Functional Roles Of Nova In Rna Metabolism, Giovanni Stefani

Nova-1 is a neuron-specific RNA binding protein, distributed both in the nucleus and the cytoplasm of several populations of neurons. The work presented here confirms that Nova-1 binds a sequence of RNA found in an intron of the glycine receptor a2 subunit pre-mRNA. Evidence that Nova-1 acts as a regulator of alternative splicing in transfected cell lines is presented. Furthermore, a direct role of Nova-1 on splicing is demonstrated by establishing an in vitro assay for Nova-l's regulatory role in splicing. Multi-drug resistance or MDR is a major impediment to the successful administration of chemotherapy. Broadly defined, the term MDR applies to any mechanism the cell uses to counter the effects of chemotherapeutic agents, protecting the cell at once against the toxicity of many, structurally dissimilar compounds. There are several routes to MDR for a cell, and they include everything from decreasing intracellular drug concentrations to increasing rates drug metabolism. The study presented here focuses on the role of drug transporters in conferring drug resistance. A measurement of the effective diffractive structure function Ffi of the antiproton obtained from a study of single diffractive dijet events produced in association with a leading antiproton in pp collisions at the center-of-mass energy y/s = 1800 and 630 GeV is presented. Inclusive samples of single diffractive events were collected during the Tevatron collider run of 1995—1996 using the Collider Detector at Fermilab (CDF) by triggering on a leading antiproton detected in a forward Roman Pot spectrometer. From these samples, single diffractive dijet subsamples were selected by requiring two or more jets with transverse energy ET > 7 GeV in …

Incorporation of chemical probes into proteins is a powerful way to elucidate biological processes and to engineer novel function. This thesis describes the ligation of synthetic molecules to target proteins in an intracellular environment, and the use of semi-synthetic proteins for whole animal studies. In the first approach a cellular protein is genetically tagged with one half of a split-intein. The complementary half is linked in vitro to the synthetic probe and this fusion is delivered into cells using a transduction peptide. Estrogen fulfills a variety of physiological roles through its nuclear receptors, estrogen receptors (ERs). The action of ERs is modulated and mediated by diverse interacting cofactors. The initial aim of this thesis work was to identify ligand-, subtype- and/or cell typespecific ER-interacting proteins. I identified a number of nuclear extract-derived proteins that interact with immobilized ER ligand binding domains (LBDs) in a 17(3-estradiol (E2)-dependent manner. The most prominent of these are components of the thyroid hormone receptor associated protein (TRAP)/Mediator coactivator complex, which interacts with ERa and ER(3 both in unfractionated nuclear extracts and in purified form equally efficiently. Potassium ion channels are ubiquitously expressed from bacteria to mammals where they are involved in various processes ranging from the regulation of osmotic pressure in a single cell to the electrical response in muscle fibers to the generation of action potentials in neurons. The B K channel family (BK for Big K+ conductance) is an interesting subfamily of K+ channels responsive to both membrane voltage and intracellular calcium ion. Identification And Characterization Of The Dna Damage Response Function Of Human Rif1 And A Search For Human Rap1 Interacting Factors, Joshua Silverman

Rifl, the ortholog of the yeast Rapl interacting factor 1, a telomere length regulator was identified. Immunofluorescence did not reveal an association of human Rifl with telomeres. Preliminary co-immunoprecipitation with known telomeric proteins and chromatin immunoprecipitation failed to show an interaction of human Rifl with telomeric protein complexes or with telomeric DNA. Unexpectedly, Rifl responded to ionizing radiation (IR), UV light, and clastogens, forming foci that co-localized with other DNA damage response factors such as 53BP1, ATM, BRCA1, Chkl, Nbsl, and Rad 17. Furthermore, Rifl localized to uncapped telomeres, as do other DNA damage response factors. Among DNA damage response …
