Conserving amphibian populations requires knowledge of a species and its habitat relationships. The four-toed salamander (Hemidactylium scutatum) is listed as Special Concern in Maine and 11 additional states and provinces, Threatened in Illinois, and Endangered in Indiana (Appendix A). Little is known of H. scutatum ecology despite the species' extensive range. Infrequent sightings of H. scutatum throughout its range may indicate either low numbers or that the species' behavior make detection difficult. Records for H. scutatum in Maine existed from only 32 sites before my study, and the total number of occurrences of this species in Maine is …

Various materials are used for the treatment of immature teeth that have been subjected to trauma or decay. It is not well established what effect these materials may have on the immature root dentin. A concern would be the resistance to fracture of the remaining root dentin. Chlorine became a major disinfectant for the removal of microbial contaminants in 1914. Current water chlorination procedures yield halogenated disinfection byproducts (DBPs), such as haloalkanes and haloacetic acids (HAAs), due to the reaction of chlorine with naturally occurring organic compounds. Various water utilities have observed decreased HAAs levels in maximum residence time locations (MRTLs), where they were expected to be higher. These MRTLs have low free chlorine residual and high heterotrophic bacteria plate counts. Xanthobacter autotrophicus, GJ-10, is a bacterium that has been shown to contain dehalogenase enzymes and, therefore, can biodegrade HAAs. A number of water-system bacteria were …

The present pilot study is a histological evaluation of mandibular condyles taken from minipigs that underwent either traditional mandibular advancement (bilateral sagittal split osteotomy), or a distraction osteogenesis procedure to advance the mandible. One pig received a bilateral sagittal split osteotomy with an immediate mandibular advancement of 14 mm, while another pig underwent distraction osteogenesis consisting of a gradual advancement of 2 mm per day to advance the mandible the same distance. The control animal received no surgery.

Percent litter, average litter depth, percent bare ground, vegetation density, vegetation canopy, and distance to an ecotone boundary were measured and compared to small mammal capture locations in the Las Vegas Wash. Neotoma lepida appear to be greatly dependent upon the foliage litter of T. ramosissima. Alternatively, ecotone boundaries appear to be the most important factor affecting Dipodomys merriami distributions. Peromyscus eremicus distributions may be predicted based on vegetation density and increased canopy during shorter, winter torpor periods. Chaetodipus penicillatus were consistently associated with dense, seed-bearing vegetation, although C. penicillatus is extremely opportunistic and vegetation density should not limit their …

The connectivity of marine populations and the degree to which they are considered open or closed has important implications for the ecology, management, and resilience of commercially harvested species. Larval exchange is a primary determinant of the level of connectivity between populations, and this thesis examines the intrinsic and extrinsic factors influencing larval transport and the distribution of larvae of the American lobster (Homarus americanus). The potential for larval transport is directly proportional to the planktonic larval duration. Our field data suggest development times in situ were up to three times faster than previous laboratory development times, indicating that potential …

High speed and/or high efficiency separations can be realized using small particles (~ 1 µm) in liquid chromatography (LC). However, due to the large pressure drop caused by small particles, conventional LC pumping systems cannot satisfy the pressure requirements needed to drive the mobile phase through the column. Use of ultrahigh pressure, elevated temperature, or both can overcome these pressure limitations and allow the use of very small particles for high speed and/or high efficiency separations.

Meanwhile, hyperthermia with radiation is important in the treatment of cancer, and it is essential for developers and users of hyperthermia systems to predict, and interpret correctly the biomass thermal and vascular response to heating. Food frequency questionnaire is a cost effective alternative for measuring dietary intake when conducting large scale surveys. We investigated the validity of a 171-item semi-quantitative food frequency questionnaire in assessing food and nutrient intakes and the prescribed intervention measure during a randomized field trial. Eighty-seven free-living adults (48 females and 39 males) aged 30-72 years provided dietary information by self-administering the Walnut Study Dietary Assessment Questionnaire (WSDAQ), which assessed intake over the past six months, and through seven unannounced non-consecutive 24-hour dietary recalls by telephone. The inductively coupled plasma - mass spectrometer (ICP-MS) is the analytical instrument of choice for trace element detection and quantification. Despite the popularity of ICP-MS, significant degradation in sensitivity and precision occurs as the result of matrix and instrument-induced effects. The sources of these effects are not well understood, characterized, or correlated to particular plasma operating condition settings or matrix compositions and involve both neutral and charged species. (a) The Robins reagent [2-acetamido-6-O-(diphenylcarbamoyl)purine] was utilized for glycosylation under Lewis acid conditions. Regioselectivity of glycosylation depends on the glycosyl donor and its 2-O- or 2-N-protecting group. Regioselective N9 glycosylation of 2-acetamido-6-O-(diphenylcarbamoyl)purine with problematic glucosamine has been accomplished by protecting the amino function as a phthalimido group with consequent stabilization of the oxocarbenium cation, and lowering the activation energy by introduction of trichloroacetimidate at the anomeric carbon.

