Characterization And Tissue-Specific As Well As Heat-Stress Expression Analysis Of Cbl-Interacting Protein Kinase Genes In Dimocarpus Longan Lour, Wei Zheng, Xueming Dong, Xuefei Yu, Qiuying Zhang

Turkish Journal of Botany

The CBL-interacting protein kinase (CIPK) gene family plays important roles in plant growth, development, and responses to abiotic stresses. To date, the CIPKs have been extensively identified and characterized in various plants. However, there have been few studies on CIPKs in Dimocarpus longan Lour (D. longan). In this study we identified eight putative CIPKs in D. longan (DlCIPKs) according to RNA-seq data. The physicochemical properties, subcellular localization, conserved motifs, phylogenetic relationships, glycosylation and phosphorylation sites, and gene ontology annotation were analyzed using bioinformatics tools. The tissue-specific and heat-stress-responsive expressions were systematically investigated as well. Ten motifs widely maintained by DlCIPKs …

Regulation Of Boron Toxicity Responses Via Glutathione-Dependent Detoxification Pathways At Biochemical And Molecular Levels In Arabidopsis Thaliana, Doğa Seli̇n Kayihan, Ceyhun Kayihan, Yelda Özden Çi̇ftçi̇ Jan 2019

Regulation Of Boron Toxicity Responses Via Glutathione-Dependent Detoxification Pathways At Biochemical And Molecular Levels In Arabidopsis Thaliana, Doğa Seli̇n Kayihan, Ceyhun Kayihan, Yelda Özden Çi̇ftçi̇

Turkish Journal of Botany

The fine-tuned regulation of the Halliwell-Asada cycle (ascorbate-glutathione pathway) in Arabidopsis thaliana under boron (B) toxicity was shown in our previous report. In this study, we investigated the expression levels of some members of the glutathione S-transferase (GST) superfamily, such as phi (GSTF2, GSTF6, GSTF7, and GSTF8), tau (GSTU19), and zeta (GSTZ1) classes in Arabidopsis thaliana that were exposed to 1 mM boric acid (1B) and 3 mM boric acid (3B). Additionally, the expression levels of genes for glutathione (GSH) and phytochelatin biosynthesis as well as miR169 and miR156 were evaluated in Arabidopsis thaliana exposed to 1B and 3B. Moreover, …

Transcriptome Analysis Reveals Differentially Expressed Genes Between Human Primary Bone Marrow Mesenchymal Stem Cells And Human Primary Dermal Fibroblasts, Eki̇m Z. Taşkiran, Beren Karaosmanoğlu Jan 2019

Transcriptome Analysis Reveals Differentially Expressed Genes Between Human Primary Bone Marrow Mesenchymal Stem Cells And Human Primary Dermal Fibroblasts, Eki̇m Z. Taşkiran, Beren Karaosmanoğlu

Turkish Journal of Biology

Stromal cells have been widely used in biomedical research and disease modeling studies in vitro. The most commonly used types of stromal cells are mesenchymal stem cells and fibroblasts. Their cellular phenotypes and differentiation capabilities are quite similar and there are no specific distinction criteria. In order to identify transcriptomic differences between these 2 cell types, we performed next-generation RNA sequencing. Using the global gene expression profile and pathway analysis, we showed that human primary bone marrow mesenchymal stem cells and human primary dermal fibroblasts have different molecular signatures. We also identified critical transcription factors that are differentially expressed between …

Variations In Ethylene Sensitivity Among Mungbean [Vigna Radiata (L.) Wilczek] Genotypes Exposed To Drought And Waterlogging Stresses, Susheel Kumar Raina, Nikhil Raskar, Lalitkumar Aher, Ajay Kumar Singh, Dhammaprakash Pandhari Wankhede, Jagadish Rane, Paramjit Singh Minhas Jan 2019

Variations In Ethylene Sensitivity Among Mungbean [Vigna Radiata (L.) Wilczek] Genotypes Exposed To Drought And Waterlogging Stresses, Susheel Kumar Raina, Nikhil Raskar, Lalitkumar Aher, Ajay Kumar Singh, Dhammaprakash Pandhari Wankhede, Jagadish Rane, Paramjit Singh Minhas

Turkish Journal of Botany

Ethylene is an important phytohormone that regulates several aspects of plant development including those crucial for drought tolerance. The main aim of the present work was to investigate variability in ethylene sensitivity of mungbeans to seed germination, waterlogging, and leaf senescence, and the underlying transcriptional regulation of ethylene biosynthetic/responsive genes. Significant variation was observed for silver-nitrate-mediated germination inhibition and flooding stress tolerance among the mungbean genotypes. Genotypes with differential ethylene sensitivity (IC-325817 and IC-325756) revealed variable leaf senescence rates upon exposure to exogenously supplied ethephon. Drought induced significant upregulation of VrACS7 and VrERF genes in the more sensitive genotype, implicating …

Genome-Wide Identification And Molecular Characterization Of Citrus Unshiu Wrkytranscription Factors In Satsuma Mandarin: Clues For Putative Involvement In Cell Growth,Fruit Ripening, And Stress Response, Mariem Ayadi, Rim Mzid, Rayda Ben Ayed, Najla Kharrat, Henda Merchaoui, Ahmed Rebai, Mohsen Hanana Jan 2019

Genome-Wide Identification And Molecular Characterization Of Citrus Unshiu Wrkytranscription Factors In Satsuma Mandarin: Clues For Putative Involvement In Cell Growth,Fruit Ripening, And Stress Response, Mariem Ayadi, Rim Mzid, Rayda Ben Ayed, Najla Kharrat, Henda Merchaoui, Ahmed Rebai, Mohsen Hanana

Turkish Journal of Agriculture and Forestry

WRKY transcription factors, being involved in several biological roles, stimulus signaling, and stress response, could be interesting candidates to prospect, particularly in the Citrus unshiu (Satsuma mandarin) genome. Our approach, mainly based on computational analyses, led to the identification of 51 CiuWRKYs followed by their molecular characterization. Syntenic relationship analysis showed significant bias towards sweet orange and Arabidopsis chromosomes. Analyses of promoter region showed that CiuWRKY1 modulates gene transcription through W-box site targeting. The deduced CiuWRKY1 protein was moderately hydrophobic. A comparative in silico analysis among Arabidopsis thaliana and Citrus sinensis WRKYs allowed us to designate orthologous genes. Lacking available …

Acute And Chronic Effects Of Thifluzamide On Daphnia Magna, Feyza İçoğlu Aksakal Jan 2019

Acute And Chronic Effects Of Thifluzamide On Daphnia Magna, Feyza İçoğlu Aksakal

Turkish Journal of Zoology

Thifluzamide is a thiazole carboxanilide fungicide used to control a wide range of basidiomycete diseases on maize, potatoes, rice, and other crops. Although this fungicide is found in aquatic environments, there is no information about its acute and chronic toxicity on aquatic invertebrates. In the present study, to investigate acute and chronic toxicity of thifluzamide, daphnids were exposed to 1.25, 1.5, 2, and 2.5 mg/L thifluzamide for 48 h and 21 days, respectively. The expression of four oxidative stress-related genes (CAT, GCLC, GST, and GPX) and life-history parameters such as days to the first brood, days to the first egg …
