Over the span of five decades, staggering progress has been made in elucidating the neural circuits involved in fear learning. Research focused on rodents and healthy individuals has been instrumental in elucidating how abnormalities in this circuitry may lend to increased prevalence of various fear and anxiety disorders. Transcription termination factor Rho is an essential protein in many bacteria. Rho is an ATP-dependent RNA helicase that accounts for 20-50% of termination events in Escherichia coli and plays an important role in preventing pervasive transcription that otherwise might be lethal to bacteria. Rho termination usually occurs in the 3’ UTRs and non-coding regions of the DNA, and the actively transcribing RNA is believed to be protected by translating ribosomes from Rho termination since both transcription and translation are a simultaneous process in prokaryotes. 1,4-dioxane is a volatile organic compound categorized by the United States Environmental Protection agency (EPA, 2017) as a possible human carcinogen potent to cause tumors in humans. Even though this compound poses significant environmental danger when it contaminates water sources, it is surprisingly less studied, and how it behaves in surface water is less understood. Staphylococcus aureus is a common commensal and opportunistic pathogen of humans. It causes a variety of diseases, ranging from skin and soft infections to life-threatening invasive diseases. Many strains of S. aureus have developed resistance to a variety of antibiotic classes, including many beta-lactams. Primates show diverse patterns of adaptive color and body size dimorphism produced by inter- and intrasexual selection. However, the specific microevolutionary processes that produce variation in secondary sexual characteristics remain largely unexplored in primates. Furthermore, sexual conflict theory predicts that female and male secondary sexual traits can coevolve in an antagonistic manner and promote speciation. This dissertation explores the microevolution of secondary sexual characteristics in mandrills (Mandrillus sphinx) and the macroevolution of these characters in anthropoid primates. In this dissertation, I aim to forecast high utilizers of emergency care and inpatient Medicare services (i.e., healthcare visits). Through a literature review, I demonstrate that accurate and reliable prediction of these future high utilizers will not only reduce healthcare costs but will also improve the overall quality of healthcare for patients. Multiplexed fluorescent imaging (MFI) has transformed biological investigation by providing spatial context to labeled analytes, such as DNA, RNA, and proteins. Multiplexed imaging techniques can track multiple analytes at once and thus has the potential to provide an unprecedented view into the complex relationships between various molecules, cells, and tissues. However, most researchers rarely use more than 3 fluorophores at a time due to the complex technological and procedural requirements for highly multiplexed imaging. Childhood growth has been a means of identifying systemic, material inequalities within populations. The plasticity of growth is responsive to multiple environmental factors, most notably adequate nutrition, but also psychosocial stress. Psychosocial stress can be a function of the social and political economic ecology, reinforcing power hierarchies within societies. This dissertation proposes that childhood growth can also reflect inequality in the distribution of psychosocial stress exposure through mothers’ feelings of a chronically stressful environment conveyed through maternal-infant interactions.

Glutamate receptors act to bring about excitatory transmission in the central nervous system. The receptors are divided into two groups: ionotropic and metabotropic glutamate receptors. Ionotropic glutamate receptors are ion channels which are activated by an agonist such as glutamate or kainate. The main receptors in the ionotropic glutamate receptor family are the α-amino-3-hydroxy-5-methyl-4-isoxazolepropionic acid (AMPA), kainate and N-methyl-D-aspartate (NMDA) receptors. In the central nervous system ionotropic glutamate receptors are found both pre- and postsynaptically. Germline stem cells differentiate into mature egg or sperm cells that go through the process of fertilization which gives rise to all sexually reproducing organisms. During this process of differentiation, germ cells undergo a switch from mitosis to meiosis that allows for proper development and specification of the future gamete. The mechanisms that facilitate this shift from mitosis to meiosis, however, are not well understood. To gain insight into this process, we used Drosophila oogenesis as our model. Group II introns are self-splicing mobile elements that are thought to be the evolutionary ancestors of eukaryotic retrotransposons, the spliceosome, and spliceosomal introns. Yet, little is known about how group II introns have coevolved within their native hosts or about the corresponding host biology.Using the