Normal aging is associated with a number of changes in behavioral and cellular function, and is often linked to increased susceptibility to cognitive impairment. The hippocampus has been widely implicated in learning and memory, and many forms of learning that are hippocampus-dependent (e.g. trace fear conditioning) are impaired in aged animals. A proposed contributor to aging-related cognitive impairment is aging-related calcium (Ca2+) dysregulation. This dysregulation is thought to result from changes in specific Ca2+-regulatory mechanisms, including abnormal Ca2+ ion channel activity or expression, as well as reduced Ca2+-binding protein (CaBP) expression, which is associated with cognitive and synaptic impairment. An estimated 1.6 million American couples struggle with infertility. Some causes for poor fertility can be clearly defined but in many instances, subfertility is unexplained. Poor oocyte quality is now considered to be a main contributing factor for many causes of infertility. Good oocyte quality is crucial for many processes including embryo development and maintaining pregnancy. There is a possibility that any alterations to the oocyte can have long lasting effects on embryo development and the health of the offspring. AIM: Sarocpenic obesity is a national concern within the United States because this metabolic syndrome is tied with reduced mobility and quality of life. Both obesity and aging are associated with insulin-resistance, chronic low-grade inflammation and muscle weakness. Skeletal muscle regeneration is a process that involves the coordinated effort of myogenic regulatory factors (MRFs), inflammatory signaling, and extracellular matrix (ECM) remodeling for optimal regeneration. It has been demonstrated that obesity and aging have a reduction in muscle regeneration. It has not been examined if sarcopenic obesity will further reduce muscle mass and the regenerative process. Aging is often associated with cognitive decline, including problems with working memory and difficulties forming new memories. These deficits can be directly linked to the hippocampus, an area of the temporal lobe of the brain that is engaged during spatial working memory. Age-related declines may be influenced by changes in important modulatory pathways that impact hippocampal function, including regulation of the metabolite lactate. When astrocytic stores of glycogen are hydrolyzed, lactate is released into the extracellular space where it can be taken up by neurons and used as fuel during moments of activation. Neural mechanisms for grip relaxation are relatively unknown and understudied, as compared to mechanisms for grip initiation. Yet, termination of motor activity is as important as initiation in daily function. This knowledge gap presents incomplete understanding of neural control of hand function and its impairment with aging and neurologic disorders. The purpose of this dissertation was to identify and examine neural mechanisms of grip relaxation in healthy young adults, with aging, and in chronic stroke survivors. Aging of wines and whiskies in oak barrels is a timely and expensive process which could be reduced by acceleration. The purpose of this study is to identify if the use of high intensity ultrasound (HIUS) assisted extraction as an alternative, accelerated aging method could be utilized in the production of an oak extract to be used in wine and whiskies. The current dissertation used a gene x environment (G x E) approach to examine the independent and interactive effects of specific genetic variants and participation in physical and cognitive/social activities (PA and CSA) on cognitive performance in 4,764 participants of the Health and Retirement Study. Using three-wave data, three sets of multi-level growth models were conducted to examine baseline, longitudinal, and interactive effects of genotype (i.e., ApoE, COMT, and BDNF) and CSA/PA on performance across five cognitive measures: immediate, delayed and total word recall, and serial 7s and backwards counting.

Chronic wounds represent an under-acknowledged socioeconomic epidemic, affecting 1.8 million new patients per year and costing the US health care system upwards of $25 billion annually. This substantial cost is rapidly growing due to a disproportionate occurrence in the ever-aging population. Key features associated with age-related impairment of wound healing include limited energy and nutrient exchange, unremitting inflammations, increased reactive oxygen species (ROS), and diminished blood flow. Most chronic wound therapies target specific molecular mechanisms; however, there are often multiple mitigating factors that prevent normal wound closure. B lymphopoiesis declines with age in humans, mice, and rabbits. Impaired B lymphopoiesis correlates with increased fat in the bone marrow (BM), suggesting that adipocytes negatively regulate this process. In fact, adipocyte factors were found to inhibit B cell development in BM cultures.
Our goal was to understand the mechanism by which adipocytes inhibit B cell development. Through culturing mouse BM cells on OP9 stromal cells in the presence of adipocyte-conditioned medium (ACM), we found that adipocytes promote the accumulation of CD11b+Gr1+ myeloid-derived suppressor cells (MDSCs). Aging is a multifactorial process associated with alterations in several physiological functions. It increases susceptibility to disease and ultimately results in mortality. Since the associated diseases of aging are highly prevalent in the geriatric population, targeting the aging process as a whole may provide a better way to delay these age related diseases, in turn delaying aging. BACKGROUND: Alzheimer’s disease (AD) is a neurodegenerative disease with the clinical presentation of memory loss and cognitive impairment. Alzheimer's disease pathology is the accumulation of beta amyloid plaques and neurofibrillary tangles.
METHOD: In this study atherogenic diet was used to induce AD in aged Brown Norway rats. Down syndrome (DS) is a chromosomal condition characterized by accelerated aging that has yet to be directly linked to a DNA repair defect. Reduced PolB and unrepaired damage from oxidative stress observed in DS, point toward defective base excision repair (BER). In this study, we report that low PolB transcript correlates with increased markers of senescence. The gene dosage effect of Trisomy 21 is likely the source for PolB downregulation. We show that the HSA21-localized miR-155 overexpression correlates with a decrease in Creb1 and PolB, thus establishing a putative regulatory pathway. Complaints associated with aging, including cognitive deficits and sleep loss, are highly prevalent and negatively impact quality of life. Further, with increased age, humans are also more likely to experience new-onset psychosocial stressors, such as divorce, loss of a spouse, and social isolation. Stress has detrimental consequences that in many ways parallel the effects of aging on sleep and cognition. The long-standing stress/ glucocorticoid hypotheses of brain aging posit that stress exposure exacerbates aging symptoms, and extensive prior studies have shown that early life stress exposure does worsen phenotypic aging symptoms.
