Development And Characterization Of Monoclonal Antibodies Directed Against Tanapox Virus, Michael Franz
Development And Characterization Of Monoclonal Antibodies Directed Against Tanapox Virus, Michael Franz
Masters Theses
The initial symptoms of a Tanapoxvirus (TPV) infection are indistinguishable from smallpox, which is caused by variola virus (VV). In the post-9/11 area, reagents that can quickly differentiate a TPV infection from smallpox are very desirable, but currently unavailable. Monoclonal antibodies (mAbs) directed specifically against TPV can be used for this purpose. Six mAbs were developed and characterized that specifically bind to TPV infected owl monkey kidney (OMK) cells, but not uninfected cells. The apparent molecular mass of the TPV protein(s) recognized by 5 mAbs were calculated using the Western blot technique. One mAb demonstrated no binding using the Western …

Historical Biogeography Of Lowland Species Of Toads (Bufo) Across The Trans-Mexican Neovolcanic Belt And The Isthmus Of Tehuantepec, Benson H. Morrill
Undergraduate Honors Capstone Projects
The eastern extension of the massive Trans-Mexican Neovolcanic Belt 1 (TMNB) contacts the Atlantic Coast in central Veracruz, Mexico. Although it is not a massive structure at this eastern terminus, the TMNB has nonetheless effected vicariance and subsequent speciation in several species groups, including bufonid toads and freshwater fishes. In this study, we examined mtDNA sequences (cyt b, 16S) from populations of two different species groups of toads (B. marinus, and the B. nebulifer-B. valliceps species pair) from a lowland transect across the eastern end of the TMNB. We also included samples from outside this region (e.g., Costa …

Distribution Of Chemistry And Sexual Fecundity In The Lichenized-Fungi, Xanthoparmelia Cumberlandia And Xanthoparmelia Coloradoensis On Boulder Mountain, Aquarius Plateau, Ut, Heather Bird Jackson
Theses and Dissertations
Three aspects of Xanthoparmelia cumberlandia and Xanthoparmelia coloradoënsis populations found at two elevations are explored: clustering of secondary chemicals and the resulting implications for taxonomic distinctions, the usefulness of thallus size as an indirect measure of sexual fecundity, and the frequency of sexual reproduction.
First, we use clustering of 46 chemicals produced by X. cumberlandia and X. coloradoënsis to evaluate the adequacy of the current taxonomic distinction between them. Using principal components analysis and UPGMA, we find that the currently recognized species boundaries indicated by the presence of stictic acid in X. cumberlandia and salazinic acid in X. coloradoënsis are …

Density Effects On Growth, Survival And Diet Of June Sucker (Chasmistes Liorus): A Component Allee Effect In An Endangered Species., David Barrett Gonzalez
Theses and Dissertations
Density-dependence is considered one of the most important regulators of population growth, and it has been documented across a wide variety of species. Typically, population growth rate and components thereof decline with increasing density (i.e., negative density-dependence); however, in species that exhibit high population densities and social behavior, positive density-dependence (i.e., Allee effect) may occur at low density. June sucker, a federally endangered lake sucker endemic to Utah Lake, Utah, USA, occurred historically at high density, and it exhibits coordinated feeding behavior. These characteristics indicate a potential for the existence of an Allee effect at current low population densities. To …
Evolution And Phylogeny Of Basal Winged Insects With Emphasis On Mayflies (Ephemeroptera), Thomas H. Ogden
Evolution And Phylogeny Of Basal Winged Insects With Emphasis On Mayflies (Ephemeroptera), Thomas H. Ogden
Theses and Dissertations
Ephemeroptera (mayflies) is a monophyletic group of semi-aquatic pterygote insects, comprising 3083 species, 376 genera, and 37 described families and are present on all continents, excluding Antarctica, being associated with freshwater and brackish water habitats. The order is unique among pterygote insects in possessing functional wings at the penultimate molt (subimago stage), prior to the full development of genitalia; in all other insects the presence of functional wings occurs only after the final molt. The purpose of this dissertation is to use molecular and morphological data, in order to investigate the position of the order Ephemeroptera among other insect orders, …
Multiple Kh Domains Of Poly C Binding Protein Are Required For Its Trans-Activation On The Proximal Promoter Of The Mu-Opioid Receptor Gene, Adnan K. Malik
Multiple Kh Domains Of Poly C Binding Protein Are Required For Its Trans-Activation On The Proximal Promoter Of The Mu-Opioid Receptor Gene, Adnan K. Malik
Seton Hall University Dissertations and Theses (ETDs)
The mu-opioid receptor (MOR) has been shown to mediate morphine induced analgesia, addiction, and tolerance. The expression of the major product of the MOR (MOR-1) gene has been found to be driven by two promoters, a distal promoter and a proximal promoter. The proximal promoter region has been shown in previous studies to be the major promoter for the regulation of MOR gene transcription in the adult mouse brain and embryonic development. Recent studies have shown that single-stranded DNA binding proteins (such as Poly C Binding Protein) are involved in transcriptional regulation of mouse MOR gene. Poly C binding protein …

