Stroke is one of the leading causes of deaths as the risk factors, both controllable and uncontrollable, are many. We first concentrated on a stress-free way to deliver the drugs. Our preference was to have the animals ingest the drugs, which led us to a reliable method for orally administering medication to the animal models. Using three different drug combinations, we tested the effects on neurogenesis without stroke. We found the drug combination of Fluoxetine/Statin/Ascorbic acid increases neurogenesis 19 fold when compared with control. The carotid bodies (CB), the primary peripheral chemoreceptors, respond to changes in blood gases with neurotransmitter release, thereby increasing carotid sinus nerve firing frequency and ultimately correcting the pattern of breathing. It has previously been demonstrated that acute application of the adipokine leptin caused perturbations of intracellular calcium and membrane ion movement in isolated CB Type I cells (Pye et al, 2015) and augmented the response of the intact CB to hypoxia (Pye et al, 2016). Ischemic strokes account for 87% of all strokes and can have debilitating effects on language, sensory, and motor skills. Currently, tPA is the only medication approved by the FDA for the treatment of ischemic stroke, but the window of time to administer the drug is very small. In this thesis, we investigate the use of a simvastatin and fluoxetine drug combination (FS) as a possible treatment for ischemic stroke victims. In this study, we hypothesize that a pharmacological drug treatment comprised of Fluoxetine, Simvastatin and Ascorbic acid together with physical rehabilitation would reduce infarct sizes. Over the period of 60 days after stroke-induction, 13 of the 23 rats were administered the drugs beginning 20-26 hours after stroke-induction and the rest were assigned to the control group. Physical rehabilitation exercises were initiated from poststroke day 8 and continued for 23 alternate days. In the present study 10-12 month female rats were examined for functional recovery from stroke and this recovery was compared with the stem cell/progenitor cell proliferation in the brain (which was measured by Ki67). The cell proliferation indicated by Ki67 showed a 6 fold increase in control animals compared to the rehabilitation animals. The contralateral functional recovery in control animals were 46.6% and in the rehabilitation animals were 24.5%. The physical rehabilitation was carried out to determine if limb rehabilitation can promote greater functional recovery. Until now, observations regarding the central morphology and organization of Ia, group II and Ib Golgi tendon organ afferents have been confined to the cat model. As the use of rodents in the study of the development and organization of segmental spinal cord circuitry increases, a complete account of the peripheral encoding and central connectivity of rodent muscle proprioceptors is necessary. In Carbon Monoxide (CO) induced Delayed Neurological Sequelae (DNS) clinical signs develop 1 to 6 weeks after CO has cleared the body. The aim of this experiment was to develop a model of CO induced DNS which closely mimics "real world" conditions both in exposure and treatment. The model was challenged with hyperbaric or normobaric oxygen, or room air. Basic behaviors were measured by Open Field test on days 1, 7, 14 post exposure and treatment.
