Isolation, Purification, And Chemical Characterization Of The Dihydroxamate-Type Siderophore, "Schizokinen," Produced By Rhizobium Leguminosarum Iari 917., Erin P. Storey

Electronic Theses and Dissertations

Iron is essential to the majority of microorganisms; it is an important cofactor in many cellular processes and enzymes. However in an aerobic environment and at biological pH, iron is primarily found as insoluble oxyhydroxides and is unavailable to microorganisms. Many bacteria have the ability to produce siderophores, low molecular weight compounds that have a high affinity for Fe3+. Siderophores are part of a multi-component system that actively transports the iron-siderophore complex into the cytoplasm. Rhizobia are characterized by their ability to form symbiotic relationships with leguminous plants, where they can fix nitrogen for the host plant and …

Re-Integrating Crops And Livestock In Maine: An Economic Analysis Of The Potential For And Profitability Of Integrated Agricultural Production, Aaron K. Hoshide

Electronic Theses and Dissertations

This thesis examines the profitability of and sustainability indicators for potato and dairy farms in Maine integrating crops and livestock in two different ways. The first is inter-farm coupling, where two or more specialized producers are close enough to exchange manure applications for crops used as livestock feed. Land base is shared between farmers. The second is where farms are on-farm integrated. Here crops and livestock are raised on the same farm and manure is applied to cash crops and livestock feed crops. Face-to-face interviews with Maine producers were used to construct integrated and non-integrated representative budgets. Assuming …

Dietary Patterns Of Mediterranean Adolescents, Sandra Ellan Brussee Aug 2005

Dietary Patterns Of Mediterranean Adolescents, Sandra Ellan Brussee

Electronic Theses and Dissertations

The Mediterranean Diet (MD) is renowned for its health benefits. However, adherence appears to be lessening in the Mediterranean region as rates of overweight, obesity, and chronic disease are increasing. The adoption of a "western diet" (high amounts of meats, refined grains, snacks, and fast-food products) has been termed the "Nutrition Transition". Adolescents appear to be the most affected; therefore their diets and eating patterns were examined in this study. The objectives were to study food group-based consumption of adolescents, compare average servings with recommendations from the USDA Food Guide Pyramid (FGP) and the MD Pyramid, identify and compare eating …

Implications Of Spatial Autocorrelation And Dispersal For The Modeling Of Species Distributions, Volker Bahn Aug 2005

Implications Of Spatial Autocorrelation And Dispersal For The Modeling Of Species Distributions, Volker Bahn

Electronic Theses and Dissertations

Modeling the geographical distributions of wildlife species is important for ecology and conservation biology. Spatial autocorrelation in species distributions poses a problem for distribution modeling because it invalidates the assumption of independence among sample locations. I explored the prevalence and causes of spatial autocorrelation in data from the Breeding Bird Survey, covering the conterminous United States, using Regression Trees, Conditional Autoregressive Regressions (CAR), and the partitioning of variance. I also constructed a simulation model to investigate dispersal as a process contributing to spatial autocorrelation, and attempted to verify the connection between dispersal and spatial autocorrelation in species' distributions in empirical …

The Effect Of Dietary Manganese On Arterial Functional Properties, Anastasia Z. Kalea Aug 2005

The Effect Of Dietary Manganese On Arterial Functional Properties, Anastasia Z. Kalea

