We have identified the irreversible protein modification S-(2-succino)cysteine, a product of the Kreb’s cycle intermediate fumarate reacting with the thiol group of cysteine (Cys) residues, also termed succination. Succination is increased in adipose tissue of diabetic mice in vivo and in adipocytes matured in high glucose medium (25 mM) in vitro. Selenocysteine (Sec) is a less common but highly conserved amino acid that has major functions in redox chemistry. Sec possesses a lower pKa (~5.4) and is much more reactive in comparison to Cys. Plants respond to these attacks by using chemical compounds including monoterpenes that are constitutively and inductively produced in some plants, and stored inside glands on their leaves. The black-footed ferret (Mustela nigripes), a North American mustelid species, was once found abundantly throughout the Midwest until extreme decline in prairie dogs, the black-footed ferrets primary food source, brought the species to near-extinction. This resulted in the development of the Black-Footed Ferret Recovery Program which included bringing the entire species into captivity with the intention of breeding the species back to sufficient numbers to allow for successful reintroduction. The most well studied cancer-related gene in history, p53, is responsible for protecting the integrity of the genome. A variety of different stresses are detected by p53 and its regulators, allowing for numerous post-translational modifications that activate p53. After activation p53 can then regulate transcription of a myriad downstream targets, resulting in either cell cycle arrest or apoptosis. One of the stress signals that activates p53 is viral infection in part through the induction of type-1 interferon. Systematic implementation of sea turtle conservation measures have occurred in the U.S. since the 1970’s. As such, this dissertation assessed the probability that loggerhead sea turtles (Caretta caretta) in the NW Atlantic will meet or exceed recovery criteria in the minimum timeframe (i.e., 50 years) specified by the Recovery Plan. Mathematical modeling (Chapter 1) of a theoretical population resembling an important nesting assemblage in a stochastic environment for 200 years revealed broad (<20k to >106k) fluctuation in annual nest counts without extinction, as well as strong contemporary environmental influence on annual nest counts. Background. Diabetes is a widespread public health concern that alters the metabolism of adipocytes through high glucose stress and hormonal dysregulation. Under diabetic conditions there are excess nutrients available and subsequently an accumulation of tricarboxylic acid cycle intermediates such as fumarate. Fumarate can irreversibly react with protein cysteine residues to form S-2-succinocysteine (2SC, protein succination). Succinated proteins can be turned over by autophagy, a mechanism of removing damaged proteins and organelles that have accumulated due to metabolic stress.

Because these positions are a potential consequence of consistent individual behavioral differences, or personality, a better understanding of potential mechanisms concerning personality is central to predicting an individuals’ location. Saltmarshes are among the most productive ecosystems globally. At North Inlet estuary, South Carolina, about one third of the primary production comes from benthic microalgae. During the tidal cycle, mobile microalgae vertically migrate through the upper 3 mm of sediment. At low tide, algae are vulnerable to a variety of grazers, including the mud snail, Ilyanassa obsoleta, which is abundant in tidal creeks. This is a floral inventory of the Gills creek watershed. It was conducted from fall 2014 through spring 2016. The final product is a list of plants that are known to presently or historically exist in the watershed. The list is comprised of 250 native species and 36 introduced and invasive species. The results indicate that Gills creek watershed’s plant diversity is severely threatened by introduced and invasive species, but still retains sizable native diversity.

Pooled next-generation sequencing allows multiple genomes to be sequenced at once in a single sample, with the resultant single nucleotide polymorphism data giving reliable estimates of allele frequencies and population genetic parameters in a cost-effective manner. This approach has potentiated new opportunities for understanding the evolution of virus populations within individual hosts over the course of infection, where the sequencing of individual genomes is exceedingly difficult and impractical. The management of saltmarshes requires detailed knowledge of the underlying processes driving their distribution in both time and space to make appropriate management decisions. With most of the world’s population living in the coastal zone and rising sea levels, one of our most important natural resources in the coastal zone faces increasing threat of collapse. This study uses the current state of Light Detection and Ranging (LiDAR) technology to model and predict saltmarsh distribution at a landscape-scale and provide evidence that a terrestrial laser scanner (TLS) can be used to estimate saltmarsh biomass for inclusion into existing models.