Ll.LtrB intron in its native host, Lactococcus lactis, I first sought to identify host factors that influence retrotransposition. Ricin is a highly lethal protein toxin derived from the seeds of the castor plant, Ricinus communis. It is a Type II ribosome inactivating protein (RIP), meaning it is a heterodimer with one subunit, ricin toxin B (RTB), that mediates cell surface attachment and intracellular trafficking and a second subunit, ricin toxin A (RTA), that irreversibly shuts down protein synthesis in the cytosol. During trafficking, RTA and RTB necessarily separate in the endoplasmic reticulum, wherein RTA unfolds and translocates into the cytosol where it refolds into an enzymatically active conformation. In this dissertation, I review the fundamental processes and mechanisms for bacterial DNA replication, especially the mechanisms employed by high-fidelity DNA polymerases to replicate the genome. Unlike the prototypical bacterial system from Escherichia coli which uses a single C-family polymerase, DNA polymerase IIIα (Pol IIIα), to replicate the genome, low-GC content Gram-positive bacteria utilize two essential C-family polymerases, PolC and DnaE. PolC and DnaE work cooperatively to replicate the genome, with DnaE initiating synthesis from RNA-primers and PolC performing the bulk synthesis. The epitranscriptome is composed of enzyme catalyzed RNA modifications that can regulate tRNA, mRNA, and rRNA structure and function. RNA modifications can play key roles in regulating gene expression by controlling the when and how much of translation of specific transcripts. The SelU catalyzed tRNA wobble uridine (U) modification, occurs on tRNA lysine, glutamic acid, and glutamine. We have shown that E. coli deficient in selU gene are more sensitive to the antibiotic chloramphenicol (CAM) than their wild type counterparts. This thesis describes a chemical method for alternative oligonucleotide purification that is non-chromatographic and gel-free and allows to routinely synthesize and purify long functional RNA strands. The purification of long RNAs is based on the bio-orthogonal inverse electron demand Diels-Alder (IEDDA) chemistry between trans-cyclooctene (TCO) and tetrazine (Tz). Target oligonucleotide strands are selectively tagged with Tz and can be captured and purified from the failure sequences with immobilized TCO. RNA strands are synthesized on solid support through a photolabile linker to avoid the loss of Tz tag. This dissertation examined the complex developmental impact of the clinically relevant synthetic progestin 17α-hydroxyprogesterone caproate (17-OHPC) on medial prefrontal cortex (mPFC) development. In rodents, the effects of 17-OHPC are subtle, but significant. In the Chapter II, we observed that 17-OHPC abolishes sex differences in dopaminergic fiber innervation and alters microglia phenotype in the prelimbic and infralimbic areas by the end of the first postnatal week. In the third chapter, the overall effect of 17-OHPC on the ontogeny of dopaminergic fiber innervation and synaptic bouton density were used to characterize the timing and duration of treatment effects in the mPFC. Phosphodiesterases (PDEs) are integral components of 3’,5’-cyclic adenosine monophosphate (cAMP) signaling pathways by degrading cAMP to modulate the concentration, duration, and localization of the cAMP signal which maintains the specificity of cAMP pathways. The human pathogen, Mycobacterium tuberculosis (Mtb) has a unique cAMP network architecture with at least 15 adenylyl cyclases (ACs) that generate cAMP, but only one characterized PDE, Rv0805, which is found exclusively in pathogenic mycobacteria. Major depressive disorder is one of the most pervasive psychiatric illnesses in the United States. Women are at greater risk for developing depression, particularly during their childbearing years. Approximately 17% of new mothers develop postpartum depression within 4 weeks after parturition. The risk for postpartum depression is even greater in women who do not breastfeed or stop breastfeeding early. Major depressive disorder and postpartum depression share the same symptomology and common etiological bases. In 1969, the enrichment of mobile element repeat sequences led Britton and Davidson to propose the hypothesis that gene expression in higher eukaryotes is regulated through the exaptation of mobile elements. In this work, I have explored the hypothesis that mobile elements can also be harnessed by the host to regulate asymmetric cell division, thereby determining cell fate. In this dissertation I provide a pre-steady-state kinetic characterization of an antibiotic-resistant mutant of a Staphylococcus aureus DNA polymerase. Staphylococcus aureus is one of the most common causes of infections in humans, and is widely known for its ability to acquire resistance to most antibiotics. Staphylococci infections pose a significant burden to the healthcare system and increase mortality, as more than 95% of Methicillin-Resistant S. aureus (MRSA) infections do not respond to first-line antibiotics.
