The initial symptoms of a Tanapoxvirus (TPV) infection are indistinguishable from smallpox, which is caused by variola virus (VV). In the post-9/11 area, reagents that can quickly differentiate a TPV infection from smallpox are very desirable, but currently unavailable. Monoclonal antibodies (mAbs) directed specifically against TPV can be used for this purpose. Six mAbs were developed and characterized that specifically bind to TPV infected owl monkey kidney (OMK) cells, but not uninfected cells. The apparent molecular mass of the TPV protein(s) recognized by 5 mAbs were calculated using the Western blot technique. One mAb demonstrated no binding using the Western …

The eastern extension of the massive Trans-Mexican Neovolcanic Belt 1 (TMNB) contacts the Atlantic Coast in central Veracruz, Mexico. Although it is not a massive structure at this eastern terminus, the TMNB has nonetheless effected vicariance and subsequent speciation in several species groups, including bufonid toads and freshwater fishes. In this study, we examined mtDNA sequences (cyt b, 16S) from populations of two different species groups of toads (B. marinus, and the B. nebulifer-B. valliceps species pair) from a lowland transect across the eastern end of the TMNB. Three aspects of Xanthoparmelia cumberlandia and Xanthoparmelia coloradoënsis populations found at two elevations are explored: clustering of secondary chemicals and the resulting implications for taxonomic distinctions, the usefulness of thallus size as an indirect measure of sexual fecundity, and the frequency of sexual reproduction.

First, we use clustering of 46 chemicals produced by X. cumberlandia and X. coloradoënsis to evaluate the adequacy of the current taxonomic distinction between them. Using principal components analysis and UPGMA, we find that the currently recognized species boundaries indicated by the presence of stictic acid in X. cumberlandia and salazinic acid in X. coloradoënsis are …

Density-dependence is considered one of the most important regulators of population growth, and it has been documented across a wide variety of species. Typically, population growth rate and components thereof decline with increasing density (i.e., negative density-dependence); however, in species that exhibit high population densities and social behavior, positive density-dependence (i.e., Allee effect) may occur at low density. June sucker, a federally endangered lake sucker endemic to Utah Lake, Utah, USA, occurred historically at high density, and it exhibits coordinated feeding behavior. Ephemeroptera (mayflies) is a monophyletic group of semi-aquatic pterygote insects, comprising 3083 species, 376 genera, and 37 described families and are present on all continents, excluding Antarctica, being associated with freshwater and brackish water habitats. The order is unique among pterygote insects in possessing functional wings at the penultimate molt (subimago stage), prior to the full development of genitalia; in all other insects the presence of functional wings occurs only after the final molt. The mu-opioid receptor (MOR) has been shown to mediate morphine induced analgesia, addiction, and tolerance. The expression of the major product of the MOR (MOR-1) gene has been found to be driven by two promoters, a distal promoter and a proximal promoter. The proximal promoter region has been shown in previous studies to be the major promoter for the regulation of MOR gene transcription in the adult mouse brain and embryonic development. Many gram-negative bacteria produce and secrete siderophores under iron-deficient conditions. Siderophores are low molecular weight compounds (600-1500 Daltons), which chelate ferric iron with an extremely high affinity, and the complex is actively transported across the outer and inner membranes of gram-negative bacteria. There are two main classes of siderophores: catechol and hydroxamate. Catechol-type siderophores chelate ferric iron via hydroxyl groups, and hydroxamate-type siderophores chelate ferric iron via a carbonyl group with an adjacent nitrogen. Rhizobia fix atmospheric nitrogen symbiotically in leguminous plants using the iron-containing enzyme nitrogenase. To satisfy their iron requirements, many rhizobia are known to produce siderophores. Environmental agents can cause an increase in cellular stress and damage DNA which can lead to diseases such as cancer. Chromium produces cellular stress by increasing the reactive oxygen species through a Fenton-like reaction, where chromium reacts with hydrogen peroxide to form hydroxyl radicals. Benzo[ a ]pyrene is an environmental carcinogen that is metabolized by cytochrome p450 enzymes into benzo[a]pyrene diolepoxide (BPDE). BPDE is highly reactive with DNA producing large DNA-adducts primarily on guanine nucleotides. The primary intracellular pathway by which mammalian cells remove damaged DNA caused by bulky mutagens is nucleotide excision repair. We have found that, in BPDE …

Efforts to create a mycolactone-negative mutant of Mycobacterium ulcerans via transposon mutagenesis were undertaken to elucidate the genes involved in the production of this toxin. Though only five PRC-confirmed insertion mutants were produced, one of these, 1615:tnp6w, was confirmed to be deficient in mycolactone production via thin-layer chromatography (TLC) of acetone soluble lipids (ASLs), cytopathicity assay of culture filtrate on L929 fibroblasts, and mass spectroscopy of ASLs. Using nested-PRC, it was determined that the transposon insertion of 1615:tnp6w had occurred in a region of DNA containing a gene 97% identical to the M. marinum ferric uptake regulator homolog (FurA) gene …

Science teachers and researchers note that students must correctly understand the role of natural selection in evolution to make sense of biology. The level of understanding of natural selection can be assessed using the Conceptual Inventory of Natural Selection (CINS) which is a 20-item multiple-choice test that incorporates student misconceptions as distractors. In the present study, Item Response Theory (IRT) was used to analyze the occurance of misconceptions on the CINS among 1192 students in an introductory non-majors biology course. The chief purpose of this study is to provide information on the habitat and breeding information of Magnolia Warblers on Unaka Mountain. Magnolia Warblers breed in Canada and the Northeastern United States. There are no current breeding records for this species in Tennessee.

