Fisetin, a flavanol with anti-inflammatory, anti-cancer, and anti-aging properties, has shown promise for reducing fat accumulation in tissue culture and animal models. This plant sourced compound has limited studies supporting its effects on fat accumulation. Therefore, this study was completed to determine fisetin’s role in fat reduction along with its mechanism of action using Caenorhabditis elegans. C. elegans is a small roundworm with roughly 65% of its genes being conserved in humans related to disease. Coffee consumption is associated with reduced risk of metabolic syndrome, obesity and diabetes. Although the mechanisms of action are not yet elucidated, the effects of coffee and its bioactive components on lipid metabolism may account for the overall coffee effects on human health. Therefore, this study investigated the molecular mechanisms of coffee and its bioactive components on lipid metabolism using Caenorhabditis elegans as a model system. Research suggests that the trajectory to obesity and its associated metabolic disorders begins early in life. Prenatal exposure to maternal gestational diabetes mellitus (GDM) is associated with increased risk, while breastfeeding is associated with reduced risk. Breastfeeding may influence obesity risk by preventing rapid postnatal weight gain. Mothers with gestational diabetes may not breastfeed as intensely as healthy mothers, potentially exacerbating the risk to their infants. We conducted three distinct investigations related to gestational diabetes, breastfeeding, and infant weight gain. All used data from the U.S. Infant Feeding Practices Study II. Prediabetes is defined by elevated blood glucose concentrations not high enough to meet criteria for type 2 diabetes. Exercise or metformin, a common “anti-diabetes” medication, may attenuate the progression from prediabetes to type 2 diabetes by improving insulin sensitivity and cardio-metabolic health. Because each treatment has its primary action in different tissues, combining exercise (muscle) with metformin (liver) may further enhance insulin sensitivity and cardio-metabolic health.