Characterization Of A Catechol-Type Siderophore And The Detection Of A Possible Outer Membrane Receptor Protein From Rhizobium Leguminosarum Strain Iari 312., Brianne Lee Clark
Electronic Theses and Dissertations
Many gram-negative bacteria produce and secrete siderophores under iron-deficient conditions. Siderophores are low molecular weight compounds (600-1500 Daltons), which chelate ferric iron with an extremely high affinity, and the complex is actively transported across the outer and inner membranes of gram-negative bacteria. There are two main classes of siderophores: catechol and hydroxamate. Catechol-type siderophores chelate ferric iron via hydroxyl groups, and hydroxamate-type siderophores chelate ferric iron via a carbonyl group with an adjacent nitrogen. Rhizobia fix atmospheric nitrogen symbiotically in leguminous plants using the iron-containing enzyme nitrogenase. To satisfy their iron requirements, many rhizobia are known to produce siderophores. Rhizobium …
Mechanism Of Chromium Reduction Of Benzo[A]Pyrene Diolepoxide Mutagenesis In Human Fibroblasts, Cara Suzanne Kriss
Mechanism Of Chromium Reduction Of Benzo[A]Pyrene Diolepoxide Mutagenesis In Human Fibroblasts, Cara Suzanne Kriss
Masters Theses
Environmental agents can cause an increase in cellular stress and damage DNA which can lead to diseases such as cancer. Chromium produces cellular stress by increasing the reactive oxygen species through a Fenton-like reaction, where chromium reacts with hydrogen peroxide to form hydroxyl radicals. Benzo[ a ]pyrene is an environmental carcinogen that is metabolized by cytochrome p450 enzymes into benzo[a]pyrene diolepoxide (BPDE). BPDE is highly reactive with DNA producing large DNA-adducts primarily on guanine nucleotides. The primary intracellular pathway by which mammalian cells remove damaged DNA caused by bulky mutagens is nucleotide excision repair. We have found that, in BPDE …

Studies On Mycobacterium Ulcerans, Terri Alford
Masters Theses
Efforts to create a mycolactone-negative mutant of Mycobacterium ulcerans via transposon mutagenesis were undertaken to elucidate the genes involved in the production of this toxin. Though only five PRC-confirmed insertion mutants were produced, one of these, 1615:tnp6w, was confirmed to be deficient in mycolactone production via thin-layer chromatography (TLC) of acetone soluble lipids (ASLs), cytopathicity assay of culture filtrate on L929 fibroblasts, and mass spectroscopy of ASLs. Using nested-PRC, it was determined that the transposon insertion of 1615:tnp6w had occurred in a region of DNA containing a gene 97% identical to the M. marinum ferric uptake regulator homolog (FurA) gene …
Prevalence Of Teleological And Lamarckian Misconceptions Of Natural Selection Among College Students, Bryce T. Battisti
Prevalence Of Teleological And Lamarckian Misconceptions Of Natural Selection Among College Students, Bryce T. Battisti
Theses and Dissertations
Science teachers and researchers note that students must correctly understand the role of natural selection in evolution to make sense of biology. The level of understanding of natural selection can be assessed using the Conceptual Inventory of Natural Selection (CINS) which is a 20-item multiple-choice test that incorporates student misconceptions as distractors. In the present study, Item Response Theory (IRT) was used to analyze the occurance of misconceptions on the CINS among 1192 students in an introductory non-majors biology course. The four most difficult topics in the CINS are: (a) how change occurs in a population; (b) origin of variation; …

Development Of Web Tools For Nlx Simulation Software, Daniel Chai Siriphongs
Development Of Web Tools For Nlx Simulation Software, Daniel Chai Siriphongs
Loma Linda University Electronic Theses, Dissertations & Projects
Valuable mathematical equations have developed within biology and other sciences (physics, chemistry, etc.) that model various processes, but the complexity of the equations are not easily understood by students, scientists, and those with limited knowledge of the subject area. Simulation software used to visually explain a model is normally proprietary to the specific model equations and cannot be easily adapted to new models by non-programmers. Simulation software needs to be modular and based upon web technologies so that the software can be run on multiple platforms. The NLX simulation software is a group of Java-based objects that can be combined …