Electronic Theses and Dissertations

Dietary manganese affects the structure and integrity of blood vessels, as well as vessel predisposition to endothelial dysfunction and cardiovascular disease. In this thesis, we studied the role of manganese on the functional properties of rat aorta as defined by the endothelial and vascular smooth muscle cell pathways for adrenergic-mediated vasoconstriction and cholinergic-mediated vasodilation. Weanling Sprague-Dawley rats were fed a manganese deficient (MnD), adequate (MnA-control group) or supplemented (MnS) diet (<1, 10-15 and 45-50ppm Mn respectively). After 14 weeks on the diet the aorta was excised and four aortic rings of three mm length were prepared from each animal. Alterations in vasoconstriction among diet groups were detected by dose-response curves to the ^-adrenergic agonist LPhenylephrine in endothelium-intact and endothelium-disrupted rings. Alterations in endothelium-dependent vasodilation among diet groups were determined by doseresponse curves to Acetylcholine. We studied the mechanism by which dietary manganese affects two different endothelium-dependent vasodilation pathways: the Larginine/ nitric oxide (NO) and the cyclooxygenase (COX) pathways. Inhibition of the enzymes for NO synthesis (NOS) with L-NMMA, and of prostanoids (COX I and II) with Mefenamic acid, determination of NOS expression, and in vitro addition of L-Arginine (substrate for NO formation) to vessel rings revealed the effect of manganese on the regulation of endothelium-mediated vasodilation and vasoconstriction. Dose-response curves to sodium nitroprusside provided data for the dietary effect on endotheliumindependent vasodilation. Supplementary dietary manganese increased adrenoreceptor-mediated vascular smooth muscle contraction, which was significantly reduced in the presence of functional endothelium. Absence of dietary manganese increased endothelial cell sensitivity to the (Xi-adrenergic vasoconstrictor agent. Manganese had a small effect on the cGMP-pathway for dilation of vascular smooth muscle but affected vasodilation primarily through an endothelium-mediated pathway, probably by preserving NO bioavailability. Inhibition of vasodilation in Mn deficiency appears to occur through an endothelium-derived vasoconstrictor, possibly thromboxane with a concomitant decrease in the synthesis of endothelium-mediated vasodilator prostanoids. Our results demonstrate that dietary manganese influences the contractile machinery of vascular smooth muscle cells and regulates the bioactivity of endothelium-mediated vasodilators to affect agonist-induced signaling pathways that participate in the regulation of vasomotor tone. This suggests possibilities for dietary intervention in blood pressure regulation.

Dynamics Of Bee And Wasp Populations In Maine Lowbush Blueberry (Vaccinium Angustifolium), Joseph E. Karem

Electronic Theses and Dissertations

Conservation of natural enemies can be an effective form of pest management. If beneficial Hymenoptera, native to the area, can be protected and encouraged to multiply, the benefits of natural insect pest control might be realized. Hymenoptera as "natural enemies" as well as "pollinators" have been studied intensively in many agroecosystems worldwide. However, lowbush blueberry is not an ecosystem where ecology of Hymenoptera has been well studied. This thesis discusses two studies conducted in lowbush blueberry fields in Washington County, Maine in 1997 and 1998. In the first study, I investigated "towers" as a method for deploying insect traps along …

Using Domination To Analyze Rna Structures., Travis Reves Coake May 2005

Using Domination To Analyze Rna Structures., Travis Reves Coake

Electronic Theses and Dissertations

Understanding RNA molecules is important to genomics research. Recently researchers at the Courant Institute of Mathematical Sciences used graph theory to model RNA molecules and provided a database of trees representing possible secondary RNA structures. In this thesis we use domination parameters to predict which trees are more likely to exist in nature as RNA structures. This approach appears to have promise in graph theory applications in genomics research.

Primary Sex Reversal In Female Betta Splendens Following Treatments With Temperature And Trenbolone Acetate., Tracy S. Smith

Electronic Theses and Dissertations

Sex reversal in fish can occur as a result of disturbances in natural conditions or laboratory manipulations. Trenbolone Acetate (TBA), a synthetic anabolic-androgenic steroid used mainly in the cattle industry, was applied to female Betta splendens, a sexually dimorphic fish. Previously, females were analyzed for secondary sex characteristics after treatment with TBA, increased temperature, or a combination of both. This study focused on primary sex characteristics by histological examination. Gonads were surgically removed and prepared following basic histological techniques. Following H&E staining, sections were observed for signs of ovarian regression, as well as signs of testicular formation. The presence …

Characterization Of Microbial Communities In Karstic Sediments : A Study Of Cave Bacteria In Central Kentucky., Justine Kay Roback 1964- May 2005

Characterization Of Microbial Communities In Karstic Sediments : A Study Of Cave Bacteria In Central Kentucky., Justine Kay Roback 1964-

Electronic Theses and Dissertations

Few previous studies have compared microbial communities in subterranean and surface environments. Chemical analyses used to characterize the surface and cave microbial environments indicated limited exchange between surface and subsurface waters. Here, bacterial communities from four distinct cave sediments collected in central Kentucky and adjacent surface streams were compared using a combination of culture and molecular methods. Predictions were that: 1) Cave communities will have unique community structures when compared to those of nearby surface sediments, and 2) Genetic diversity of bacterial communities in cave sediments will be higher than those in nearby surface sediments. Since diversity and community structure …

Soil Microbial Community Dynamics In Florida Scrub Ecosystem, Maria Albarracin Jan 2005

