Despite the chronic debate in the etiology of crohn's disease (cd), a debilitating inflammatory bowel disease (ibd) closely related to ulcerative colitis (uc), an emerging interest in a possible mycobacterial role has been marked. Granuloma and pathologic manifestations in cd resemble aspects found in tuberculosis, leprosy and paratuberculosis. The latter, a chronic enteritis in cattle, goat, sheep and primates, which is similar to human enteritis, also known as cd, is caused by a fastidious, slow growing mycobacterium avium subspecies paratuberculosis (map). Malaria kills at least one million people each year, mostly children - a death every 30 seconds. Almost one half of the world population is at risk from malaria. Antimalarial drugs are the only means for the treatment of about 500 million annual global malaria cases. Because of prevalent drug-resistance it is extremely urgent to identify new drug targets. Many proteins involved in eukaryotic signal transduction and cell cycle progression undergo post-translational lipid modification by a prenyl group. An intensive, replicated monthly sampling of snake communities inhabiting four habitat types was conducted at John F. Kennedy Space Center, Brevard County, Florida from November 2002 through October 2003. Thirteen species (580 individuals plus 74 recaptures) plus one hybrid were captured. The three most commonly captured species, Coluber constrictor, Thamnophis sirtalis, and Thamnophis sauritus, combined made up 85% of the sample. These three species were active during every month of the year, but showed modal activity patterns typical of Temperate Zone snakes. Monthly snake captures were correlated with monthly captures of potential prey species and with mean monthly temperature. Species …

To gain an understanding of the ecology of drift algae in the Indian River Lagoon system along the east coast of central Florida, four questions were addressed: 1) What is the composition and rate of accumulation of drift? 2) How much movement and turnover occurs within drift accumulations? 3) Do growth rates differ for drift versus attached algae? 4) Is there a difference in photosynthetic performance in drift versus attached algal species? The vast majority of valuable agronomic traits are encoded polygenetically. Chloroplast genetic engineering offers an alternate approach to multigene engineering by allowing the insertion of entire pathways in a single transformation event, while being an environmentally friendly approach. Stable integration into the chloroplast genome and transcription of the phaA gene coding for β-ketothiolase was confirmed by Southern and northern blots. Coomassie-stained gel and western blots confirmed hyperexpression of β-ketothiolase in leaves and anthers, with high enzyme activity. The transgenic lines were normal except for the male sterile phenotype, lacking pollen. Scanning electron microscopy revealed a collapsed morphology of the pollen …

Productivity, diversity and survival of estuaries are threatened by explosive coastal population growth and associated recreational activities. One major area of recreational growth has been the number of small pleasure craft motoring in shallow waters at high speeds. On the east coast of Central Florida in the Indian River Lagoon system, intense boating activity occurs year-round and intertidal reefs of the eastern oyster Crassostrea virginica with dead margins (piles of disarticulated shells) on their seaward edges are commonly found adjacent to major boating channels. Epithelial morphogenesis is contingent upon cell shape changes. Cell shape changes are the driving force for the metamorphosis of the adult Drosophila leg from the leg imaginal disc precursor. Genetic analysis has identified several Drosophila genes involved in regulating cell shape changes during leg disc morphogenesis. These include members of the RhoA signaling pathway and the product of the Stubble-stubbloid (Sb-sbd) locus, a transmembrane serine protease. Mutations in the Sb-sbd gene interact genetically with the members of the RhoA signaling pathway, however the nature of the relationship between Sb-sbd serine protease activity and RhoA signaling is not understood. Traumatic brain injury (TBI) induces a state of microglialactivation, which includes upregulation of macrophage functions and release inflammatory mediators such as certain inflammatory cytokines. Current literature suggests that interleukin-1Beta is an important cytokine mediator, which is dramatically increased after brain injury. Previous studies indicate that ATP is released by traumatically injured astrocytes and serves as a cell-to-cell mediator through purinergic receptors after in vitro injury. Land use affects the water quality of lakes. Different land use types yield different effects due to varying amounts and constituents of runoff. In this study, the effects of surrounding land use on the water quality of 50 lakes in Seminole County, Florida was assessed. Using GIS, I placed buffers of 100 and 500 m around each lake. The percentages of land use type were calculated within these buffers for 1990 and 1995. In the Indian River Lagoon, the nudibranch Okenia zoobotryon and its egg masses are found attached to the bryozoan Zoobotryon verticillatum throughout the year. Okenia zoobotryon is thought to live, feed, and reproduce exclusively on Z. verticillatum, which would make this a very specialized predator-prey interaction. The primary goal of my study was to document certain aspects of the ecological and chemical relationships between Z. verticillatum and O. zoobotryon. Mycobacterium avium subspecies paratuberculosis (map) is an intracellular pathogen that is known to parasitize macrophages and monocytes. Map infiltrates gastrointestinal tract host tissue where it is the known etiological agent of johne's disease in ruminants and implicated in the etiology of crohn's disease in humans. Map's ability to survive within macrophages enables it to disseminate throughout the rest of the host, possibly infecting other circulating blood leukocytes. OmiHtrA2 is a highly conserved mammalian serine protease that belongs to the HtrA family of proteins. Omi shares homology with the bacterially expressed heat shock protease HtrA, which functions as a protease at higher temperatures and a chaperone at lower temperatures. Additionally, Omi shares sequence similarity with the mammalian homologs L56/HtrA1 and PRSP/HtrA3. Omi was first isolated as an interacting protein of Mxi2, an alternatively spliced form of the p38 stress-activated kinase, using a modified yeast two-hybrid system. Insight into blood values for free-ranging populations of chelonians is very incomplete. A better understanding of marine turtle ecological physiology is necessary for population health assessments and conservation management plans. Vitamins A and E are fat-soluble compounds required by vertebrates. Vitamin A is required for growth, differentiation and integrity of epithelial tissue, bone remodeling, reproduction and vision. Vitamin E is a constituent of cell membranes where it is an antioxidant and free radical scavenger. Both nutrients are required for reproduction and immune system function.
