Bootsie – Estimation Of Coefficient Of Variation Of Aflp Data By Bootstrap Analysis, Justin Payne, Erica Lindroth, K. M. Kneeland, Steven R. Skoda, Fatima Mustafa, Muhammad Irfan Ullah, John E. Foster

Bootsie is an English-native replacement for ASG Coelho's “DBOOT” utility for estimating coefficient of variation of a population of AFLP marker data using bootstrapping. Bootsie improves on DBOOT by supporting batch processing, time-to-completion estimation, builtin graphs, and a suite of export tools for creating data files for other population genetics software.

The spined soldier bug, Podisus maculiventris (Say), is an important biological control agent of agricultural and forest pests; and it preys on eggs and larvae of lepidopteran and coleopteran species. Genetic variability among field samples collected from Michigan, Mississippi, Missouri, Florida and established laboratory colonies was examined using Amplified Fragment Length Polymorphism-Polymerase Chain Reaction (AFLP-PCR). Four AFLP primer pairs generated a total of 340 molecular markers for evaluation. Results from Analysis of Molecular Variance showed that the majority of the genetic variation occurred within populations (individuals from each sample site). The western corn rootworm, Diabrotica virgifera virgifera, is a damaging pest of cultivated corn that was controlled by applications of cyclodiene insecticides from the late 1940s until resistance evolved ~10 years later. Range expansion from the western plains into eastern USA coincides with resistance development. An alanine to serine amino acid substitution within the Rdl subunit of the gamma-aminobutyric acid (GABA) receptor confers resistance to cyclodiene insecticides in many species. The stable fly, Stomoxys calcitrans (L.), is a cosmopolitan pest of livestock and humans. The pestiferous nature and painful bite cause stress to cattle and other animals. The stress and resulting avoidance behaviors manifest as reductions in weight gain or milk production in cattle; estimated annual economic loss in the United States exceeds US$2 billion. Understanding the population genetics of stable flies could provide information on their population dynamics, origins of outbreaks, and geographical patterns of insecticide resistance, resulting in a tactical advantage for developing management strategies. The western bean cutworm, Striacosta albicosta (Smith), is a secondary pest of maize (Zea mays L.) and dry beans (Phaseolus vulgaris L.) in the western United States. Recently, this insect has undergone a major territory expansion into the eastern United States and has become a pest throughout much of the Corn Belt. This study was instigated to examine the population genetics of this pest to facilitate control and resistance management, as well as to shed light on the current habitat expansion. The western corn rootworm, Diabrotica virgifera virgifera LeConte (Coleoptera: Chrysomelidae), is a major pest of corn, Zea mays L., in North America that has recently invaded Europe. A loss of ovipositional fidelity to cornfields has allowed the species to circumvent crop rotation as a means of control in part of its range in the United States. Analyses of variation at eight microsatellite loci provided no evidence for general genetic differentiation between samples of western corn rootworm collected in soybean, Glycine max (L.) Merr., fields and those collected in cornfields both inside and outside the rotation-resistance problem area. This result suggests …

Genetic variation in stable fly, Stomoxys calcitrans (L.),populations from Nebraska, Canada, and Texas was sampled. Four of 12 allozyme loci were polymorphic, with an average of 1.7 alleles per locus. Observed and expected heterozygosities were 0.086 and 0.070,respectively. Nei's genetic distance between populations averaged 0.001 and ranged from 0.000 to 0.005. Wright's F statistics revealed greater variation within than among populations. Allele frequencies were homogeneous among temporal samples from a single population. Polymerasechain reaction—restriction fragment length polymorphism (PCR—RFLP) analysis of 6.4 kb of the mitochondrial DNA genome with 16 restriction enzymes revealed no variation in stable fly populations from Canada, …

Allozyme variation in 4 populations of secondary screwworm, Cochliomyia macellaria (F.), representing North, Central, and South America was examined . Variability was observed in 12 of 13 enzyme loci, and the frequency of the most common allele was IS statistics indicated random mating within populations . Wright's FST statistics and chi-square contingency analyses indicated homogeneous gene frequencies among the populations. These data suggest that C. macellaria is a panmictic species with high levels of gene flow between populations.

Allozyme variation in screwworm, Cochliomyia hominivorax (Coquerel), and secondary screwworm, C. macellaria (F.), populations from northwest Costa Rica was examined . Variability was observed in 11 of 13 enzyme loci and the frequency of the most common allele was <0.95 for 5 loci in screwworm . In secondary screwworm, 12 of 13 loci were variable and the frequency of the most common allele was <0.95 for 6 loci . Expected heterozygosities were 0.149 and 0.160 for screwworm and secondary screwworm, respectively. Goodness-of-fit statistics for Hardy-Weinberg equilibrium and Wrights F statistics indicated that both species are panmictic with no evidence of population substructuring. Nei ' s genetic distances were 0.000–0.001 for intraspecific comparisons and 0.899–0.916 for interspecific comparisons. The data indicate a high level of gene flow between populations within each species.
