During the first phase of the Biomass Burn Operational Project (BBOP) field campaign, conducted in the Pacific Northwest, the DOE G-1 aircraft was used to follow the time evolution of wildfire smoke from near the point of emission to locations 2-3.5 h downwind. In nine flights we made repeated transects of wildfire plumes at varying downwind distances and could thereby follow the plume's time evolution. On average there was little change in dilution-normalized aerosol mass concentration as a function of downwind distance. This consistency hides a dynamic system in which primary aerosol particles are evaporating and secondary ones condensing. Organic …

Bartonella bacilliformis, the etiological agent of Carrión's disease, is a Gram-negative, facultative intracellular alphaproteobacterium. Carrión's disease is an emerging but neglected tropical illness endemic to Peru, Colombia, and Ecuador. B. bacilliformis is spread between humans through the bite of female phlebotomine sand flies. As a result, the pathogen encounters significant and repeated environmental shifts during its life cycle, including changes in pH and temperature. In most bacteria, small non-coding RNAs (sRNAs) serve as effectors that may post-transcriptionally regulate the stress response to such changes. Encapsulated microbes have been used for decades to produce commodities ranging from methyl ketone to beer. Encapsulated cells undergo limited replication, which enables them to more efficiently convert substrate to product than planktonic cells and which contributes to their stress resistance. To determine how encapsulated yeast supports long-term, repeated fed-batch ethanologenic fermentation, and whether different matrices influence that process, fermentation and indicators of matrix durability and cell viability were monitored in high-dextrose, fed-batch culture over 7 weeks. At most timepoints, ethanol yield (g/g) in encapsulated cultures exceeded that in planktonic cultures. And frequently, ethanol yield differed among the four matrices …

Evaluating our understanding of smoke from wild and prescribed fires can benefit from downwind measurements that include inert tracers to test production and transport and reactive species to test chemical mechanisms. We characterized smoke from fires in coniferous forest fuels for >1,000 hr over two summers (2017 and 2018) at our Missoula, Montana, surface station and found a narrow range for key properties. ΔPM2.5/ΔCO was 0.1070 ± 0.0278 (g/g) or about half the age-independent ratios obtained at free troposphere elevations (0.2348 ± 0.0326). Reactive nitrogen (Nr, defined as all nitrogencontaining compounds except for N2 and N2O) is one of the most important classes of compounds emitted from wildfire, as Nr impacts both atmospheric oxidation processes and particle formation chemistry. In addition, several Nr compounds can contribute to health impacts from wildfires. Understanding the impacts of wildfire on the atmosphere requires a thorough description of Nr emissions. Bartonella are Gram-negative bacterial pathogens that trigger pathological angiogenesis during infection of humans. Bartonella bacilliformis (Bb) is a neglected tropical agent endemic to South America, where it causes Carrión's disease. Little is known about Bb's virulence determinants or how the pathogen elicits hyperproliferation of the vasculature, culminating in Peruvian warts (verruga peruana) of the skin. In this study, we determined that active infection of human umbilical vein endothelial cells (HUVECs) by live Bb induced host cell secretion of epidermal growth factor (EGF) using ELISA. The Kathmandu Valley in Nepal is a bowl-shaped urban basin that experiences severe air pollution that poses health risks to its 3.5 million inhabitants. As part of the Nepal Ambient Monitoring and Source Testing Experiment (NAMaSTE), ambient air quality in the Kathmandu Valley was investigated from 11 to 24 April 2015, during the premonsoon season. Ric-8A is a cytosolic Guanine Nucleotide exchange Factor (GEF) that activates heterotrimeric G protein alpha subunits (Gα) and serves as an essential Gα chaperone. Mechanisms by which Ric-8A catalyzes these activities, which are stimulated by Casein Kinase II phosphorylation, are unknown. We report the structure of the nanobody-stabilized complex of nucleotide-free Gα bound to phosphorylated Ric-8A at near atomic resolution by cryo-electron microscopy and X-ray crystallography. The mechanism of Ric-8A GEF activity differs considerably from that employed by G protein-coupled receptors at the plasma membrane. A barrier to cost-efficient biomanufacturing is the instability of engineered genetic elements, such as plasmids. Instability can also manifest at the whole-genome level, when fungal dikaryons revert to parental species due to nuclear segregation during cell division. Here, we show that by encapsulating Saccharomyces cerevisiae-Pichia stipitis dikaryons in an alginate matrix, we can limit cell division and preserve their expanded metabolic capabilities. To better understand the effects of wildfires on air quality and climate, it is important to assess the occurrence of chromophoric compounds in smoke and characterize their optical properties. This study explores the molecular composition of light-absorbing organic aerosol, or brown carbon (BrC), sampled at the Missoula Fire Sciences laboratory as a part of the FIREX Fall 2016 lab intensive. Bartonella henselae is a facultative intracellular pathogen that occurs worldwide and is responsible primarily for cat-scratch disease in young people and bacillary angiomatosis in immunocompromised patients. The principal source of genome-level diversity that contributes to B. henselae's host-adaptive features is thought to be horizontal gene transfer events. However, our analyses did not reveal the acquisition of horizontally-transferred islands in B. henselae after its divergence from other Bartonella. Rather, diversity in gene content and genome size was apparently acquired through two alternative mechanisms, including deletion and, more predominantly, duplication of genes. Moose (Alces alces) in Isle Royale National Park impact the ecosystem by browsing tree species and serving as the primary food source for wolves. As a heat-sensitive species, moose are susceptible to the impacts of climate change in the southern extent of their range which includes Isle Royale. Understanding how temperature impacts moose behavior is valuable and can be used to predict how moose may respond to changing temperature in the future. Dragons have been depicted in human art as early as 4500 BCE. For centuries, these fantasy creatures have inspired countless folk and fantasy tales, as well as appearing in the art of different cultures around the world. Now there are thousands of different depictions of these huge, flying, fire-breathing lizards, but are any of them possible? In this study, I reference peer-reviewed scientific articles, phylogenetic analysis, and paleoart studies to create biologically-sound dragons. Top-down predator-prey effects that alter the abundance, biomass, or productivity of a population community across more than one link in a food web are referred to as trophic cascades. While these effects have been extensively studied in aquatic environments, fewer studies have examined trophic cascades in terrestrial ecosystems. And fewer still terrestrial studies have tested for trophic cascades between vertebrates and grassland vegetation. Across the globe, grassland plant biomass is driven by both precipitation and non-linear positive feedbacks between grazing and plant productivity, as predicted by the Intermediate Grazing Hypothesis. Ground dwelling birds must scale all kinds of complex terrain in order to survive in their natural environments. For instance, Alectoris chukar live on steep hillsides with slopes of up to 60° or 172.3% slope. We undertook the present study to improve understanding of how birds successfully traverse such complex terrain. Using a high speed camera, we analyzed the hind limb kinematics of chukars during normal locomotion on a 10° and 35° incline, decline, and level slope. Human cytomegalovirus (HCMV) is widely spread throughout the world and immunocompromised individuals can suffer severe diseases from HCMV infection. Once the infection is established, HCMV can spread through the body and infect many major somatic cell types. The glycoproteins H and L (gH/gL) on HCMV envelope can be bound by either gO or the UL128-131 proteins to form complexes gH/gL/gO and gH/gL/UL128-131 that are critical for viral entry and spread, and these two complexes are important targets of neutralizing antibodies. Interdisciplinary and transdisciplinary approaches are needed to effectively address the challenges facing our complex social-ecological systems. To meet this need, many approaches, including co-production, have been proposed to overcome the difficult relationship between science and governance, often termed a ‘gap’, which can continue to impede natural resource problem solving. Co-production is an iterative process that engages scientists, managers, and community members in knowledge creation and problem solving. Migration by ungulates has traditionally been thought of as a strategy that increases access to forage quality or reduces exposure to risk of predation, but the benefits of migration may be waning globally. In partially migratory populations, the persistence of both migrant and resident strategies is an intriguing ecological phenomenon, because migrants and residents often face contrasting fitness consequences. Partial migration is common in mule deer (Odocoileus hemionus), a species that has experienced widespread declines across the western United States during recent decades. Remote sensing can be utilized by land management organizations to save money and time. Mapping vegetation using either aerial photographs or satellite imagery and the applications for forest management are of particular interest to the Montana Department of Natural Resources. In 2018, the organization began a pilot program to test the incorporation of raster analysis of remotely sensed data into their inventory program and had limited success. This analysis identified two areas of improvement: the selection method of inventory plots and the imagery used for classification and metrics. Across much of North America, moose populations (Alces alces) are declining due to disease, predation, climate, and anthropogenic pressures. Despite this, populations of moose in Colorado have continued to grow. Studying successful (i.e., persistent or growing) populations of moose can facilitate the continued conservation of the species by identifying habitat features critical for moose persistence.

