Creating memories is a fundamental challenge for the human brain. To create memories, defining features of experiences must be stored distinguishably without forgetting other memories. Memory associations represent co-occurring features and defining features across experiences. Memory associations are represented as networks of information that are stored in the brain. New memory associations are encoded during experiences and can be used to update existing memory associations during offline intervals. However, the mechanisms that underlie how encoded memory associations are stored within existing networks during offline intervals remains unclear. Learning and memory formation at the cellular level involves decoding complex electrochemical signals between nerve cells, or neurons. Understanding these processes at the molecular level requires a comprehensive study of calcium-sensitive proteins that serve as signal mediators within cells. More specifically, the protein calcium/calmodulin-dependent protein kinase II (CaMKII) is a key regulator of downstream cellular signaling events in the brain, playing an important role in long term memory formation. CaMKII is encoded in humans on four different genes: alpha, beta, gamma and delta. CD4+ and CD8+ T cells are lymphocytes of the adaptive immune system that play essential roles in immunity. Both T cell subsets recognize their cognate antigen through the T cell receptor (TCR), which induces the proliferation and differentiation of these antigen-specific cells into effector T cells. CD4+ T cells have the potential to differentiate into one of multiple lineages of helper T (Th) cells and participate indirectly in antigen clearance by orchestrating the function of other cells. CD8 T cells are the cytotoxic effectors of the adaptive immune response, clearing virally infected and cancerous cells within the host. CD8 T cells acquire their cytotoxic function by differentiating into cytotoxic T lymphocytes (CTLs). Once the CTLs have cleared the antigen, the majority of responding cells will die during contraction; however, a small population of the antigen-specific responders will remain, differentiating into long-lived memory cells that provide potent protection to the host upon re-encounter with the antigen. A mild traumatic brain injury (mTBI), also known as a concussion, is defined as an injury that results in an alteration of consciousness or mental status. Previous studies have shown mTBI populations experience a number of chronic (> 1 year) symptoms, such as sleep disturbances (e.g., sleep stage alterations), mood alterations (e.g., depressive symptoms), and cognitive alterations (e.g., poor concentration). The three chapters of this dissertation sought to explore these long-term sequelae and the possible interrelations between them. Nocturnal sleep has been shown to benefit memory in adults and children. During the preschool age range (~3-5 years), the distribution of sleep across the 24-hour period changes dramatically. Children transition from biphasic sleep patterns (a nap in addition to overnight sleep) to a monophasic sleep pattern (only overnight sleep). In addition, early childhood is a time of neuronal plasticity and pronounced acquisition of new information. Healthy aging is associated with a reduction in slow-wave sleep (SWS), crucial for declarative memory consolidation in young adults; consequently, previously observed benefits of sleep on declarative learning in older adults could reflect a passive role of sleep in protecting memories from waking interference, rather than an active, stabilizing effect. To dissociate the passive and active roles of sleep, a visuo-spatial task was administered; memory was probed after a 12 hr interval consisting of either daytime wake or overnight sleep and post-wake/post-sleep stability of the memories was tested following task-related interference.