Soil Microbial Community Dynamics In Florida Scrub Ecosystem, Maria Albarracin

Electronic Theses and Dissertations

Pyrogenic ecosystems are maintained by fires which vary in frequency, seasonality, and intensity. Florida oak-saw palmetto scrub ecosystem is characterized by fires occurring at intervals of 10-20 years. Diverse factors as private land acquisition and development has created a patchy distribution of scrub ecosystems and also interrupted the natural fire cycle. The effects of fire over plant regeneration and fauna habitat utilization of the scrub have been well characterized in previous research. In the present paper the objective is to characterize the short- and long-term fire effects on the soil microbial community. Fire effects were studied in a chronosequence, comprising …

Effects Of A Shore Protection Project On Loggerhead And Green Turtle Nesting Activity And Reproduction In Brevard County, Florida, Kelly Brock Jan 2005

Effects Of A Shore Protection Project On Loggerhead And Green Turtle Nesting Activity And Reproduction In Brevard County, Florida, Kelly Brock

Electronic Theses and Dissertations

Marine turtle reproductive success is strongly correlated with the stability and quality of the nesting environment. Because females show fidelity to key nesting beaches, the management and physical characteristics of these beaches directly affect future generations of marine turtles and may be essential for the recovery of these threatened and endangered species. The impacts of beach restoration on loggerhead turtles (Caretta caretta) and on green turtles (Chelonia mydas) were investigated. Previous studies concerning beach nourishment projects have focused on loggerhead turtles. I compared data between nourished and non-nourished areas and between loggerhead and green turtles. I found, at one season …

Juvenile Green Turtle (Chelonia Mydas) Foraging Ecology:Feeding Selectivity And Forage Nutrient Analysis, Eliza Gilbert Jan 2005

Juvenile Green Turtle (Chelonia Mydas) Foraging Ecology:Feeding Selectivity And Forage Nutrient Analysis, Eliza Gilbert

Electronic Theses and Dissertations

For the endangered green turtle, Chelonia mydas, a fundamental component of recovery and conservation is an understanding of its foraging ecology. Foraging optimality models suggest animals will select resources of high quality over those of low quality. For green turtles, this behavior is important, as sufficient quantities of nutritionally adequate forage items are necessary for growth and reproduction. One intrinsic element in the understanding of green turtle foraging ecology is to identify and document the availability and quality of forage resources preferred by green turtles. The objectives of this study were: 1) determine whether juvenile green turtles showed a feeding …

Historical Responses Of Marine Turtles To Global Climate Change And Juvenile Loggerhead Recruitment In Florida, Joshua Reece Jan 2005

Historical Responses Of Marine Turtles To Global Climate Change And Juvenile Loggerhead Recruitment In Florida, Joshua Reece

Electronic Theses and Dissertations

Marine turtle conservation is most successful when it is based on sound data incorporating life history, historical population stability, and gene flow among populations. This research attempts to provide that information through two studies. In chapter I, I identify historical patterns of gene flow, population sizes, and contraction/expansion during major climatic shifts. In chapter II, I reveal a life history characteristic of loggerhead turtles previously undocumented. I identify a pattern of juvenile recruitment to foraging grounds proximal to their natal nesting beach. This pattern results in a predictable recruitment pattern from juvenile foraging ground aggregations to local rookeries. This research …

Effects Of Food Deprivation On Blood Lipid Concentration And Composition In Steller Sea Lions (Eumetopias Jubatus), Michelle Lea Berman

Electronic Theses and Dissertations

Steller sea lions, the largest Otariid, fast during their breeding season; during this time they refrain from ingesting food for a period of 12-43 days. Fasting, while undertaking an extremely energetically demanding activity (breeding and pupping), requires specific physiological adaptations. This study examined the physiological response to fasting of two age classes, juveniles and sub-adults, during the breeding and non-breeding seasons to determine how these animals utilize lipids and the pattern of fatty acid mobilization from lipid stores during fasting. Four juveniles and 5 sub-adults were fasted for one and two weeks, respectively, and blood samples were collected approximately every …

Novel Protein-Protein Interactions Regulate The Proteolytic Activity Of The Pro- Apoptotic Serine Protease, Omi/Htra2, Supriya Singh

