Insulin Sensitivity And The Onset Of Hyperphagia In Fatty Rats, Holiday Agnes Durham Dec 2004

Insulin Sensitivity And The Onset Of Hyperphagia In Fatty Rats, Holiday Agnes Durham

Masters Theses

Hyperphagia emerges suddenly after 22 days of age in Zucker fatty (fa/fa) rats, and fatties become visibly obese within days. This rapid increase in growth rate is likely to be associated with substantial changes in glucose metabolism. The objective of these experiments was to determine the relationship between insulin resistance and the onset of hyperphagia. Insulin tolerance tests showed that fatties were remarkably insulin resistant prior to the onset of hyperphagia, hyperinsulinemia, and obesity, but become as insulin sensitive as their lean littermates after the onset of hyperphagia. Furthermore, fatties became increasingly insulin resistant prior to the onset of …

Antimicrobial Activity Of Trypsin And Pepsin Hydrolysates Derived From Acid-Precipitated Bovine Casein, Kellie Parks Burris

Masters Theses

Foodborne pathogens are a major concern to the food industry and consumers but they may be controlled with antimicrobials. Naturally occurring antimicrobials may be isolated from a variety of plant, animal and microbial sources. Previous studies have demonstrated that peptides isolated from enzyme hydrolyzed milk proteins may have in vivo and in vitro antimicrobial activity. Such compounds could be of use as inhibitors of foodborne pathogens. The objectives of this study were to determine the antimicrobial effectiveness against Salmonella Typhimurium and Listeria monocytogenes of digests of bovine acid-precipitated casein with the enzymes pepsin and trypsin and to determine if these …

Relationships Between Muskrats And Freshwater Mussels Prior To River Otter Restoration At Mammoth Cave National Park, Kentucky, Kimberly Ann Asmus

Masters Theses

The southeastern United States has a great diversity of freshwater mussel species, many of which are threatened with extinction. The muskrat (Ondatra zibethicus) is one of the few animals to extensively prey on freshwater mussels, and it has been implicated in retarding the recovery of, or further threatening, some endangered mussel populations. Muskrat predation on mussels may be significant at Mammoth Cave National Park (MCNP) where a river otter (Lontra canadensis) restoration has been proposed. Anecdotal observations suggest that the reintroduction of river otters dramatically reduces muskrat populations, which, in tum, may reduce the number of …

Levels And Patterns Of Genetic Diversity In The Rare And Endangered Cumberland Stitchwort, Minuartia Cumberlandensis (Caryophyllaceae), Charles Thomas Winder

Masters Theses

Sequences of a highly variable nuclear gene (G3pdh) were used to characterize genetic diversity within and among populations of the endangered rockhouse endemic, Minuartia cumberlandensis (Wofford and Kral) McNeill (Caryophyllaceae), and compared to a widespread and abundant related species, M. glabra (Michaux) McNeill. By reconstructing phylogenetic relationships among G3pdh variants (haplotypes) and observing the geographical distribution of those ordered variants, an attempt was made to gauge the effects of historical and contemporary population processes acting within the species, particularly those with potential implications for long-term conservation. Both M. cumberlandensis and M. glabra were found to have high overall …

Development And Characterization Of Monoclonal Antibodies Directed Against Tanapox Virus, Michael Franz Dec 2004

Development And Characterization Of Monoclonal Antibodies Directed Against Tanapox Virus, Michael Franz

Masters Theses

The initial symptoms of a Tanapoxvirus (TPV) infection are indistinguishable from smallpox, which is caused by variola virus (VV). In the post-9/11 area, reagents that can quickly differentiate a TPV infection from smallpox are very desirable, but currently unavailable. Monoclonal antibodies (mAbs) directed specifically against TPV can be used for this purpose. Six mAbs were developed and characterized that specifically bind to TPV infected owl monkey kidney (OMK) cells, but not uninfected cells. The apparent molecular mass of the TPV protein(s) recognized by 5 mAbs were calculated using the Western blot technique. One mAb demonstrated no binding using the Western …

The Dynamics, Distribution, And Activity Of Viruses In Lake Erie, Amanda Dean Dec 2004

The Dynamics, Distribution, And Activity Of Viruses In Lake Erie, Amanda Dean

Masters Theses

As viruses-- specifically pervasive pathogens of aquatic microorganisms-- are now included in most studies of aquatic food webs and nutrient cycles, presented here is new information from samples collected in July 2003 which demonstrates that bacteria lysed by viruses released 122 - 1080 nM of phosphorus per day throughout Lake Erie, implying that viruses acted as a major mechanism of phosphorus recycling in the lake. Viruses have been shown to be ubiquitous throughout Lake Erie, with a weak correlation between their distribution and the distribution of bacteria. Moreover, evidence suggests that virus production rates are tightly tied to the growth …

