Urinary tract infections (UTI) are one of the most common bacterial infections, of which, the majority are caused by uropathogenic Escherichia coli (UPEC). Many bacterial pathogens including UPEC synthesize and secrete chemically diverse metabolites called siderophores, which are classically defined by their ability of bind and deliver iron(III), an essential nutrient, to pathogens during infections. UPEC isolates can express multiple siderophore systems, of which, the virulence-associated siderophore yersiniabactin (Ybt) binds to both iron(III) and copper(II) during urinary tract infections. In this thesis we show that Ybt interacts with multiple physiologic transition metals and acts as a metallophore system to deliver …

Copper oxide nanoparticles (CuO NPs) are an up and coming technology increasingly being used in industrial and consumer applications and thus may pose risk to humans and the environment. In the present study, the toxic effects of CuO NPs were studied with two model organisms Saccharomyces cerevisiae and Caenorhabditis elegans. The role of released Cu ions during dissolution of CuO NPs in growth media were studied with freshly suspended, aged NPs, and the released Cu2+ fraction. Exposures to the different Cu treatments showed significant inhibition of S. cerevisiae cellular metabolic activity. Inhibition from the NPs was inversely proportional to size …

It is imperative to not only understand the impact of engineered nanomaterials (ENMs) in edible plants, but also the interactions they have with various additives used in agriculture. In this study, kidney bean plants were grown in potting soil treated with nano copper (nCu), bulk copper (bCu), and copper chloride (CuCl2) at concentrations of 0, 50, and 100 mg/kg for 55 and 90 days. At 15 days of growth, 0, 10, and 100 μm of kinetin (KN) were applied to plants. Plant tissue samples were harvested at 55 days and seeds were reaped at 90 days. The mechanisms behind metal-metal interactions in freshwater environments are currently not well understood. Freshwater environments consist of many different types of metals, from those naturally present such as copper (Cu) and those that originate from anthropogenic sources like silver (Ag). Both Cu and Ag use apical sodium (Na+) channels for uptake into the gills of freshwater fish. In the gills, the mechanisms of Cu2+ and Ag+ toxicity appear to be similar to one another, which is by inhibiting Na+ /potassium (K+)- adenosine triphosphatase (NKA) and carbonic anhydrase (CA). Anthropogenic activities negatively impact fresh water ecosystems through toxic contaminants that are released into the environment. Copper (Cu) is a water contaminant that is fundamentally persistent once introduced into the environment that has the potential for bioaccumulation. Although Cu toxicity has been studied for decades, there is still a continuing problem with new sources and pathways. New approaches are needed to understand distribution and transport of Cu and its potential for complex biological impacts beyond the simple assessment of lethality. Industrially important metals, such as Cu and Ni, sometimes are present at elevated concentrations in lakes, including those in the Sudbury, ON region. Although they are essential metals, their divalent-cation state (Cu2+ and Ni2+) can be toxic at high concentrations in the water. The free-ion toxicity of each of these metals has been studied in isolation, but rarely as a mixture. The economic importance of Cu2+ and Ni2+ makes them essential to study in the context of mixture toxicity.