Non-Parametric Algorithms For Evaluating Gene Expression In Cancer Using Dna Microarray Technology, Virginie Aris
Dissertations
Microarray technology has transformed the field of cancer biology by enabling the simultaneous evaluation of tens of thousands mRNA expression levels in a single experiment. This technology has been applied to medical science in order to find gene expression markers that cluster diseased and normal tissues, genes affected by treatments, and gene network interactions. All methods of microarray data analysis can be summarized as a study of differential gene expression. This study addresses three questions, 1) the roles of selectively expressed genes for the classification of cancer, 2) issues of accounting for both experimental and biological noise, and 3) issues …

Does Seed Size Affect The Rate Of Germination And Early Seedling Growth In Hairy Vetch?, Joy Ekpo
Does Seed Size Affect The Rate Of Germination And Early Seedling Growth In Hairy Vetch?, Joy Ekpo
McCabe Thesis Collection
Size is an important characteristic of seed quality because larger seeds with larger volume contain more resources and are likely to exhibit greater vigor than smaller seeds (Ellis, 1992). In a range of plant species, seed size has been shown to affect germination rate, emergence rate, success of establishment, and growth (Black 1957; Bentley et al. 1980; Sanderson et al. 2002). Hairy vetch is an annual legume that is used for improvement of soil for pasture and to provide ground cover protecting against erosion (Duke 1981; Undersander 2002; Sattell et al 1988). Problems facing the establishment of hairy vetch are …
Genetic Analysis Of The Putative Streptolysin O Regulator From Streptococcus Pyogenes, Robert Tyrone Billingslea
Genetic Analysis Of The Putative Streptolysin O Regulator From Streptococcus Pyogenes, Robert Tyrone Billingslea
McCabe Thesis Collection
Group A streptococci (Streptococcus pyogenes) is one of the most frequent pathogens of humans. It is estimated that between 5-15% of normal individuals harbor the bacterium, usually in the upper respiratory tract, without signs of disease. S. pyogenes can initiate an active infection when defenses are compromised or when the organism is able to penetrate the host's defenses. When the bacteria invade vulnerable tissues, a variety of suppurative infections can occur. Acute S. pyogenes infection may be present as pharyngitis ("strep throat"), scarlet fever (rash), impetigo (infection of the superficial layers of the skin) or cellulitis (infection of the deep …
Moisture Potential Affects Rate Of Leaf Appearance In Cool Season Grasses, Nina Cherone Terrell
Moisture Potential Affects Rate Of Leaf Appearance In Cool Season Grasses, Nina Cherone Terrell
McCabe Thesis Collection
The rate of leaf appearance and onset of growth in cool season grasses may be influenced by soil moisture potential and exposure to below freezing temperatures. Levels of moisture potential and days exposed to freezing temperatures, which will restrict leaf appearance, are not well defined. The first part of this study was to determine the effects of different moisture potentials on the leaf appearance rates in two cool season grasses. Italian ryegrass (IRG) and tall wheat grass (TWG) were grown hydroponically at 4 moisture potentials in seed pouches in an incubator under a photoperiod of 13 hours light and 11 …
Natural History, Demography, And Home Range Characteristics Of A Southern California Population Of Phrynosoma Mcallii Inhabiting Atypical Habitat, Kirk Setser
All Graduate Theses and Dissertations, Spring 1920 to Summer 2023
Phrynosoma mcallii inhabiting a topographically complex, sparsely vegetated, "mudhill" terrain site in southern California in 1999 and 2000 had low mortality rates, and small clutch sizes compared to previous data from this species. Females likely produced a single clutch of eggs in each year. Although this population occurs near a heavily used OHV trail, I did not detect any mortality due to OHV use.
Lizards did not center home ranges on obvious habitat features. Lizards maintained smaller home ranges and exhibited greater range fidelity than did lizards at other sites in other years. Phrynosoma mcallii share many area-use characters with …

