In their natural environments, photosynthetic organisms are often exposed to widely varied light environments. Species adapted to shade light, often found growing in lower layers of photosynthetic biofilms, must survive on filtered light alone. Filtered light is highly enriched in far-red wavelengths, which are normally unavailable for photosynthetic energy production in most oxygenic phototrophs. To overcome light limitations in filtered light environments, some species of algae and cyanobacteria utilize specialized photosynthetic pigments and antenna systems to harvest these far-red wavelengths. By sampling the natural environment and using custom-built far-red light growth chambers, I have isolated several species of oxygenic phototrophs …

Harmful algal blooms (HABs) and nuisance algal blooms (NABs) are a worldwide phenomenon with implications for human health and safety. HABs occur when algae or bacteria grow in high enough densities to harm animals and humans. A primary component of harmful algal blooms is cyanobacteria, which are aquatic, photosynthesizing microorganisms that produce toxins at high concentrations. Cyanobacterial biomass has increased worldwide in recent decades, raising concern about the future of fresh- and marine-water systems in a changing climate. Raceway ponds can be used to cultivate microalgae for purposes such as wastewater treatment and feedstock for animal feeds and biofuels. Besides nitrogen assimilation in biomass, nitrification can be a wastewater treatment mechanism but one that is hindered by the low nighttime dissolved oxygen concentrations (DO) common in wastewater treatment raceways. The large diel difference in pond conditions and changing influent wastewater quality also raises the question of the optimal frequency and timing of grab sample collection, to obtain representative water quality data. Microbial biofilm associated infections are a significant threat to patients with medical devices and are partially responsible for the increased resistance seen in nosocomial infections. The biofilm chemical and physical properties restrict access of chemotherapeutic agents. Therefore, there is a need to enhance the antimicrobial effects of current chemotherapeutic agents. Microalgae live in competitive environments that include film-forming, opportunistic pathogens S. maltophilia and C. albicans. Extracts from 5 diverse species of microalgae were screened for antimicrobial enhancing effects against established biofilms with two different extraction solvents, 3:1 hexanes and isopropyl (HIPA) and 2-methyltetrahydrofuran (2-MTHF).

Microalgae remain a promising, but underdeveloped source of lipids for sustainable biodiesel. Some of the obstacles to cost-effective commercial-scale production have been culture contamination and expensive harvest methods. A chrysophyte isolated from Berkeley Pit Lake and identified as Chromulina freiburgensis, was found to grow rapidly in a pH 2.5 liquid medium and to amass numerous intracellular lipid bodies. This research addresses the scarcity of published knowledge on the topic of chrysophyte species as potential lipid sources for biodiesel. It investigates how growth phase, culture conditions, and harvest timing influence the quantity and composition of lipids produced by this alga. …

Freshwater ecosystems are some of the most biologically diverse environments on Earth. Billions of humans rely on functioning freshwater ecosystems for drinking water and many other services. These ecosystems are increasingly threatened by human impacts including nutrient pollution, invasive species, and climate change. Here I contribute four research chapters that investigate freshwater diversity and ecosystem threats using the Characeae, a family of freshwater green macroalgae, as a study system. Chlorella vulgaris is a unicellular green algae grown throughout the world. Due to its multiple trophic modes as well as its ability to maintain high rates of growth under adverse conditions, it has been of global interest for use in ecological contamination studies, biofuel feedstock optimization, and studies of photosynthetic electron transfer. Using a wide-range of methods for physiological and photosynthetic characterization, the studies within seek to further extend the usefulness of C. vulgaris in a variety of environmentally important studies.