Electronic Theses and Dissertations

Omi/HtrA2 is a mitochondrial serine protease with high homology to the bacterial HtrA proteins. Omi promotes caspase-dependent apoptosis by binding and degrading IAPs-inhibitor of apoptosis proteins. Omi can also induce caspase-independent apoptosis but the actual mechanism is still unknown. IAP's are not the only substrates cleaved by Omi. There are at least two more known substrates of Omi, the HAX-1 and the ped/pea-15 proteins. HS1-associated protein X-1 (HAX-1) is a mitochondrial protein, degraded by Omi after induction of caspase-dependent apoptosis. Ped/pea-15 is also an anti-apoptotic protein and is cleaved by Omi after induction of caspase-independent apoptosis. The proteolytic activity of …

Responses Of The Florida Mouse (Podomys Floridanus) To Habitat Management, Jason Depue

Electronic Theses and Dissertations

The Florida mouse (Podomys floridanus), a species restricted to the Lake Wales Ridge and the Atlantic Coastal Ridge, is recognized by the Florida Fish and Wildlife Conservation Commission as a species of special concern, highlighting its status as a species that is threatened by habitat loss statewide. Publicly owned lands offer protection for the species, but management is generally focused on protecting biodiversity in general and not a particular species. The response of the Florida mouse to land management practices such as mechanical treatment and prescribed fires is poorly documented. This research examined the population responses of Florida mice on …

The Effects Of Arsenic On Selenoprotein Biosythesis, Fanta Konate Jan 2005

The Effects Of Arsenic On Selenoprotein Biosythesis, Fanta Konate

Electronic Theses and Dissertations

Arsenic contamination of drinking water is a real public health problem in certain areas of South-East Asia where chronic exposure has been correlated to higher rates of lung, skin, bladder, kidney, and liver cancer. Although arsenic carcinogenicity is well established, the mechanism by which it induces cancer is poorly understood. Recent evidence suggests that oxidative stress could be a possible mechanism for the carcinogenic effects of arsenic. Selenium, in the form of selenocysteine, is necessary for the activity of several enzymes with a role in the defense against reactive oxygen species (ROS), primarily thioredoxin reductases (TrxR) and glutathione peroxidases (Gpx). …

A Novel Binding Interaction For The Paxillin Ld3 Motif: Paxillin Ld3 Mediates Merlin-Paxillin Binding At Paxillin Binding Domain 1, Sandra E. Beattie Geden

Electronic Theses and Dissertations

Neurofibromatosis type 2, an autosomal dominant genetic disorder, causes predisposed individuals to develop various benign central and peripheral nervous system tumors. The characteristic tumors of this disease are schwannomas, which are tumors of the Schwann cells, typically on the vestibular nerve. These and the other associated tumors slowly compress nervous system structures causing deafness and loss of balance, resulting in an average life-span of less than 40 years. The product of the Nf2 gene is the protein named merlin or schwannomin. In individuals diagnosed with NF2, merlin is either absent or mutated to the point of inactivation. As such, merlin …

Real Time Rt-Pcr For Direct Detection Of Viable Mycobacterium Avium Subspecies Paratuberculosis In Chron's Disease Patients And Association Of Map Infection With Downregulation In Interferon-Gamma Receptor (Infg1) Gene In Crohn's Disease Patients, Mounir Chehtane

Electronic Theses and Dissertations

Association of Mycobacterium avium subspecies paratuberculosis (MAP) with Crohn's disease (CD) and not with ulcerative colitis (UC), two forms of inflammatory bowel disease (IBD), has been vigorously debated in recent years. This theory has been strengthened by recent culture of MAP from breast milk, intestinal tissue and Blood from patients with active Crohn's disease. Culture of MAP from clinical samples remained challenging due to the fastidious nature of MAP including its lack of cell wall in infected patients. The advent of real time PCR has proven to be significant in infectious disease diagnostics. In this study, real time reverse transcriptase …

Site Preparation Methods For Restoration Of Non-Native Pasturelands To Native Upland Habitat, Krisann Joie Kosel

Electronic Theses and Dissertations

The flatwoods ecosystem of Florida has been heavily depleted over time but remains one of the most important systems to many threatened and endangered species. Areas that have been converted into non-native pastures may be restored to provide not only this invaluable ecosystem but also restore connectivity of the surrounding ecosystems. The pasture areas on The Disney Wilderness Preserve in central Florida were surveyed, and a conceptual plan for restoration was written in 1996. That same year a pilot study was developed to assess five methods for removing non-native pasture grasses. The treatments studied were single herbicide, single disc, multiple …
