The chemical synthesis of highly specialized peptides that might possess catalytic action provides one of the approaches to an understanding of the mechanism of enzyme action. The experiments reported in this thesis involve the chemical synthesis of model compounds designed to explore how trypsin and chymotrypsin may function. Since several lines of evidence indicate a catalytic role for histidine residues in these enzymes, it was proposed to prepare imidazole derivatives with carefully designed side chains which might attract and orient similarly shaped substrates. Central to the modern theory of the electrical properties of cells is the permeability of the plasma membrane to the ions passing across this structure; it has been our purpose to explore, both theoretically and experimentally, the physical implications of this point-of-view. The cornerstone of our analysis has been the fundamental flux equations of Nernst and Planck, which describe the movement of ions in solution subject to gradients of electrochemical potential. Genetic Recombination In Pneumococcal Transformation: The Role Of Integrity And Heterogeneity Of Deoxyribonucleate Molecules, As Reflected In Kinetic Analysis, Joan Louise Kent

Pneumococcal transformation has been used to study intramolecular genetic recombination. A kinetic technique is developed whereby the cumulative number of multiply-marked transformants is measured as a function of the duration of exposure to a multiply-marked DNA. Transformants accumulate with time at a rate characteristic of the particular markers and the number of DNA particles involved, thus singly-marked transformants accumulate linearly with time of exposure to DNA, as do some multiply-marked transformants. Other unlinked pairs of markers accumulate exponentially with time, by a second-order process, at a rate very close to that expected from random interaction of one cell with two …

Reoviruses occur widely in the respiratory and enteric tracts of man and animals, but their relation to disease is not clear. The present study concerns the physical-chemical characteristics and interaction with cells of reovirus type 3. A main objective of the study is to relate some of the distinguishing biological characteristics of the virus to the structure of its RNA.

The growth of the RNA-containing bacteriophage f2 has been studied. DNA synthesis appears normal after phage infection. It is possible to dissociate the synthesis of viral RNA from the synthesis of DNA using thymineless bacteria with fluorinated pyrimidines and Mitomycin C as inhibitors of DNA synthesis. It is concluded that the growth of f2 is independent of DNA synthesis.

The initial observation that delayed-type hypersensitivity could be transferred from an allergic to a normal animal by means of living white cells was made at The Rockefeller Institute many years ago. Since that time it has been recognized that cellular transfer appears to be a unifying principle underlying both contact dermatitis and tuberculin hypersensitivity. Later studies in man have suggested that the phenomenon can be reproduced with use cell-free extracts. Similar attempts have been made with extracts of cells from sensitized guinea pigs but, there exist only three reports in the literature which claim that transfer of contact or tuberculin …
