Cytokines play a pivotal role in regulating inflammation, which is a condition that makes the tissue vulnerable to different pathological and physiological conditions. Thus, how cytokines are regulated is an important area of study. Skin that receives ultraviolet B radiation (UVB), a major pro-oxidative stressor, results in the release of multiple cytokines and chemokines like tumor necrosis factor (TNF)-alpha and interleukin (IL)-8. Previous studies from our group and others have demonstrated synergistic release of TNF-alpha when UVB is combined with IL-1 or the lipid mediator Platelet-activating factor (PAF). Of interest, subcellular microvesicle particles (MVP) have been proposed to play an …

Kinetics Of Microvesicle Particle Release In Keratinocytes, Pariksha Thapa Jan 2019

Kinetics Of Microvesicle Particle Release In Keratinocytes, Pariksha Thapa

Microvesicle particles (MVPs) are subcellular particles that could be involved in inter-cellular communication because they carry various bioactive substances including cytokines. Previous studies from our lab has shown that that the lipid mediator Platelet-activating Factor (1-alkyl-2-acetyl-glycerophosphocholine; PAF) and ultraviolet B radiation (UVB) enhances the release of MVP in various cell types like primary keratinocytes, epithelial cell lines, and murine skin. We hypothesized that there may be synergistic increases in MVP release after combination of treatment of keratinocytes with UVB and PAF agonist (CPAF)/phorbol ester (PMA). The combination treatment significantly increases MVP and cytokine release at 4 to 8 hours time …

Extracellular Microvesicles As A Novel Biomarker For Wound Healing, Walid Omran Mari Jan 2017

Extracellular Microvesicles As A Novel Biomarker For Wound Healing, Walid Omran Mari

Browse all Theses and Dissertations

Introduction: Wound healing is a sophisticated dynamic process that involves complex coordination among a variety of resident cells in a suitable extracellular environment. Chronic wounds are defined as wounds that fail to heal within a period of 3 months. Negative pressure wound therapy (NPWT) is a closed suction drainage system is used to enable mass transport of fluid from the body as an adjunct to surgical procedures. (Oasis-ultra) is a triple layer extracellular matrix containing different types of glycosaminoglycans (GAGs) like heparin sulfate, hyaluronic acid and chondroitin sulfate. In addition, it has adhesion molecule such as fibronectin and laminin along …

Effect Of Oasis-Ultra Matrix On The Healing Rate Of Stage Iv Pressure Wounds, Abdelfatah Shaban Abou Issa Jan 2016

Effect Of Oasis-Ultra Matrix On The Healing Rate Of Stage Iv Pressure Wounds, Abdelfatah Shaban Abou Issa

Browse all Theses and Dissertations

Introduction: (Oasis-ultra) is an extra cellular collagen rich matrix derived from porcine intestinal sub-mucosa. A prospective, multi-centered, randomized, single-blinded clinical trial was conducted to study the effects of Oasis-ultra combined with negative pressure wound therapy (NPWT) on the healing rate of stage IV pressure wounds versus NPWT alone. Materials and Methods: Twelve subjects were involved in the study: six patients in the study group and six in the control group. NPWT was changed twice a week for all subjects, and Oasis-ultra was applied weekly. The wounds were measured weekly, and the healing rate was calculated for each subject for 12 …

Developing Proteomic And Cytokine Biomarkers For Vulvodynia, Ashvin Iyer Jan 2015

Developing Proteomic And Cytokine Biomarkers For Vulvodynia, Ashvin Iyer

Browse all Theses and Dissertations

Vulvodynia is a chronic, heterogeneous, and multifactorial disease. This condition may affect up to 18 percent of the female population including Caucasians, African Americans, Africans and Hispanics particularly those sexually active at child bearing age. The etiology of this condition is complex and multifactorial and it is frequently accompanied by physical disabilities, psychological distress and sexual dysfunction. Clinically, vulvodynia can be generalized or localized and pain can be provoked or unprovoked. Patients may also describe vulvar paresthesias or dysesthesias that may last hours. The International Society for the Study of Vulvar Disease (ISSVD) recognizes vulvar pain related to a specific …

Expression Of Inflamatory Response Genes In Ferrets Challenged With H5n1 Avian Influenza Virus, Brock M. Miniard Jan 2012

Expression Of Inflamatory Response Genes In Ferrets Challenged With H5n1 Avian Influenza Virus, Brock M. Miniard

Browse all Theses and Dissertations

Influenza A H5N1 has emerged as a potential to be the next pandemic influenza. Ferrets are a promising model of H5N1 infection because the disease progression is similar to that known in humans, however reagents to characterize infections in ferrets are few. We developed real-time PCR assays for the ferret cytokines IL-1β, IL-2, IL-4, IL-8, IL-12p40, TNF-α, IFN-α, IFN-β, IFN-γ, TGFB-β1, and TGFB-β2. Forty-eight ferrets were vaccinated with either one of three vaccine formulations (Vaccine 1, Vaccine 2, or Vaccine 3), or saline (0.9% NaCl), then challenged intranasally with a lethal dose of H5N1 strain A/Vietnam/1203/04. Cytokine assays were then …

The Influence Of Stored Product Mites On The Function Of Skin Microvascular Endothelial Cells, Jennifer Graff Curp Jan 2010

The Influence Of Stored Product Mites On The Function Of Skin Microvascular Endothelial Cells, Jennifer Graff Curp

Browse all Theses and Dissertations

The prevalence of allergenic diseases and sensitization to stored product mites is increasing worldwide. Stored product mites are prevalent in homes, foods, and some work environments. Stored product mites need to be further studied so we can characterize their inflammatory properties to understand their role in skin disease, and provide proper treatment to patients with mite allergies. The purpose of the study was to show the role of stored product mites in modulating the inflammatory and immune response to human dermal endothelial cells in the skin. The epidermis serves as the first line of defense between the body and the …

Immunomodulation Of Human Skin Cells By Extracts Of The Scabies Mite, Sarcoptes Scabiei, Jeremi Stevan Mullins Jan 2008

Immunomodulation Of Human Skin Cells By Extracts Of The Scabies Mite, Sarcoptes Scabiei, Jeremi Stevan Mullins

Browse all Theses and Dissertations

Sarcoptes scabiei infests its host through burrowing into the stratum corneum layer of skin. Keratinocytes and fibroblasts responding to the burrowing mite(s) and/or mite products produce inflammatory and immune activating cytokines and chemokines. The purpose of this study was to determine if an extract of S. scabiei can modulate cytokine and chemokine expression from keratinocytes and fibroblasts in vitro. Normal human keratinocytes and fibroblasts were exposed to an extract of S. scabiei var. canis with or without various pro-inflammatory stimuli. Cytokine and chemokine expression were measured by enzyme-linked immunosorbent assay (ELISA). Scabies extract increased expression of interleukin-6 (IL-6), growth-related oncogene-α …
