In Vitro Mutagenesis Of Etlingera Elatior (Jack) And Early Detection Of Mutation Using Rapd Markers, Muhamad Fahmi Yunus, Maheran Abd Aziz, Mihdzar Abdul Kadir, Siti Khalijah Daud, Azmi Abdul Rashid

Turkish Journal of Biology

Mutation breeding techniques in combination with tissue culture and molecular marker methods provide a powerful tool for improvement of vegetatively propagated plants. The aim of this study was to develop a protocol for shoot regeneration and mutation induction of Etlingera elatior. The results of irradiation on in vitro buds of E. elatior showed LD_{50} to be 10 Gy, with the survival of explants being sharply reduced at this dosage. All 8 selected gamma irradiated regenerants were differentiated from the untreated control based on the banding patterns obtained using 9 primers, which generated 59 reproducible bands, whereby 35 (55.31%) were found …

Development Of Short Gssrs In G. Arboreum And Their Utilization In Phylogenetic Studies, Tayyaba Shaheen, Yusuf Zafar, James M. Stewart, Mehboob-Ur Rahman

Turkish Journal of Agriculture and Forestry

Microsatellite regions in DNA fragments of Gossypium arboreum were explored and PCR products 500-800 bp long were analyzed for genomic simple sequence repeats (gSSRs). From 39 segments, 23 short gSSRs were identified; on average they occurred every 1.6 kb. Primers were used to amplify fragments from 18 diploid species representing 6 diploid genomes and 1 tetraploid species, G. hirsutum (AD). The phylogeny of diploid genomes was subsequently determined. No primer amplified the DNA from all genotypes; however, amplification occurred in 59% of 437 PCR events. All genomes were grouped into 2 clusters (a and b). The obtained phylogeny of the …

Assessment Of Genetic Diversity In Naturally Growing 29 Trifolium L. Taxa From Bolu Province Using Rapd And Ssr Markers, Emel Uslu, Gözde Damla Ertuğrul, Mehmet Teki̇n Babaç Jan 2013

Assessment Of Genetic Diversity In Naturally Growing 29 Trifolium L. Taxa From Bolu Province Using Rapd And Ssr Markers, Emel Uslu, Gözde Damla Ertuğrul, Mehmet Teki̇n Babaç

Turkish Journal of Biology

Two molecular marker technologies, RAPD and SSR, were used to determine the genetic diversity of 29 taxa of Trifolium L. collected from Bolu Province. The analysis was carried out using 11 RAPD and 5 SSR markers. A total of 446 and 331 fragments were produced by RAPD and SSR markers, respectively. The amplification products were 300-3500 bp with RAPD and 125-3000 bp with SSR markers. These bands were scored as absence and presence for all taxa. Data obtained were used to estimate genetic similarity using Jaccard’s coefficient, and dendrograms were constructed by the UPGMA method using either RAPD and SSR …

Genetic Fidelity Assessment Of Encapsulated In Vitro Tissues Of Bacopa Monnieri After 6 Months Of Storage By Using Issr And Rapd Markers, Joe Virgin Largia Muthiah, Karutha Pandian Shunmugiah, Ramesh Manikandan Jan 2013

Genetic Fidelity Assessment Of Encapsulated In Vitro Tissues Of Bacopa Monnieri After 6 Months Of Storage By Using Issr And Rapd Markers, Joe Virgin Largia Muthiah, Karutha Pandian Shunmugiah, Ramesh Manikandan

Turkish Journal of Botany

The present study has investigated the efficiency of regrowth of Bacopa monnieri (L.) Pennell after 6 months of storage along with its clonal fidelity checking. Shoot tips and nodal segments of in vitro grown plantlets were encapsulated and stored at different temperatures and the highest regeneration was achieved by storing shoot tips at 4 °C for a period of 6 months. The response of shoot tips was found to be much better when compared to nodal segments in the case of forming healthy shoots and roots. The genetic stability of regrown plantlets was analysed by using RAPD and ISSR markers. …
