Two Essays On Whole Farm Modeling And Crop Marketing In Western Kentucky, Benjamin A. Martin

This thesis is composed of two essays that investigate whole farm planning and crop marketing in western Kentucky. In the first essay, contracting decisions between food corn producers and a mill are analyzed to observe factors affecting the bushel amount farmers contract. Unbalanced panel data containing seven years’ worth of pricing and contract information are used with a fixed-effects model to generate parameter estimates and quantify their effect on bushels contracted. It was found that contract attributes, market condition, and relationship-specific assets had a significant effect on producers’ food corn contracting decisions. The second essay utilizes mixed-integer programming to optimize …

Although China is generally known as a tea-consuming country, both the production and consumption of coffee in China has seen double-digit growth in the last decade. This upward trend is projected to continue as long as investment is sustained and levels of disposable income continue to increase. In this rapidly expanding market, it is important for coffee producers and retailers in China to understand the preferences of emerging Chinese coffee consumers. Characterization And Evaluation Of Androgen-Binding Protein, Sex Hormone-Binding Globulin, And Thyroxine-Binding Globulin In The Horse, Blaire O'Neil Fleming

The objectives of this study are to characterize two carrier proteins in the horse that significantly decrease in humans following anabolic androgenic steroid administration: sex hormone-binding globulin (SHBG) and thyroxine-binding globulin (TBG). For SHBG characterization, qPCR, RNA sequencing, and immunohistochemistry were performed on testes and equine livers. Free and total testosterone immunoassays were utilized to confirm the presence of a carrier protein in equine circulation. SHBG was detected in the testes using qPCR, RNA sequencing, and IHC, indicating the presence of the isoform androgen-binding protein (ABP). SHBG was not detected in any liver samples. Exposure to environmental pollutants poses numerous risk factors for human health, including increasing incidence of cardiovascular disease and diabetes. Persistent organic pollutants, such as polychlorinated biphenyls (PCBs) have been strongly linked to the development of these chronic inflammatory diseases and the primary route of exposure is through consumption of contaminated food products. Thus, the gastrointestinal tract is susceptible to the greatest levels of these pollutants and is an important facet to study.

Arterial blood pressure (BP) undergoes a 24-hour oscillation that peaks in the active day and reaches a nadir at night during sleep in humans. Reduced nocturnal BP fall (also known as non-dipper) is the most common disruption of BP circadian rhythm and is associated with increased risk of untoward cardiovascular events and target organ injury. Up to 75% of diabetic patients are non-dippers. However, the mechanisms underlying diabetes associated non-dipping BP are largely unknown. The purpose of this mixed methods study was to learn how cultural sensitivity was integrated and perceived in community nutrition education of an existing program offered by the University of Kentucky Cooperative Extension Service Nutrition Education Program. Data collection consisted of a quantitative cultural competence assessment survey tool used with nutrition education paraprofessionals (n=74), qualitative in-depth interviews of nutrition education paraprofessionals (n=8) and focus groups of Hispanic/Latino nutrition education program participants (n=39). Mastitis and milk quality affect every dairy farmer across the globe. Sand bedded freestalls are the industry standard for cow comfort, welfare, and the control of environmental mastitis. Compost bedded packs may be a viable alternative to the sand bedded freestall.

Compost bedded packs are maintained at a consistent level of moisture, nutrients, and aeration to favor compost microorganisms. Greater bacteria counts in bedding have traditionally been associated with increased mastitis rates and mastitis pathogens can be found in the pack and on the teats of cattle housed in even well managed compost bedded pack barns. RNA interference (RNAi) is a useful reverse genetics tool for investigation of gene function as well as for practical applications in many fields including medicine and agriculture. Due to the variability in RNAi efficiency, RNAi-based methods are currently being developed for controlling only coleopteran insects which are known to be amenable to RNAi. Emerald ash borer (EAB), Agrilus planipennis (Coleoptera: Buprestidae), is an aggressive invader from Asia that has killed millions of trees in North America. Recently EAB has been documented developing in a novel host, white fringetree, Chionanthus virginicus. I evaluated larval performance in two common ash species and white fringetree by infesting excised bolts with emerald ash borer eggs. Vitamin D Works Through The Lipid Droplet Protein Plin2 To Augment Mitochondrial Function In Skeletal Muscle, David M. Schnell

