The goal of this project is to be able to streamline a protocol for conducting ERGs on zebrafish from mice ERG protocol already present in Dr. Renna’s lab. The objective is to create a protocol specifically for zebrafish and to eliminate any issues that occur when transiting from a mice ERG protocol to a zebrafish ERG protocol and to obtain a light response from zebrafish retinae in differing light intensities. In May of 2023, I embarked on an exciting research journey to Moorea, French Polynesia, alongside fellow students and faculty members from the University of Akron and Syracuse University. This expedition was part of the university-sponsored Tropical Vertebrate Biology course, where we delved into the exploration of various tropical species inhabiting the island, including sea urchins, geckos, and my primary focus, the blackspotted rockskipper.

Studies have shown that HERs are involved in the progression of human cancers. Researchers have found that HER2 mutations in colorectal cancer are similar to mutations found in breast cancer. Recombinant antibodies are effective therapeutic treatments for cancers. Antibodies can also be used to determine how proteins function under normal and pathophysiological conditions while also identifying interacting partners. INTRODUCTION: Ventilatory threshold (VT) is where minute ventilation (VE) increases nonlinearly with increasing exercise intensity. Prior studies have shown that subjects can recognize VT by changes in breathing during exercise. Teaching recreational athletes to use VT as a training method could result in better quality training. PURPOSE: Determine if recreational athletes can accurately perceive changes in ventilation associated with VT during maximal exercise testing. Even though poor glycemic control puts diabetics at risk for a variety of disease complications, there are many barriers to achieving adequate blood glucose regulation. Diabetic cardiac autonomic neuropathy (CAN) is a progressive autonomic denervation of the cardiovascular system resulting from chronic hyperglycemia and characterized by orthostatic hypertension, sinus tachycardia, low heart rate variability, and fatal arrhythmias. Available CAN therapeutics are limited to symptom alleviation, and do not prevent or reverse cardiac nerve damage. The diffusion in and out of these vesicles will be tracked by using singular lipid molecules. These singular lipid molecules will be tagged with fluorescence and viewed using an internal reflection fluorescence microscope. The rise in e-cigarette use is likely due to the overwhelming evidence regarding the addictive properties of nicotine and the damage caused by traditional tobacco cigarettes. I investigated the effects of diet and sex on zebrafish bone metabolism. Zebrafish were subjected to either a high-calorie or low-calorie diet over a five-week period. After this diet, zebrafish scales were used to measure alkaline phosphatase (ALP) and tartrate-resistant acid phosphatase (TRAP) enzyme activity with a fluorescent substrate. The enzyme activities were markers of the osteoblasts (ALP) and the osteoclasts (TRAP). These data were compared among sex and diet of the zebrafish using a repeated measures ANOVA statistical test. I also measured the number of resorption lacunae per scale (another indicator of osteoclast activity). The results showed higher TRAP …

It is important that native plants are able to successfully emerge in wetlands because they play an important role in maintaining diversity. The relationship between seed buoyancy and seedling emergence was analyzed for three wetland species of the same genus: Elymus canadensis (FACU), Elymus macgregorii (FACW), and Elymus virginicus (FACW). E. macgregorii seemed to be the most buoyant, and E. virginicus and E. canadensis demonstrated very similar, less buoyant levels. Based on the idea that more buoyant seeds will be deposited at a shallow depth, and less buoyant seeds will be deposited at a deeper soil level, a relationship between …

Damp granular media is a difficult environment to study because it is both practically complex and it lacks equations which fully describe its behavior. In this study, an oscillatory lateral head movement and its effects while penetrating damp granular media were tested using a robophysical model. This experimental research was inspired by the burrowing behavior of the clade Amphisbaenia, a group of usually limbless squamates that employ a variety of different burrowing behaviors, but it can apply to a wide range of burrowers. This research could help with both human burrowing technologies and the further investigation of animal behaviors.

