, implanted wires have been known to cause physical damage and damage from electrical currents. Here, we describe a proof of concept to optically drive cardiac function in a model organism, Drosophila melanogaster. We expressed the light sensitive channelrhodopsin protein ChR2.XXL in larval Drosophila hearts and examined light‐induced activation of cardiac tissue. Serotonin modulates various physiological processes and behaviors. This study investigates the role of 5-HT in locomotion and feeding behaviors as well as in modulation of sensory-motor circuits. The 5-HT biosynthesis was dysregulated by feeding Drosophila larvae 5-HT, a 5-HT precursor, or an inhibitor of tryptophan hydroxylase during early stages of development. The effects of feeding fluoxetine, a selective serotonin reuptake inhibitor, during early second instars were also examined. 5-HT receptor subtypes were manipulated using RNA interference mediated knockdown and 5-HT receptor insertional mutations. The master switch of the sexual differentiation and dosage compensation pathway in Drosophila is the sex lethal gene, Sxl. The early promoter, SxlPe, is activated in females, resulting in female-specific splicing of later transcripts (notably the late Sxl transcript SxlPm), while inactive in males. Chromatin immunoprecipitation (ChIP) assays have previously shown association of two conventionally heterochromatin-localized proteins, HOAP and HP1, at SxlPe, and in situ hybridization as well as RT-PCR assays have confirmed a repressive role for HOAP and both repressive and activating roles for HP1. Bacteria in modern society have often been perceived as having negative effects on humans with complications and potential death to their hosts. In recent years, the gut microbiota has shown that not all bacteria inhabiting a host cause negative side effects, but they instead can provide essential nutrients to their host and even directly impact growth rate and development. A Drosophila melanogaster genome-wide transcriptome dataset is
available for studies on temporal patterns of gene expression. Gene
expression was measured using two-dye color oligonucleotide arrays
derived from Version 2 of the Drosophila Genomics Resource Center.
A total of 15,158 oligonucleotide probes corresponded to a high
proportion of the coding genes in the genome. The source of the flies
was a highly genetically heterogeneous population maintained in an
overlapping generation population regime. This regime was
designed to maintain life history traits so that they were similar to

The ability to discriminate between prospective mates is potentially important not only for the avoidance of unsuccessful or incompatible matings, but also for the selection of higher “quality” mates. Inbreeding, reproduction between closely related individuals, has long enjoyed the attention of biologists, and has classically been associated with fitness consequences that are primarily negative, termed “inbreeding depression”. Indeed, the costs of mating with a relative are well documented across a wide variety of species. Anthropogenic lead (Pb) pollution is ubiquitous in the environment and a risk factor for both human and wildlife health. Pb exposure has the potential to alter reproductive strategies with respect to mate choice and reproductive output. This could be especially deleterious if these changes disrupt adaptive behavioral and reproductive life history strategies. Therefore, the overall aim of this body of work was to examine the consequences of developmental Pb exposure on reproductive strategies, using Drosophila melanogaster as a model system.