Vitamin D has been connected with increased intramyocellular lipid (IMCL) mitochondrial function in skeletal muscle. It is also shown to prevent lipotoxicity in several tissues, but this has not yet been examined in skeletal muscle. Perilipin 2 (PLIN2), a lipid droplet protein upregulated with vitamin D treatment, is integral to managing IMCL capacity and lipid oxidation in skeletal muscle. Increased lipid storage and oxidation is associated with increased tolerance to a hyperlipidic environment and resistance to lipotoxicity. Therefore, I hypothesized that vitamin D increases β-oxidation and lipid turnover though a PLIN2 mediated mechanism, thereby preventing lipotoxicity.

Small scale protected zones are valuable for helping the health and productivity of fisheries at Lake Tanganyika (East Africa). Spatial placement of protected areas relies on accurate maps of benthic habitats, consisting of detailed bathymetry data and information on lake-floor substrates. This information is unknown for most of Lake Tanganyika. Fish diversity is known to correlate with rocky substrates in ≤ 30 m water depth, which provide spawning grounds for littoral and pelagic species. These benthic habitats form important targets for protected areas, if they can be precisely located.

Estrogens seem to play a role in the locomotor activating effects of cocaine. Japanese quail provide a good model for hormonal manipulation as alterations of their photoperiod controls hormone levels. The current study aims to examine the role of early life photoperiod manipulation in cocaine-induced behavioral sensitization in quail. It was expected that if quail were raised on a short photoperiod, they would have a reduction in gonadal hormones and this reduction in hormones would affect the acquisition of cocaine-induced behavioral sensitization. Adult skeletal muscle is highly plastic and responds readily to environmental stimuli. One of the most commonly utilized methods to study skeletal muscle adaptations is immunofluorescence microscopy. By analyzing images of adult muscle cells, also known as myofibers, one can quantify changes in skeletal muscle structure and function (e.g. hypertrophy and fiber type). Skeletal muscle samples are typically cut in transverse or cross sections, and antibodies against sarcolemmal or basal lamina proteins are used to label the myofiber boundaries.

Background: To determine the role of muscle mass in sex-dependent differences in power output during flywheel resistance training (FRT).

Methods: Twenty recreationally active (≥ 2 resistance exercise bouts per week), subjects (10 M, 10 F) completed 2 bouts of resistance exercise using a flywheel resistance training (FRT) device (Exxentric kbox 4 Pro) separated by at least one week. Each session consisted of 3 sets of 4 exercises (squat, bent-over row, Romanian deadlift, and biceps curl) with varying moments of inertia (0.050, 0.075, and 0.100 kg/m2, respectively) in random order. It is estimated that nearly 50,000 individuals in the United States will be diagnosed with head and neck cancer in 2017 (American Cancer Society www.cancer.org). Ninety percent of oral cancers are head and neck squamous cell carcinoma (HNSCC). Major obstacles in the treatment of HNSCC are recurrence and treatment resistance, which contributes to increased mortality. Pathogenic fungi are increasingly associated with epidemics in wildlife populations and represent a significant threat to global biodiversity. Snake fungal disease is an emerging disease caused by the fungus, Ophidiomyces ophiodiicola, and appears to be widespread in the eastern United States. Yet an evaluation of field diagnostics, and an understanding of the population-level consequences of the disease, are lacking. Changes in land use such as mountaintop removal mining with valley fills (MTR/VF) affect chemical, physical, and hydrological properties of headwater streams. Although numerous stream taxa have experienced significant declines from MTR/VF, stream salamanders appear to be particularly sensitive. Yet, the specific mechanism(s) responsible for the population declines has eluded researchers. We sampled salamander assemblages across a continuous specific conductivity (SC) gradient in southeastern Kentucky and estimated occupancy rates and abundance estimates along this gradient. Land Use Land Cover (LULC) changes can take place at the expense of degrading environmental conditions and undermining ecosystem’s capacity to deliver benefits to people. In the Appalachian region, surface mining for coal is a major driver of LULC change. The Surface Mining Control and Reclamation Act (SMCRA) of 1977 requires mine site reclamation but typical reclamation practices often result in land cover dominated by grass and shrubs. Consumption of endophyte-infected (E+) grasses has long been associated with health problems in animals. In cattle E+ tall fescue consumption leads to fescue toxicosis, and in horses it leads reproductive problems. The health-related issues associated with endophyte consumption have been attributed to the effects caused by the ergot alkaloids produced by the fungus. These ergot alkaloids are considered D2-like receptor agonists, and 5-HT2 serotonin and α-adrenergic receptor partial agonists. Prenatal alcohol exposure (Fetal Alcohol Syndrome [FAS] and Fetal Alcohol Spectrum Disorders [FASD’s]) represents the leading preventable cause of intellectual disabilities in the western world, with FASDs estimated to affect approximately 2-5% of live births in the United States at an approximate annual cost of $3.6 billion (CDC, 2015; May et al., 2009). Precision dairy monitoring is used to supplement or replace human observation of dairy cattle. This study examined the value dairy producers placed on disease alerts generated from a precision dairy monitoring technology. A secondary objective was calculating the accuracy of technology-generated disease alerts compared against observed disease events. A final objective was determining the economic viability of investing in a precision dairy monitoring technology for detecting estrus and diseases.

