Kisspeptin, a neuroendocrine regulator of gonadotropin releasing hormone, is hypothesized to integrate nutrition and hormones critical to metabolism and the regulation of reproduction. Since the negative energy balance of early lactation is associated with reduced fertility via suppression of gonadotropin secretion and enhanced growth hormone (GH) responsiveness, this experiment was designed to determine the effects of stage of lactation and negative energy balance on kisspeptin-10 (Kp-10) stimulated luteinizing hormone (LH) and GH concentrations. Five nonlactating [5.1 ± 0.8 (SEM) years; 577 ± 19 kg body weight (BW)] and five lactating [4.1 ± 0.6 years; 608 ± 11 kg BW] multiparous …

Forestry, Trees, and Timber
You are unique. The title “Forest Landowner” is one not held by many Tennesseans. Your ownership carries with it a sense of treasure, of enjoyment, of wildness. But it’s also a privilege, one that imparts responsibility, one that can be observed and one that increasingly can be measured.
If you were to be judged on your forestry practices, how would you score? Would you even know which parameters to assess or what questions to ask? Which of these forest uses aesthetics, recreation, soil, timber, water, wildlife—are more important? Have you ever intelligently, and intentionally, began a planning process for your …

Mutational Optimization Of The Coelenterazine-Dependent Luciferase From Renilla, Jongchan Woo, Albrecht G. Von Arnim
Faculty Publications and Other Works -- Biochemistry, Cellular and Molecular Biology
Renilla luciferase (RLUC) is a popular reporter enzyme for gene expression and biosensor applications, but it is an unstable enzyme whose catalytic mechanism remains to be elucidated. We titrated that one RLUC molecule can turn over about one hundred molecules of coelenterazine substrate. Mutagenesis of active site residue Pro220 extended the half-life of photon emission, yielding brighter luminescence in E. coli. Random mutagenesis uncovered two new mutations that stabilized and increased photon emission in vivo and in vitro, while ameliorating substrate inhibition. Further amended with a previously identified mutation, a new triple mutant showed a threefold improved k …

W192-Why Will My Weed Problem Not Go Away? 8 Common Reasons Why Herbicides Fail, The University Of Tennessee Agricultural Extension Service
Home Garden, Lawn, and Landscape
The frustration can be unsettling. ”Why won’t this weed die? I sprayed it with an herbicide, but for the life of me I can not figure out why it’s still here.” This is a very common story we hear from many people trying to control tough weeds in lawns, vegetable gardens, flower beds, nurseries, forests and agricultural fields. Many times just a small change in your approach to controlling the weed is all that is needed, but sometimes the problem can be a bit more complex. Whatever the weed control challenge, it is likely someone else has shared this same …

Pb1621-Best Management Practices For Planting Ornamental Plants, The University Of Tennessee Agricultural Extension Service
Home Garden, Lawn, and Landscape
While spring is a popular planting season, fall may be the best time to plant. Fall planting can help woody ornamental plants survive in the landscape. In fall, plant shoots need fewer nutrients because winter dormancy is approaching. In fall too, carbohydrate plant “food” is produced in leaves and moves to roots, which helps plant growth and survival. Importantly, roots continue to grow until soil temperatures drop below 45-50 F (7-10 C). It is extremely important to water when planting in fall because October and November are usually very dry months with little rainfall. But fallplanted plants also will not …

Pb1621-S-Prácticas Gerenciales Óptimas Para La Plantación De Plantas Ornamentales, The University Of Tennessee Agricultural Extension Service
Home Garden, Lawn, and Landscape

Background
Arsenic is toxic to plants and a common environmental pollutant. There is a strong chemical similarity between arsenate [As (V)] and phosphate (Pi). Whole genome oligonucleotide microarrays were employed to investigate the transcriptional responses of Arabidopsis thaliana plants to As (V) stress.
Results
Antioxidant-related genes (i.e. coding for superoxide dismutases and peroxidases) play prominent roles in response to arsenate. The microarray experiment revealed induction of chloroplast Cu/Zn superoxide dismutase (SOD) (at2g28190), Cu/Zn SOD (at1g08830), as well as an SOD copper chaperone (at1g12520). On the other hand, Fe SODs were strongly repressed in response to As (V) stress. Non-parametric rank …

Abstract
Background
The placenta is an important site for iron metabolism in humans. It transfers iron from the mother to the fetus. One of the major iron transport proteins is transferrin, which is a blood plasma protein crucial for iron uptake. Its localization and expression may be one of the markers to distinguish placental dysfunction.
Methods
In the experimental study we used antibody preparation, mass spectrometric analysis, biochemical and immunocytochemical methods for characterization of transferrin expression on the human choriocarcinoma cell line JAR (JAR cells), placental lysates, and cryostat sections. Newly designed monoclonal antibody TRO-tf-01 to human transferrin was applied …

Sp290-N The Greenbug In Grain Sorghum, The University Of Tennessee Agricultural Extension
Field & Commercial Crops
Greenbugs are small (1.6 mm long), soft-bodied insects that are oval in shape. Greenbugs usually feed in colonies by sucking sap from leaves. Feeding may cause mottling and discoloration of the plant. Heavily infested leaves turn red or yellow, shrivel and die. Damaging levels of greenbugs can result in the destruction of entire grain sorghum fields.

Hybridization Produces Novelty When The Mapping Of Form To Function Is Many To One, Nicholas F. Parnell, C Darrin Hulsey, J. Todd Streelman
Faculty Publications and Other Works -- General Biology
Background
Evolutionary biologists want to explain the origin of novel features and functions. Two recent but separate lines of research address this question. The first describes one possible outcome of hybridization, called transgressive segregation, where hybrid offspring exhibit trait distributions outside of the parental range. The second considers the explicit mapping of form to function and illustrates manifold paths to similar function (called many to one mapping, MTOM) when the relationship between the two is complex. Under this scenario, functional novelty may be a product of the number of ways to elicit a functional outcome (i.e., the degree of MTOM). …

Gene Alterations By Peroxisome Proliferator-Activated Receptor Gamma Agonists In Human Colorectal Cancer Cells, Maria Cekanova, X Li, J Yuan, K B. Kim, Seung J. Baek
Faculty Publications and Other Works -- Biochemistry, Cellular and Molecular Biology
The peroxisome proliferator-activated receptor gamma (PPARgamma) is a nuclear transcription factor that controls the genes involved in metabolism and carcinogenesis. In the present study, we examined the alteration of gene expression in HCT-116 human colorectal cancer cells by PPARgamma agonists: MCC-555 (5 microM), rosiglitazone (5 microM), and 15-deoxy-Delta12,14-prostaglandin J2 (1 microM). The long-oligo microarray data revealed a list of target genes commonly induced (307 genes) and repressed (32 genes) by tested PPARgamma agonists. These genes were analyzed by Onto-Express software and KEGG pathway analysis and revealed that PPARgamma agonists are involved in cell proliferation, focal adhesion, and several signaling pathways. …