Alkaloids And Old Lace: Pollen Toxins Exclude Generalist Pollinators From Death Camas, Melissa Weber
Alkaloids And Old Lace: Pollen Toxins Exclude Generalist Pollinators From Death Camas, Melissa Weber
Undergraduate Honors Capstone Projects
Many plants produce toxins to which specialist herbivores - typically insects - have evolved counter-adaptations, sometimes resulting in a co-evolutionary arms race. Although many non-social bee species are likewise taxonomic host specialists, the pollination guilds at their floral hosts frequently include diverse floral generalists as well, even on plants that are otherwise chemically defended. In this study, we show that pollen and nectar of foothills death camas (Toxicoscordion [=Zigadenus] paniculatum) contains zygacine, the alkaloid responsible for this plant's notorious mammalian toxicity. Many adults and larvae of the generalist solitary bee, Osmia lignaria (Megachilidae), were paralyzed and …
Host Shifts In Phytophagous Insects: A Mathematical Modeling Perspective, Meenakshi Venkataraman
Host Shifts In Phytophagous Insects: A Mathematical Modeling Perspective, Meenakshi Venkataraman
All Graduate Theses and Dissertations, Spring 1920 to Summer 2023
I explored changes in predator-prey system stability with the addition of spatial heterogeneity in prey density dependent mortality. The results indicated that this addition led to greater stability for all prey spatial distributions if prey was aggregated or loosely aggregated. These results indicated prey spatial distribution was critical to population stability. I next explored whether predator emigration under the above conditions would lead to a host shift. I used Chesson and Murdoch's templates of host density independent (HDI) aggregation and host density dependent (HDD) aggregation in predators as the basis for my metapopulation models. I varied prey density dependent mortality …
Phylogeny Of The Polyneopterous Insects With Emphasis On Plecoptera: Molecular And Morpological Evidence, Matthew Dana Terry
Phylogeny Of The Polyneopterous Insects With Emphasis On Plecoptera: Molecular And Morpological Evidence, Matthew Dana Terry
Theses and Dissertations
Polyneoptera is an assemblage of eleven insect orders comprising the "orthopteroid" insects. It includes familiar insects such as grasshoppers, roaches, termites, earwigs and preying mantises; as well as the more obscure web-spinners, angel insects and ice-crawlers. We present a phylogenetic analysis of the polyneopteran orders based on 18S rDNA, 28S rDNA, Histone 3, and a coded morphology matrix for an extensive sampling of taxa. We investigate the use of congruence between separate datasets as an a priori measure of alignment quality. Our results support the paraphyly of Polyneoptera, the monophyly of Dictyoptera, sister taxon relationships between Embiidina + Phasmatodea and …
Phylogenetic Systematics And Evolution Within The Family Scincidae, Alison Swindle Whiting
Phylogenetic Systematics And Evolution Within The Family Scincidae, Alison Swindle Whiting
Theses and Dissertations
Scincidae (skinks) comprise one of the largest families of lizards, and the more than 1300 species show great variation in body size and form, and are found worldwide in a diversity of habitats. The group presents many interesting questions ranging from the colonization of oceanic islands, to the evolution of limb loss, yet most of these and other questions remain understudied. The purpose of this dissertation is to use multiple mitochonidrial and nuclear DNA markers in connection with current cladistic methods to address evolutionary questions at many levels within Scincidae. In chapter one, a molecular phylogenetic study, based on six …

The pond is approximately 36 m long and 38 m wide and is located on a south-facing hillside at 202 m in elevation. A drift fence composed of landscaping cloth was constructed to completely encircle the site. At every 5.15 m, funnel traps were positioned on both sides of the fence. Traps were checked daily …
The Status Of Leptodactylus Validus As A Single Taxon And Its Relationship To Letptodactylus Pallidirostris, Kenneth Christopher Yanek
The Status Of Leptodactylus Validus As A Single Taxon And Its Relationship To Letptodactylus Pallidirostris, Kenneth Christopher Yanek
Master's Theses
Leptodactylus validus exhibits an unusual distribution inhabiting Trinidad, Tobago, and the Lesser Antilles, but not the mainland of South America. This distribution is inconsistent with other distribution patterns observed for these islands. Although slight variation in adult morphology has been observed among the island populations, call data suggest the presence of a single species. Calls of Leptodactylus pallidirostris from Venezuela suggested that this taxon might be conspecific with L. validus. Herein sequence data from the 12S and 16S mt rRNA genes were collected and analyzed in order to address: 1.) whether more than one species is represented within L. validus, …

Habitat Use And Seasonal Activity Of Selected Snakes On John F. Kenned, Karen Dyer
Electronic Theses and Dissertations
An intensive, replicated monthly sampling of snake communities inhabiting four habitat types was conducted at John F. Kennedy Space Center, Brevard County, Florida from November 2002 through October 2003. Thirteen species (580 individuals plus 74 recaptures) plus one hybrid were captured. The three most commonly captured species, Coluber constrictor, Thamnophis sirtalis, and Thamnophis sauritus, combined made up 85% of the sample. These three species were active during every month of the year, but showed modal activity patterns typical of Temperate Zone snakes. Monthly snake captures were correlated with monthly captures of potential prey species and with mean monthly temperature. Species …