The monitoring of Mojave Desert plant communities during and following disturbance is an important process that could provide invaluable information about disturbance/recovery regimes in similar arid environments across the globe. Fuego is dominated by peatlands; they are important locally and in the context of global climate change, because they store large quantities of organic carbon.

Occupational exposure to asbestos has been associated with increased incidence of pulmonary interstitial fibrosis, mesothelioma of the pleura, and bronchogenic carcinoma. Although the mechanism by which asbestos causes cancer remains unknown, iron associated with asbestos is thought to play a role in the pathogenic effects of fibers.

The West has lost up to 60% of its historic aspen stands over the last century, probably as a result of the successional tendency of aspen to be replaced by coniferous species in the absence of periodic fires. One of several major impacts of this change is the loss of understory forage as conifer canopy cover increases. I measured understory biomass in aspen stands ranging from 0% to 81 % absolute conifer cover in the canopy and found that understory production declines exponentially as conifers replace aspen. An inexpensive detector for proteins is described. The detection technique was based on two-photon excitation intrinsic protein fluorescence using a visible 532 nm diode-pumped nano laser as the excitation source. Proteins that exhibit intrinsic fluorescence must contain at least one tryptophan, tyrosine, or phenylalanine residue in their amino acid sequences. The detector was characterized and was found to have a detection limit of 4 micro-molar for tryptophan, 22 micro-molar for tyrosine and 500 micro-molar for phenylalanine. Capillary electrophoretic (CE) separations were successfully coupled to time-of-flight mass spectrometric (TOFMS) detection for the analysis of three families of biological compounds that act as mediators and/or indicators of disease, namely, catecholamines (dopamine, epinephrine, norepinephrine) and their O-methoxylated metabolites (3-methoxytyramine, norepinephrine, and normetanephrine), indolamines (serotonin, tryptophan, and 5-hydroxytryptophan), and angiotensin peptides. While electrophoretic separation techniques provided high separation efficiency, mass spectrometric detection afforded specificity unsurpassed by other types of detectors.
Both catecholamines and indolamines are present in body fluids at concentrations that make it possible for them to be determined by capillary zone electrophoresis coupled to TOFMS without employing any …

The desert tortoise (Gopherus agassizii) populations have been in decline and have been placed on the endangered species list. No single factor is ultimately responsible for the tortoise decline. It is a combination of events that collectively have a negative impact. I chose to focus my study on only one factor, off-road vehicles. By evaluating the damage done to the terrain by off-road vehicles, a better understanding of their impact on the tortoise populations can be assessed and protective measures taken.

A. californica can be considered a rare endemic species, which is believed to be restricted by unique soil relationships. These relationships make the species vulnerable to anthropogenic habitat disturbance. Although A. californica is listed as critically endangered by the state of Nevada, further research is needed before the species can be listed as federally endangered or threatened. This study used primary observational data and secondary GIS compatible data to characterize A. californica habitat. The purpose of this study was to assess the effects of environmentally relevant concentrations of perchlorate on the development and growth of Rana pipiens. Ammonium perchlorate is a rocket fuel oxidizer that is known to interfere with the function of the thyroid gland and some of the highest United States surface water concentration exists in the Las Vegas Wash, NV. Perchlorate not only blocks the body’s ability to incorporate iodine into T3 and T4 hormones through the sodium iodine symporter system, but also depletes the thyroid glands’ internal stores of iodine. Beta-catenin is an essential cell adhesion and signaling protein, associated with high prolactin levels in rat pituitary tumor cells. It has been shown that phosphorylation affects the location and activity of b-catenin. Glycogen synthase kinase (GSK3-b) is a serine-threonine kinase that phosphorylates b-catenin on N-terminal residues, targeting it for proteasomal degradation. Studies have shown that C-terminal tyrosine phosphorylation decreases the association of b-catenin with cadherin. In 235-1 rat pituitary tumor cells, our lab has shown that the glucocorticoid analog dexamethasone (Dex) decreases the half- life of b-catenin while increasing the activity of GSK3-b. The diseases Dermo and MSX have devastated Chesapeake Bay populations of the eastern oyster, Crassostrea virginica. The protozoan Perkinsus marinus, which causes Dermo, is particularly problematic since it persists over a wide range of salinities and temperatures. An objective of this dissertation was to determine whether specific wild oyster stocks had developed natural resistance to Dermo and if several parameters (survival, growth, condition and energy reserves) were associated with resistance. Striped bass (Morone saxatilis) in Chesapeake Bay, USA, are experiencing an epizootic of mycobacteriosis. This disease, caused by bacteria in the genus Mycobacterium, causes granulomatous lesions of the skin and viscera. Diseased fish are often emaciated, and fish with skin lesions may be significantly disfigured. The overall goal of this work was to examine aspects of the pathobiology of mycobacteria in striped bass via laboratory exposure studies and cellular assays. Sequence similarity searching is commonly used to help clarify the biochemical and physiological features of newly discovered genes or proteins. An efficient similarity search relies on the choice of tools and their associated subprograms and numerous parameter settings. To assist researchers in selecting optimal programs and parameter settings for efficient sequence similarity searches, the web-based expert system, Smart Sequence Similarity Search (S4) was developed.
