Immunoreactivity Of Infiltrating Macrophages During Gastric Ulcer Healing In Rats, Erkmen Tuğrul Epi̇kmen, Emrah İpek, Recai̇ Tunca
Turkish Journal of Veterinary & Animal Sciences
This study investigated a timeline of changes in the immunoreactivity of infiltrating macrophages during gastric ulcer healing. To this end, gastric tissue samples were obtained from an acetic acid-induced gastric ulcer model in rats on the 1st, 3rd, 7th, and 14th days of ulcer induction. Ulcerated gastric tissues were subjected to histologic and immunohistochemical evaluations for macrophage subtypes. The number of Iba-1+ macrophages in the gastric ulcer area significantly increased on day 3, reaching a maximum number on day 7, followed by a decrease on day 14. No Gal-3+ macrophages were seen in the gastric ulcer area until day 14. …

Primary goat synovial cells were developed to be cocultivated with caprine arthritis encephalitis virus (CAEV)-infected monocytes. Two CAEV-seropositive goats and one CAEV-seronegative goat were confirmed for viral particles in the blood using the PCR method. Thereafter, heparinized blood samples from the three goats were collected and isolated for monocytes, which were further cultivated to differentiate into macrophages. Cultivated macrophages of 7-10 days old were cocultivated with the primary goat synovial cells for 7 days. Microscopic examination was performed every 2 days for multinucleated syncytial formation, and immunofluorescence assay was applied using monoclonal antibody against CAEV on day 7 to detect …

Macrophage-derived foam cells are the predominant component of arterial plaques in the early stages of atherosclerosis. The deposition of arterial plaques is effected by several factors that are influenced by a person’s daily nutritional habits. One factor that poses a major risk for plaque development is high levels of plasma LDL resulting from the consumption of a high-fat meal. In order to understand how an individuals’ diet effects arterial plaque deposition via the process of foam cell formation, we measured the acute response in circulating monocyte activity after consuming a high-fat meal. Samples were acquired on a FlowSight (EMD Millipore) …

Transmission Electron Microscopic Study Of Renal Haemopoietic Tissues Of Channa Punctatus (Bloch) Experimentally Infected With Two Species Of Aeromonads, Rajarshi Ghosh, Sumit Homechaudhuri
Turkish Journal of Zoology
The ultrastructural pathological changes in the renal haemopoietic tissue of Channa punctatus (Bloch) experimentally infected with 2 species of aeromonads, Aeromonas hydrophila and Aeromonas salmonicida, were reported. Adult C. punctatus specimens were injected intramuscularly, once at a time, with a chosen sublethal dose (2 × 10^9 cfu mL^-^1) of aeromonads at 5 µL g^-^1 body weight of fish and were exposed to 72 h of duration. The leucocytes (especially macrophages and melanomacrophages) and other associated cells from the pronephric head kidney of aeromonad-infected C. punctatus were studied by fixed ultrathin sections for transmission electron microscopy. The sinusoidal cells, barrier cells, …

Sulphur dioxide as a criterion of air pollution supresses macrophage functions. Some immunological changes in diabetes mellitus have been detected. In the present study, we examined the effect of sulphur dioxide on macrophage phagocytic activity in diabetic rats. The animals were separated into four groups as: 1) Control, 2) Sulphur dioxide, 3) Diabetes mellitus, 4) Diabetes mellitus+sulphur dioxide. The first and the third groups of animals were respirated with filtered atmosphere air in a special chamber one hour daily for eight weeks, while the other groups were exposed to 10 ppm sulphur dioxide in the same conditions and periods. The …
Retinal Changes In A Mutant Form Of Goldfish With Megalophthalmia, David T. Yew, Winnie W. Y. Li, C. Au, H. L. Choi, Q. D. Yang, Patrick K. H. Chan
Retinal Changes In A Mutant Form Of Goldfish With Megalophthalmia, David T. Yew, Winnie W. Y. Li, C. Au, H. L. Choi, Q. D. Yang, Patrick K. H. Chan
Scanning Microscopy
The retinal changes of a mutant strain of goldfish with megalophthalmia were studied by histology, electron microscopy and biochemistry. Changes in the morphology of the pigment epithelium, decrease in number of photoreceptors, thinning out of retinal layers and the existence of spaces in the retina were all features as the eyes grew in size. Invasion of macrophages was also evident in the retina. A decrease in leucine uptake per milligram of retina was also detected as the eye grew beyond 0.8 millilitre in volume. These changes, although related to volume changes (i.e., growth) of the eyes, were found to have …
Scanning Electron Microscopy Of Early Atherosclerosis In Rabbits Using Aortic Casts, Ralph G. Kratky, Margot R. Roach
Scanning Electron Microscopy Of Early Atherosclerosis In Rabbits Using Aortic Casts, Ralph G. Kratky, Margot R. Roach
Scanning Microscopy
Our research involves measuring the size and location of atherosclerotic lesions on the intimal surface of arteries. To this end we have developed a new method in which scanning electron micrographs of vascular casts with impressions of these lesions on their surface are used. This method is compared with light microscopy and scanning electron microscopy of tissue with lesions. All three methods are found to detect lesions equally well. We also examine the cellular elements in the lesion to determine how the images are formed.

Expression Of Major Histocompatibility Complex Antigens On Macrophages: Correlative Study Using Flow Cytometry, Radioimmunoassay, And Colloidal Gold Immunolabeling, Lynne E. Guagliardi, Donna M. Paulnock, Ralph M. Albrecht
Scanning Microscopy
Correlative scanning electron microscopy (SEM), radioimmunoassay (RIA), and flow cytometric analysis were used to characterize levels of class I and class II major histocompatibility complex-encoded (MHC) antigen expression on peritoneal exudate cells of mice chronically infected with Chlamydia psittaci. Analysis of peritoneal macrophages by all three techniques revealed a marked induction of H-2 K,D (class I) and I-A, l-E (class II) antigens on cells from infected C3H mice when compared to uninfected controls.
Scanning electron micrographs further document that the increases in class I and II MHC antigens are due to an increase in la/H-2 bearing cells as well …
