A Non-Circadian Role For Clock-Genes In Sleep Homeostasis: A Strain Comparison, Paul Franken, Ryan Thomason, H. Craig Heller, Bruce F. O'Hara

Biology Faculty Publications

BACKGROUND: We have previously reported that the expression of circadian clock-genes increases in the cerebral cortex after sleep deprivation (SD) and that the sleep rebound following SD is attenuated in mice deficient for one or more clock-genes. We hypothesized that besides generating circadian rhythms, clock-genes also play a role in the homeostatic regulation of sleep. Here we follow the time course of the forebrain changes in the expression of the clock-genes period (per)-1, per2, and of the clock-controlled gene albumin D-binding protein (dbp) during a 6 h SD and subsequent recovery sleep in three inbred strains of mice for which …

MicroRNAs (miRNAs) are a novel class of small noncoding RNAs that regulate gene expression at the post-transcriptional level and play a critical role in many important biological processes. Most miRNAs are conserved between humans and mice, which makes it possible to analyze their expressions with a set of selected array probes. Here, we report a simple array platform that can detect 553 nonredundant miRNAs encompassing the entire set of miRNAs for humans and mice. The platform features carefully selected and designed probes with optimized hybridization parameters. Potential cross-reaction between mature miRNAs and their precursors was investigated. The array platform was …

BACKGROUND: Long-term use of non-steroidal anti-inflammatory drugs (NSAIDs) is associated with a reduced incidence of Alzheimer's disease (AD). We and others have shown that certain NSAIDs reduce secretion of Abeta42 in cell culture and animal models, and that the effect of NSAIDs on Abeta42 is independent of the inhibition of cyclooxygenase by these compounds. Since Abeta42 is hypothesized to be the initiating pathologic molecule in AD, the ability of these compounds to lower Abeta42 selectively may be associated with their protective effect. We have previously identified R-flurbiprofen (tarenflurbil) as a selective Abeta42 lowering agent with greatly reduced cyclooxygenase activity that …

The easy accessibility of skin makes it an excellent target for gene transfer protocols. To take advantage of skin as a target for gene transfer, it is important to establish an efficient and reproducible delivery system. Electroporation is an established technique for enhancing plasmid delivery to many tissues in vivo. A critical component of this technique is the electrode configuration. Electroporation parameters were optimized for transgene expression with minimal tissue damage with a novel electrode. The highest transgene expression and efficiency of individual cell transformation with minimal damage was produced with eight 150 ms pulses at field strength of …

Renewed Selection For Heat Loss In Mice: Direct Responses And Correlated Responses In Feed Intake, Body Weight, Litter Size, And Conception Rate, J. M. Mcdonald, Merlyn K. Nielsen Jan 2007

Renewed Selection For Heat Loss In Mice: Direct Responses And Correlated Responses In Feed Intake, Body Weight, Litter Size, And Conception Rate, J. M. Mcdonald, Merlyn K. Nielsen

Department of Animal Science: Faculty Publications

Divergent selection in mice was renewed in 3 independent replicates for high (MH) and low (ML) heat loss. An unselected control (MC) was maintained in all replicates. Heat loss was measured for individual male mice for 15 h, overnight in direct calorimeters. After 16 initial generations of selection followed by 26 generations of relaxed selection, divergent selection resumed for 9 generations. The realized selection applied was very close to the maximum possible selection according to the criteria and protocol. Selection differentials were greater for high than for low selection due to greater variation in the MH line. When corrected for …

Measuring Ultrasonic Communication Between Mouse Pups And Adult Mother Mice, Katie Ludwig Jan 2007

Measuring Ultrasonic Communication Between Mouse Pups And Adult Mother Mice, Katie Ludwig

Undergraduate Research Symposium (UGRS)

Measuring ultrasonic communication provides us with a way to study parental influence on animals. In this study I measured the ultrasonic communication between mouse pups and two maternal females, one of which who had given birth to the pups and the other had raised them. I found that there was no significant difference between the amount of noise expressed by pups in response to each the biological mother and foster mother test groups. Mouse pups call to maternal females regardless of genetic relatedness. Communication in mice may be a more complicated model because of their communal nature.
