Valorization of Xylan in Agroforestry Waste Streams.

Microwave-assisted deep eutectic solvent and gamma-valerolactone metallic chloride catalyzed conversion of xylan to furfural were investigated using a 2x3 factorial experimental design at two levels of percent microwave power, reaction time, and catalyst concentration. The levels of each factor studied were (20%, 60% microwave power, 2, 4 minutes, and 10, 20mg) respectively. The effect of three metallic chloride catalysts (LiCl, FeCl3.6H20, CuCl2) on the conversion of xylan to furfural was also investigated. The Mediterranean region of northern California is projected to get increasingly warmer under all Intergovernmental Panel on Climate Change (IPCC) emission scenarios, with future precipitation projections not showing much of a trend. This poses a problem to the already dry summers that are experienced in the Mediterranean region of California. If precipitation does not increase alongside temperatures, the dry seasons will likely only get drier. Soils in the Monongahela National Forest (MNF) are acidic due to sandstone parent material, acid deposition, uptake of base cations by vegetation, and release of organic acids by organic matter (OM) decomposition. Increases in soil acidity have caused declines in forest health and changed species composition and nutrient status. Liming can neutralize soil acidity, but no large-scale liming projects have been done on acid forest soils in the USA. The hemlock woolly adelgid (HWA), Adelges tsugae Annand (Hemiptera: Adelgidae), is an invasive insect that is causing mortality of eastern hemlock trees, Tsuga canadensis, and Carolina hemlock trees, Tsuga caroliniana, across the eastern United States. To protect these ecologically important tree species, a neonicotinoid insecticide, imidacloprid, is commonly used. Imidacloprid is an effective treatment and can remain effective against HWA for four to six years but long-term (≥ one year after application) non-target effects of imidacloprid on forest ecosystems are not well-studied. There is a growing concern regarding the microbial safety of farmers’ markets as fresh produce consumption in the United States is gaining popularity. The CDC reported 46% of foodborne illnesses are related to fresh produce. Consuming fresh produce raw removes a processing step, increasing the risk of cross-contamination, leading to illness. However, this risk can be mitigated by washing produce with antimicrobial solutions during post-harvest processes. Previous studies on one of the horned beetle species, Onthophagus taurus, that utilized comparative gene expression analyses, suggested legs to be a strong candidate of the origin of horns. Little is known about how above-ground vegetation may differentially influence the below-ground microbial community structure, abundance, and function. Abundance of soil microbial N cycling genes responsible for nitrification (amoA), denitrification (nirK and nirS), and nitrous oxide reduction (nosZ) may vary with tree species and increasing N availability, and these variables may be used to predict production of NO3- and N2O from soil. Refined Liquid Smoke (RLS) (1%) was incorporated during osmotic dehydration of apple cubes (10 mm each side) to observe its effect on solid gain, water loss, yield, water activity, product quality, and consumer preference. Three different treatments, POD (pure osmotic dehydration, no RLS), LSISS (RLS inside sugar solution) and PTLS (pre-treatment with RLS), were compared in this study. Osmotic dehydration was carried out using 42 °Brix sugar solution, and the fruit to solution ratio was kept at 1:4. The blood-brain barrier (BBB) is a critical interface between the systemic circulation and the brain. It is a specialized multicellular unit composed of brain microvascular endothelial cells (BMECs), pericytes, a basement membrane, and astrocytic end foot processes. BMECs are a principal component of the BBB that provide the structural framework needed for the stringent transport of molecules into the brain. BMEC dysfunction permits the trafficking of neurotoxins from systemic circulation into the brain, which ultimately exacerbates BBB dysfunction and neuroinflammation. Perineuronal nets (PNN) are a type of specialized extracellular matrix in the central nervous system. The PNN forms during postnatal development but the ontogeny of the PNN has yet to be elucidated. Studying the PNN in the rat brain may allow us to further understand the PNN’s role in development, learning, and memory. The PNN is fully developed in the deep cerebellar nuclei (DCN) of rats by post-natal day 18. By using enzymatic digestion of the PNN with chondroitinase ABC (ChABC), we studied how digestion of the PNN affects cerebellar-dependent eyeblink conditioning (EBC) and performed electrophysiological recordings from DCN neurons. …

The Ohio River is continuing to recover from a long history of anthropogenic impacts. Blue Catfish were considered locally extirpated from the West Virginia sections and have been reintroduced through fingerling stockings beginning in 2002. Flathead Catfish are naturally sustaining in the pool and combined with Blue Catfish provide two popular recreational fisheries. The Blue Catfish and Flathead Catfish studies shared similar objectives: (1) evaluating size structure, (2) estimating growth rates, and (3) estimating annual mortality. Modern shallow brine environments contain an abundance of microorganisms. Microorganisms, mineral crystals, and air bubbles can be trapped with those brines in primary fluid inclusions in bedded halite as it grows in shallow saline surface waters. Primary fluid inclusions and their contents can be preserved, unaltered, for millions of years.

The ~830 million-year-old Browne Formation of central Australia contains intervals of bedded halite with unaltered primary fluid inclusions. Metastatic breast cancer causes the vast majority of cancer-associated deaths, especially in triple negative breast cancers (TNBC). TNBC is still poorly understood and has no effective treatment. Here we reveal that presence of Aurora-A Kinase (AURKA) in the nucleus and metastatic dissemination are molecularly connected through HIF1 (Hypoxia induced factor-1) signaling. The nuclear AURKA in the complex with constitutively expressed HIF-1β subunit activates transcription of “hypoxia induced genes” under normoxic conditions (the phenomenon called pseudohypoxia) without upregulation of oxygen-sensitive HIF-1α subunit. We uncover that AURKA preferentially binds to and phosphorylates HIF-1β, and co-localizes with HIF complex on DNA. The mass …

The primary objective of my thesis was to develop and utilize a biocompatible multifunctional trityl spin probe for concurrent measurement of pO2, pHe, and [Pi] in vivo by electron paramagnetic resonance (EPR) spectroscopy (Chapter 2). My first goal was to synthesize the proposed probe we are terming HOPE71. Secondly, HOPE71 was characterized by X-band and L-band EPR spectroscopy. Pseudomonas aeruginosa is a Gram-negative opportunistic pathogen which can cause severe, recurrent, and chronic infections. The pathogen is highly adaptable, and pneumonia caused by it is associated with high morbidity and mortality, especially in individuals with the genetic disorder cystic fibrosis. Antibiotic resistance amongst clinical isolates of P. aeruginosa is steadily increasing, and multi-drug resistant strains are prevalent. There is currently no vaccine available for commercial use against P. aeruginosa.
