Using forelimb asymmetry analysis along with Montoya Staircase methodology we tested how a fluoxetine simvastatin ascorbic acid drug cocktail would affect recovery from a demyelinating event in a rat model, with the hypothesis that if administered then it would help female recovery but hinder male recovery. It was found that a fluoxetine simvastatin and ascorbic acid drug cocktail did not significantly enhance recovery from a demyelination model injury in female rats, and that the same drug cocktail significantly slowed male rat recovery from the same type of injury. Whereas immune modulation has proven beneficial in multiple sclerosis (MS), we hypothesized that targeting down-stream modulators of neurogenesis and subsequent remyelination may offer an additional, if not superior, point of intervention in an attempt to repair damage and recover lost function. Stroke is one of the leading causes of deaths as the risk factors, both controllable and uncontrollable, are many. We first concentrated on a stress-free way to deliver the drugs. Our preference was to have the animals ingest the drugs, which led us to a reliable method for orally administering medication to the animal models. Using three different drug combinations, we tested the effects on neurogenesis without stroke. We found the drug combination of Fluoxetine/Statin/Ascorbic acid increases neurogenesis 19 fold when compared with control. In this current study, we have investigated this a combination of fluoxetine, simvastatin and ascorbic acid administered daily beginning at 20-26 hours after stroke induction. We hope to understand therapeutic abilities by studying its effectiveness on the blood brain barrier permeability and gene expression changes of the microglial subtypes involved in neuro-inflammation and neurogenesis factors in the peri-infarct region. Our results indicate that S-enantiomer of fluoxetine may be more beneficial compared to the R-enantiomer. Ischemic strokes account for 87% of all strokes and can have debilitating effects on language, sensory, and motor skills. Currently, tPA is the only medication approved by the FDA for the treatment of ischemic stroke, but the window of time to administer the drug is very small. In this thesis, we investigate the use of a simvastatin and fluoxetine drug combination (FS) as a possible treatment for ischemic stroke victims. In this study, we hypothesize that a pharmacological drug treatment comprised of Fluoxetine, Simvastatin and Ascorbic acid together with physical rehabilitation would reduce infarct sizes. Over the period of 60 days after stroke-induction, 13 of the 23 rats were administered the drugs beginning 20-26 hours after stroke-induction and the rest were assigned to the control group. Physical rehabilitation exercises were initiated from poststroke day 8 and continued for 23 alternate days. Stroke is one of the leading causes of death and the top cause of long term disability. Currently there exists no standardized treatment of care for ischemic stroke patients during the days following stroke other than giving aspirin. A drug combination containing fluoxetine, one of two statins, and ascorbic acid was used to try and improve motor function following strokes in rats. Endothelin induced stroke survival surgery was performed on the right cortex of Long Evans and Sprague Dawley rats. Our laboratory has found increased functional recovery post-stroke with Long Evans rats that received a combination of fluoxetine, simvastatin, and vitamin C. The objectives of this study were to test that treatment and other drug combinations on Sprague Dawley rats, to determine whether Forelimb Asymmetry is a reliable behavioral measure for stroke studies, and to determine the most optimal surgical procedure on Sprague Dawley rats. There were more significant increases in functional recovery observed on the Forelimb Asymmetry than on Montoya Staircase behavioral tests, but there were no significant differences between treatment groups and control.