Maximizing Northern Red Oak (Quercus Rubra) Seedling Growth To Sustain Oak-Dominated Ecosystems In East Tennessee, Brian Andrwe Barwatt

Masters Theses

The success of northern red oak (Quercus rubra L.) in the oak-hickory forest type and its growth and development into the overstory is of great concern due to the value of this species as a source of forest products and mast for wildlife. There is a general consensus that rapid oak seedling height growth occurs above certain threshold levels of light, soil moisture, and nutrients. If these specific levels and the relative importance of competition between northern red oak (NRO) seedlings and other individual plant species can be determined, then the implementation of more precise management practices that promote …

Molecular Weight And Degree Of Acetylation Of Ultrasonicated Chitosan, Shari Rene Baxter Aug 2004

Molecular Weight And Degree Of Acetylation Of Ultrasonicated Chitosan, Shari Rene Baxter

Masters Theses

Chitosan is a glucosamine polymer produced by deacetylation of chitin from crustacean shells. The functional properties of chitosan, such as thickening, film-formation and antimicrobial activity, are related to its molecular weight and degree of acetylation (DA). High intensity ultrasonication has the potential to modify molecular weight of chitosan and thus alter or improve chitosan functional properties. The objective of this research was to determine the DA and molecular weight of chitosan molecules as a function of sonication intensity and treatment time.

High molecular weight shrimp chitosan was purified by alkaline precipitation and dialysis from aqueous solution. A 1 % (w/v) …

Identifying The Signature Of The Natural Attenuation Of Mtbe In Groundwater Using Molecular Methods And "Bug Traps", Anita Eva Biernacki Aug 2004

Identifying The Signature Of The Natural Attenuation Of Mtbe In Groundwater Using Molecular Methods And "Bug Traps", Anita Eva Biernacki

Masters Theses

Natural attenuation through intrinsic bioremediation is the risk-based management approach commonly used for gasoline (BTEX) contamination sites. This approach has not yet been utilized for the fuel oxygenate methyl tertiary-butyl ether (MTBE). MTBE is more resistant to biodegradation than BTEX. MTBE is more abundant than benzene in oxygenated gasoline, has a greater water solubility than BTEX, and sorbs weakly to soil. These properties complicate developing a risk based management option to be implemented as easily as for BTEX. The purpose of this project is to contribute to a growing database containing information on MTBE contaminated sites nationwide with a variety …

Mycorrhizal Symbiosis And The Response Of Sorghum Plants To Combined Drought And Saline Stresses, Keun Ho Cho Aug 2004

Mycorrhizal Symbiosis And The Response Of Sorghum Plants To Combined Drought And Saline Stresses, Keun Ho Cho

Masters Theses

Arbuscular mycorrhizal (AM) symbiosis has been reported often to improve the abilities of host plants to tolerate drought stress. The physiological mechanism is uncertain, but one idea is that the effect might be linked to resistance to salt stress. Several studies have shown more growth in crop plants colonized with AM fungi than nonAM control plants under salt stress. Drought and salt stresses frequently occur together in nature and their initial symptoms in plants are similar. It may be interesting to scrutinize their physiological interaction in host plant as a function of AM fungi. Therefore, the objectives of my studies …

Sol-Gel Encapsulation Of A Bioluminescent Bioreporter, Elizabeth F. Mitchell Aug 2004

Sol-Gel Encapsulation Of A Bioluminescent Bioreporter, Elizabeth F. Mitchell

Masters Theses

Genetic engineering has allowed for the development of a number of whole-cell bacterial bioreporters. In order for these cells to be used in certain devices and field applications, they must be maintained, and protected, but exposed to environmental conditions. One approach used to accomplish this goal is cellular encapsulation. The recent development of techniques to form inorganic matrices, in particular through the sol-gel process, displays promise as a system of encapsulation. The silica sol-gel process was originally designed for the fabrication of glass, and given that reactions are performed in standard lab conditions and room temperature, the process can be …

Prescribed Burning In Tennessee: Importance And Barriers, Goals, And Information Needs Of Private, State, And Federal Managers, Brian T. Hemel Aug 2004

Prescribed Burning In Tennessee: Importance And Barriers, Goals, And Information Needs Of Private, State, And Federal Managers, Brian T. Hemel

Masters Theses

Early in the 20th century, intense and severe wildfires were frequent due to abundant fuels generated by logging and the presence of anthropogenic ignition sources in close proximity to these fuels. As timber was depleted, this period of lumbering ceased and the realization that vast tracts of cutover land had to be reforested and protected led to the inception of fire suppression organizations and techniques. The fire exclusion that occurred over much of the 20th century due to organized fire suppression efforts has led to two sets of problems: the buildup of fuels and the decline of fire-adapted …
