Model organisms subject to dietary restriction (DR) generally live longer. Accompanying this lifespan extension are improvements in overall health, based on multiple metrics. This indicates that pharmacological treatments that mimic the effects of DR could improve health in humans. To find new chemical structures that extend lifespan, we screened 30 000 synthetic, diverse drug-like chemicals in Caenorhabditis elegans and identified several structurally related compounds that acted through DR mechanisms. The most potent of these NP1 impinges upon a food perception pathway by promoting glutamate signaling in the pharynx. This results in the overriding of a GPCR pathway involved in the …

Mucopolysaccharidosis IIIA (MPS IIIA) is a lysosomal storage disease caused by a mutation in the gene that codes for the enzyme heparan sulfamidase. The decreased enzyme activity of heparan sulfamidase results in the accumulation of heparan sulfate (HS). HS accumulation in the brain causes severe central nervous system (CNS) complications, including learning and memory deficits and seizure. MPS IIIA patients have short life expectancies and there currently is no cure for the disease. As with any complex biological pathway, the splicing process has both advantages and obstacles with respect to the diversity and fidelity of protein production. The potential benefits of being able to produce multiple versions of a gene (isoforms) must be weighed against the additional complexity introduced by the noisy and mechanistically complicated process of splicing. Indeed, research has found that errors in splicing can be implicated in an increasing number of disorders. Integrity Of The Barrier Epithelium In Drosophila Gut Is Mediated By Factors That Regulate Intestinal Stem Cell Activity, Adam A. Kinion

My thesis work on the Drosophila intestinal epithelium (IE) involved characterizing the mitochondrial unfolded protein response (UPRmt), validating proliferation specific regulatory genes, and proposing gut barrier function–specific genes as regulators of commensal bacteria density. The results from this project have shown that intestinal stem cells (ISCs) respond systemically in the presence of autonomously induced, visceral muscle (VM)-localized UPRmt. A phenotypic correlation between autonomous electron transport chain (ETC) dysfunction was observed in the visceral muscle and the systemic UPRmt response of ISCs in the posterior midgut. Horseshoe crabs are xiphosuran chelicerates, the sister groupto arachnids. As such, they are important for understandingthemost recent common ancestor of Euchelicerata and the evolution and diversification of Arthropoda. Limulus polyphemus is the most investigated of the four extant species of horseshoe crabs, and the structure and function of its visual system have long been a major focus of studies critical for understanding the evolution of visual systems in arthropods. Likewise, studies of genes encoding Limulus opsins, the protein component of the visual pigments, are critical for understanding opsin evolution and diversification among chelicerates, where knowledge of opsins is limited, and …

Plasmodium falciparum, a protozoan parasite known as malaria, widely impacts human health; thus antimalarial drug investigations are critical. Doxycycline is a commonly used antimalarial prophylactic, but its mechanism of action is unclear. In prokaryotes, doxycycline works as an antibacterial by disrupting protein translation via the small subunit ribosome. Interestingly, P. falciparum has a small subunit ribosome of prokaryotic origins in the apicoplast, a plastid-like organelle. Therefore, we hypothesized that doxycycline works in P. falciparum by inhibiting protein synthesis via the small subunit ribosomal RNA and that mutations in the gene encoding the P. falciparum apicoplast small subunit ribosomal RNA …

One might not realize that blockbuster movies can be used as an educational tool in the classroom setting to enhance the learning process. While these types of movies are not crafted as pedagogical tools, like animated, demonstrative video clips, there is educational value to be found in them. Inherent Variability Of Cancer-Specific Aneuploidy Generates Metastases., Mathew Bloomfield, Peter Duesberg

BACKGROUND: The genetic basis of metastasis is still unclear because metastases carry individual karyotypes and phenotypes, rather than consistent mutations, and are rare compared to conventional mutation. There is however correlative evidence that metastasis depends on cancer-specific aneuploidy, and that metastases are karyotypically related to parental cancers. Accordingly we propose that metastasis is a speciation event. This theory holds that cancer-specific aneuploidy varies the clonal karyotypes of cancers automatically by unbalancing thousands of genes, and that rare variants form new autonomous subspecies with metastatic or other non-parental phenotypes like drug-resistance - similar to conventional subspeciation.