First, I evaluated calving success of moose in Colorado and the impact of willow habitat quality and nutrition. I then estimated the probability of female moose having a calf using repeated observations in a Bayesian occupancy model. I assigned values for dry matter …

Introduction: Wildland firefighters (WLFF) undergo a critical training (CT) period immediately before the firefighting season. The intensity of CT exercise could lead to muscle damage, as previously reported cases of rhabdomyolysis in WLFFs have been documented. This study established the effects of activities performed during a two-week CT period on acute markers of muscle damage in WLFFs.

Introduction: Wildland firefighters (WLFFs) are confronted with numerous physical and mental stressors. Pre-fire season includes an intense 2-week critical training (CT) period; a preparatory phase of multiple activities that can result in injury, illness, and rhabdomyolysis. The purpose of this study was to identify physiologic changes in metabolic, inflammatory, and oxidative stress biomarkers during 2 weeks of CT in WLFFs.

In 2012, the U.S. Forest Service (USFS) promulgated new forest planning regulations that significantly altered national forest management. One of the most controversial and important advancements was the inclusion of what were meant to be stronger biodiversity protections. An analysis of USFS’s rationale in revising the biodiversity regulations provides insights into how to interpret the substantively and procedurally new ecosystem and species protections. Around the turn of the 20th century, lake trout (Salvelinus namaycush) were widely introduced in several lakes and reservoirs outside their native range in western North America. Since then, lake trout have become problematic in many lakes where they were introduced, causing significant declines in popular sport fishes and native species, most notably federally protected bull trout (Salvelinus confluentus). The Northern Rocky Mountain fisher population (Pekania pennanti), is classified as a Species of Greatest Conservation Need and is of special concern to state, federal, and tribal entities. In this thesis, we present methods to effectively survey fishers across the northern Rockies of Idaho and Montana, and provide estimates of fisher distribution at a population range-wide scale through occupancy analyses. We also assess factors that influence fisher occurrence through covariate analyses and identify core fisher habitat in the northern Rockies through spatial occupancy modeling. Introduction: Wildfire suppression is characterized by high total energy expenditures and water turnover rates. Hydration position stands outline hourly fluid intake rates. However, dose interval remains ambiguous. We aimed to determine the effects of micro- and bolus-dosing water (MW, BW) or carbohydrate-electrolyte (MCE, BCE) solutions on fluid balance, heat stress, and carbohydrate oxidation during extended thermal exercise. Methods: Males (n=12), in a repeated measures cross-over design, completed 4, 120-minute treadmill trials (1.3 m.s-1, 5% grade, 33°C, 30%RH) wearing a USFS uniform and 15kg pack.