For the habitat analysis, trees were identified on individual subplots. Shrubs were analyzed using the point-intercept method. Most of the major tree and shrub species were different than those found in Maine, which is a major breeding region for this species. The general goals of this study were to evaluate transplant methods for Lake Pontchartrain submersed aquatic vegetation (SAV) restoration, and to compare survival and expansion of transplanted SAV among four sites. Two experiments were conducted from summer 2000 through spring 2001 at two north shore and two south shore sites in Lake Pontchartrain. Experiment 1 involved testing the effects of transplanting with matting, staples, exclosures, and interspecific interaction on survival and expansion of Ruppia maritima and Vallisneria americana transplants in Lake Pontchartrain. Size is an important characteristic of seed quality because larger seeds with larger volume contain more resources and are likely to exhibit greater vigor than smaller seeds (Ellis, 1992). It is estimated that between 5-15% of normal individuals harbor the bacterium, usually in the upper respiratory tract, without signs of disease. S. pyogenes can initiate an active infection when defenses are compromised or when the organism is able to penetrate the host's defenses. The rate of leaf appearance and onset of growth in cool season grasses may be influenced by soil moisture potential and exposure to below freezing temperatures. Levels of moisture potential and days exposed to freezing temperatures, which will restrict leaf appearance, are not well defined. The first part of this study was to determine the effects of different moisture potentials on the leaf appearance rates in two cool season grasses. Phrynosoma mcallii inhabiting a topographically complex, sparsely vegetated, "mudhill" terrain site in southern California in 1999 and 2000 had low mortality rates, and small clutch sizes compared to previous data from this species. Females likely produced a single clutch of eggs in each year. Although this population occurs near a heavily used OHV trail, I did not detect any mortality due to OHV use.

Lizards did not center home ranges on obvious habitat features. Lizards maintained smaller home ranges and exhibited greater range fidelity than did lizards at other sites in other years. Phrynosoma mcallii share many area-use characters with …

In the Southern Appalachians, there are growing concerns about Spotted Salamanders (Ambystoma maculatum) due to habitat destruction and the uncertain future of their forested riparian habitat. In this study, I establish baseline data of a population of Spotted Salamanders so future monitoring of this population can be performed. The study site is located in northeastern Tennessee, just east of the city of Bristol. Many plants produce toxins to which specialist herbivores - typically insects - have evolved counter-adaptations, sometimes resulting in a co-evolutionary arms race. Although many non-social bee species are likewise taxonomic host specialists, the pollination guilds at their floral hosts frequently include diverse floral generalists as well, even on plants that are otherwise chemically defended. In this study, we show that pollen and nectar of foothills death camas (Toxicoscordion [=Zigadenus] paniculatum) contains zygacine, the alkaloid responsible for this plant's notorious mammalian toxicity. I explored changes in predator-prey system stability with the addition of spatial heterogeneity in prey density dependent mortality. The results indicated that this addition led to greater stability for all prey spatial distributions if prey was aggregated or loosely aggregated. These results indicated prey spatial distribution was critical to population stability. I next explored whether predator emigration under the above conditions would lead to a host shift. I used Chesson and Murdoch's templates of host density independent (HDI) aggregation and host density dependent (HDD) aggregation in predators as the basis for my metapopulation models. Polyneoptera is an assemblage of eleven insect orders comprising the "orthopteroid" insects. It includes familiar insects such as grasshoppers, roaches, termites, earwigs and preying mantises; as well as the more obscure web-spinners, angel insects and ice-crawlers. We present a phylogenetic analysis of the polyneopteran orders based on 18S rDNA, 28S rDNA, Histone 3, and a coded morphology matrix for an extensive sampling of taxa. We investigate the use of congruence between separate datasets as an a priori measure of alignment quality. A farm pond approximately 40 years old located in Wayne County, West Virginia was studied to determine the ingress and egress of amphibians and reptiles. In addition to the pond, 2 small marshes were also examined. The study extended from February 2003 to November 2003. The pond is approximately 36 m long and 38 m wide and is located on a south-facing hillside at 202 m in elevation. Leptodactylus validus exhibits an unusual distribution inhabiting Trinidad, Tobago, and the Lesser Antilles, but not the mainland of South America. This distribution is inconsistent with other distribution patterns observed for these islands. Although slight variation in adult morphology has been observed among the island populations, call data suggest the presence of a single species. Calls of Leptodactylus pallidirostris from Venezuela suggested that this taxon might be conspecific with L. validus. An intensive, replicated monthly sampling of snake communities inhabiting four habitat types was conducted at John F. Kennedy Space Center, Brevard County, Florida from November 2002 through October 2003. Thirteen species (580 individuals plus 74 recaptures) plus one hybrid were captured. The three most commonly captured species, Coluber constrictor, Thamnophis sirtalis, and Thamnophis sauritus, combined made up 85% of the sample. These three species were active during every month of the year, but showed modal activity patterns typical of Temperate Zone snakes. Monthly snake captures were correlated with monthly captures of potential prey species and with mean monthly temperature. Species …
