Precise regulation of gene expression is required for embryonic stem cell (ESC) differentiation. Transcription factor (TF) networks coordinate the balance of pluripotency and differentiation in response to extracellular and intracellular signals. Chromatin factors work alongside TFs to achieve timely regulation of gene expression for differentiation process. Our previous studies showed that a member of the Sin3a corepressor complex, Arid4b, is critical for proper mouse ESC differentiation into mesoderm and endoderm. We found elevated histone 3 lysine 27 acetylation (H3K27Ac) in a subset of genomic loci in meso/endoderm directed arid4b? cells, coincident with their derepression. DNA methylation is one of the major epigenetic modifications influencing the regulation of gene expression. The opium poppy is an important medicinal plant. Its latex contains opium, which is a rich source of pharmaceutical benzyisoquinoline alkaloids (BIA). Here, the methylation-sensitive amplification polymorphism (MSAP) profiling technique using 21 MSAP molecular markers was applied in order to compare levels of DNA methylation between 6 opium poppy varieties. MSAP profiling reflected the different methylation statuses among opium poppy varieties having divergent BIA content. We have previously shown that DNA damage induced by doxorubicin was associated with an increase in the level of DNA methylation, 5meC, in mouse embryonic fibroblasts using a developed immunostaining methodology. Doxorubicin (also known as adriamycin), which induces double-strand DNA breaks, has been widely used in cancer therapy. Cytosine modifications are dynamically regulated in the genome during lifespan, but abnormal changes in the 5meC pattern can be associated with diseases in particular cancer progressions and DNA damage. Multicellular organisms comprise a variety of cell types that have the same genotype but distinct phenotypes. This diversity is possible because of the establishment of a specific epigenetic landscape that stabilizes gene expression profiles that are exclusive for a particular cell type. Accumulating data indicate that the maintenance, loss, and reacquisition of pluripotency are dynamically regulated by epigenetic alterations evoked by a subset of cellular factors. A better understanding of the epigenetic mechanisms involved in stem cell biology and differentiation will improve our ability to use these cells in the clinical context.