I made endospanin 1 knockout (KO) zebrafish to examine its effects on lipid and bone metabolism. Endospanin 1, or leptin receptor overlapping transcript (leprot), is a cytosolic protein linked to the protein hormone leptin that influences the trafficking of leptin receptors to the plasma membrane of cells. Genes for endospanin and tyrosinase (a pigmentation enzyme) were targeted via a microinjection of guide RNAs and CRISPR Cas9 into zebrafish embryos at 2-4 cell stages. I was able to disrupt the endospanin 1 gene (based upon the disruption of tyrosinase), but very few mutant zebrafish fully developed into adults. Only low KO …

Particulate calcium carbonate (PCC) is an important filler in industrial products, including paints, which are obtained through mining and sintering (heating to 1,100°C), which is responsible for up to 2% of global emissions. A potential solution to reduce these emissions is the use of calcium carbonate produced by bacteria from atmospheric , through a process called microbially induced carbonate precipitation (MICP). The pollutants measured in this study include hardness, bromine, residual chlorine, iron, copper, lead, nitrate, nitrite, ammonium chloride, total chlorine, fluoride, carbonate, pH, total alkalinity and cyanuric acid. This study aims to examine the relationship between e-cigarettes (both with nicotine and without) and the effects on the standard metabolic rates of crayfish. In this experiment, 42 crayfish were divided into three treatment groups and exposed to a designated solution of either a control solution or a solution containing e-cigarette (nicotine or non-nicotine) vapor. Their standard metabolic rates were then measured by using oxygen uptake as a proxy. The results of this experiment indicate that there is no statistically significant relationship between the use of electronic cigarettes and the standard metabolic rate of crayfish. Pollinator visitation rates are a helpful way to monitor the health of ecosystems; however, there lacks a standardized method for obtaining these rates. The pollinator visitation rates for five plant species were collected and the relationship between the standard error of these rates and the time interval was determined. Monarda, Echinacea purpurea, Pycnanthemum muticum, and Baptisia alba all exhibit more accurate pollinator visitation rates as the time interval increased. Trifolium repens exhibited less accurate pollinator visitation rates as the time interval increased.

According to Wolff’s law, bone is constantly subject to change based on loading patterns (Ruff et al., 2006). Those subjected to greater stress become more robust by increasing bone mineral density and often changing geometric properties. Previous studies of primate anatomy have found significant differences in bone robustness between the fore- and hindlimbs in adult leaping primates. Because leaping primates, such as Galago senegalensis and Galago moholi, subject their hindlimbs to much greater stress, this difference in robustness may come as a result of bone functional adaptation/Wolff’s law. Not only are there academic demands to remain eligible for competition, but there are physical and mental demands that often go unnoticed. With all these demands, it is understandable how one's nutrition, body image, and mental health may go on the back burner, resulting in insufficiencies in all three categories. The treatment groups were control, nicotine, and non-nicotine. The mass, sex, and condition of their appendages were recorded before random assignment. For the purpose of this study only male crayfish were used as there were not enough female crayfish. This experiment will aim to explore the heterodimerization of Dopamine (D2) and Adenosine (A2A) receptors in cells when bound to different ligands. Borroto-Escuela et al. (2018) investigated the dimerization of the A2AR-D2R in rats when administered with cocaine, and they found that disrupting the A2AR-D2R complex restores the cocaine reward system in rats. This was a strong influence as to why we chose to investigate A2AR-D2R dimerization in Chinese hamster ovary (CHO) cells.

Ohio forests are threatened by the invasive ecosystem engineer A. agrestis. A. agrestis invasion typically co-occurs with the ecosystem engineer, Odocoileus virginianus, where their impacts may synergize. To determine the direct effects of A. agrestis invasion, fenced plots across the Cuyahoga Valley National Park that excluded deer were utilized. The species richness, Shannon diversity and evenness of woody understory flora was measured in each plot. Mustard extraction was used to determine earthworm abundance. This study investigated the viability of bioremediating rubber cryogrind using enriched indigenous bacteria. To begin the experiment, soils from three highway roadside locations in Kansas, KS 96 and West, KS 400 and 143rd, and 199th, were collected and transported to the lab to be studied. An initial soil characterization was run on the soil samples using distilled (DI) water mixture and 0.01 M CaCl2 to assess conductivity. Previous work involving 3D culture of human liver cells with fluorinated chitosan based microparticles has shown that their incorporation provides needed structural cues that the culture of cells alone does not provide, such as increased gas transport. However, using bare microparticles to grow large 3D cellular structures is not practical as they tend to collapse before any meaningful research can be done on them. The oral behavior of drinking includes interactions between the tongue, jaw, and liquid being ingested to provide hydration to the animal. Different mechanisms may be used to ingest liquid such as lapping, licking, or sucking. The objective of the study was to compare the mechanism of lapping across marsupial species through comparison of the length of gape cycle, and the amplitude of jaw pitch and tongue protraction between the two marsupial species, Didelphis virginiana and Monodelphis domestica.