Purpose: The purpose of this study was to determine if a novel physical activity intervention game (Bingocize®) designed for use in confined spaces and modified to include age appropriate activities would increase the time spent engaged in physical activity (PA) during indoor recess. Methods: Fifty-two third grade children wore triaxial accelerometers during three different recess conditions. The recess conditions included: “typical” indoor recess (TIR), indoor recess with children engaged in Bingocize® (IRB), and “typical” outdoor recess (TOR). Results: There were significant (p < 0.05) differences among the recess conditions for the time spent in sedentary, light, moderate-to-vigorous physical activity (MVPA) intensity categories, as well as PA counts, and steps. During IRB, TIR, and TOR the subject were sedentary 42.4% (±0.10), 71.9%(±0.10), and 17.5% (±0.10) of the recess time, respectively. During IRB, TIR and TOR the subjects spent 43.4%(±0.10), 18.5%(±0.10), and 74.2%(±0.12) engaged in MVPA, respectively. Conclusions: These results indicate that Bingocize® promotes increased PA during times when inclement weather necessitates indoor recess and larger space requirements for activity are not available.

Epilepsy is a neurological disorder that is characterized by aberrant electrical activity in the brain resulting in at least two unprovoked seizures over a period longer than 24 hours. Approximately 60% of individuals with epilepsy are diagnosed with temporal lobe epilepsy (TLE) and about one third of those individuals do not respond well to anti-seizure medications. This places those individuals at high risk for sudden unexpected death in epilepsy (SUDEP). Age-related neurologic disease is a significant and growing burden on our society. Although the largest share of research effort has typically been devoted to the common neurodegenerative illnesses (such as Alzheimer’s disease, or AD), the reality is that nearly all cases of neurodegenerative disease possess elements of mixed pathology. Vascular contributions to cognitive impairment and dementia (VCID) is a complex form of dementia, combining aspects of vascular disease and other forms of dementia, such as Alzheimer’s disease. Modern maize hybrids exhibit higher yields, increased biomass production, stress tolerance and greater nitrogen (N) use efficiency. Increased biomass accumulation can influence nutrient uptake and lead to increased nutrient removal. Hybrids were tested at seeding rates (SR) of 74000 (low) and 148000 (supraoptimal) plants ha-1 and at N rates of 0 (deficient) and 390 (non-limiting) kg N ha-1. Plants were sampled at V7, V14, R3, R5 and R6 and separated into vegetative and reproductive fractions for determination of dry matter and N accumulation. Grain yield was harvested at R6.
