Spider beetles are a poorly known group of North American beetles in the family Ptinidae. Protection of biodiversity requires knowledge of species- and population-level distinctiveness, so data regarding genetic structure of spider beetles can shed light on conservation priorities. Spider beetles in the genus, Coleotestudus, are distributed broadly in the southwestern US. Given their small size and lack of flight, it is presumed that the dispersal capability of Coleotestudus is limited, leading to strong population structure on a small spatial scale. The results of this project will explore a novel approach to lipid metabolism in pregnant women by exploring the relationship between maternal gene expression of genes critical for fat metabolism, protein expression, and maternal and neonatal outcomes. The study will also examine gene expression in lean pregnant women compared to obese pregnant women, shedding light into the adverse pathology of obesity during pregnancy. These findings may uncover a mechanism related to gene-expression that connects unfavorable maternal fat metabolism to inflammation and/or insulin resistance, which may help to uncover future targets for interventions.

A radula is an anatomical structure found exclusively in mollusks that acts as both tongue and teeth in feeding. Its structure has been studied for more than a century, yet published methods for identifying, dissecting, and cleaning radulae for microscopy are almost unintelligible to anyone not already familiar with molluscan anatomy. The purpose of this project was to identify the best method of garden slug radula isolation and present it in a manner that anyone could understand and successfully use.

In most ecosystems, primary production is conducted by plants or photosynthetic microbes; however, extremophilic sulfur-oxidizing bacteria can use hydrogen sulfide gas (H2S) as an electron donor to produce metabolic energy in a type of metabolism called chemoautotrophy. These ecosystems are a prominent feature of some karst landscapes, and two have been observed in the Mammoth Cave system, Sulphur River in Parker Cave and Marianne’s Pass in Mammoth Cave National Park respectively.

Telomeres are repetitive DNA sequence motifs found at eukaryote chromosome ends. Telomeres help protect chromosome ends from DNA damage and promote chromosome stability. Chromosomes play important roles in aging, mutation, and cancer. Eukaryotic pathogens also use telomeres to mutate and manage virulence genes. In response to chromosome end breakage newly formed telomeres, called de novo telomeres, are formed to recreate the lost telomere and sub-telomeric regions.

Amphibian populations are threatened by increasing levels of environmental pollution resulting from human activities. Areas with mixed land use that include agriculture and urban development experience increased contamination from surface herbicide and road deicer application. Upon entering water sources, these chemicals have the capacity to disrupt the natural dynamics of aquatic ecosystems. This study aimed to evaluate the effects of these contaminants on amphibian populations. I used the spotted salamander (Ambystoma maculatum) as a model, rearing larvae from hatching to metamorphosis. When sleep is interrupted, so that an individual is awakened for some period before going back into sleep, the sleep can be described as fragmented. Zooplankton are small aquatic animals that serve an important role in transferring energy from phytoplankton to higher trophic levels. The zooplankton community composition in Barren River Lake from 2008 and 2020 was compared. A field study was conducted to not only describe the seasonal population dynamics of zooplankton in Barren River Lake, but also to determine if an invasive zooplankton, Daphnia lumholtzi, was present. While D. lumholtzi was found throughout Barren River Lake in 2008, it was absent from both resampled sites in 2019 and 2020. Visual and haptic (tactile) modes of perception, while occasionally exercised independently, most often occur concurrently. The degree to which the ordering of the two modes of perception affects successful recognition of three-dimensional shapes varies. Some have found that the cross-modal orders (vision followed by haptics or vice versa) produce equal performance (Caviness, 1964; Lacey, Peters, & Sathian, 2007; Norman et al., 2006), while other researchers found visual-haptic (VH) performance to be superior to haptic-visual (HV) performance (Davidson, Abbott, & Gershenfeld, 1974; Norman, Clayton, Norman, & Crabtree, 2008). Microbacteriophage Hasitha is a virus that infects Microbacterium foliorum, a bacterium associated with grasses that was first discovered in Germany. Hasitha was isolated from an enriched compost sample and is of particular interest due to its unusual growth pattern. Most bacteriophages require actively growing host cells to produce new phage progeny. However, Hasitha can infect and kill stationary (non-replicating) bacterial cells. We discovered this unusual characteristic through a fortuitous observation of infected lawns that were allowed to incubate in the lab workspace for approximately one month. From 1980 to 2020, drought events accounted for 11.4% of the billion-dollar disasters in the United States (U.S.) yet caused the second highest total amount in damages at $236.6 billion. Bees are threatened by environmental changes, pathogens, and pesticides (Pettis, 2012, Meeus et al., 2018). The goal of the study is to compare and evaluate the bee abundance and species richness in three different field types. My hypothesis is that bees would favor the fields planted for their benefit, and that the tall grass dominated plantings would be preferred over mowed fescue. My findings have low power considering few replicates and the use of relative abundance and relative species richness for statistical analyses. The overuse of antibiotics has led to an increase in antibiotic resistant bacteria since some antibiotics are sent into the wastewater system by waste produced by both humans and agriculture, making exposure to these bacteria more likely. The evolution of these bacteria in groundwater is of particular concern, as groundwater is used as a source of drinking water, and infections by these bacteria would be more difficult to treat. Sleep is an important process required for vertebrates, including humans, to function. When sleep is disrupted, it leads to deleterious effects such as inflammatory responses throughout the body. Past studies have shown that acute (24 h) sleep fragmentation (SF) leads to an inflammatory response in white adipose tissue. However, whether brown adipose tissue responds in a similar fashion is unknown. State Wildlife Action Plans (SWAPs) were created in order to identify Species of Greatest Conservation Need (SGCN) and develop plans to prevent their populations from further decline and the possibility of being listed as threatened or endangered. Which SGCNs are included are decided based on characteristics that determine their vulnerability. As of early 2021, approximately 720stoneflies (Insecta, Plecoptera) species are found in the U.S., but only 143 are listed as SGCN. Mudskippers are intertidal fishes that can survive both in and out of the water. They are territorial and exhibit behaviors to attract mates and defend against competitors. Recently, it has been shown that vibrations are produced and transmitted through the mud during such displays in one species, Periophthalmodon septemradiatus. To see if similar vibrations are produced in other species of mudskippers, I recorded the behavioral interactions of pairs of the mudskipper Periophthalmus barbarus via digital video and acoustic signals via accelerometer. There are extremely important uses for platinum when it comes to the treatment of cancers and dementias. One potential therapeutic combines platinum with a structure known as phenanthroline. This platinum phenanthroline structure is normally extremely water-insoluble, rendering it difficult to study and use in biological systems. The Williams’ Biochemistry Laboratory endeavored to find a simpler synthetic pathway for a water-soluble phenanthroline product. Obstructive sleep apnea is identified by recurring events of airway collapse during sleep, intermittent hypoxia, and perturbations in sleep continuity, known as sleep fragmentation. There is evidence to suggest that elderly patients are more at risk of developing obstructive sleep apnea. Antibiotic resistance is a rapidly spreading global threat to human health. Previously in our lab, we developed a simple method for facile detection of antibiotic resistance genes (ARGs) in bacteria. This DNA detection system takes advantage of the CRISPR-Cas system (Clustered Regularly Interspaced Short Palindromic Repeat and CRISPRAssociated Protein). We designed CRISPR in complex with nuclease deactivated Cas9 (dCas9) on graphene oxide (GO) sheets conjugated with Cas9 antibodies. Single guide RNA (sgRNA) was customized to program dCas9 for target DNA binding. sgRNA was covalently labeled with fluorescein to provide fluorescence signal. All bacteria contain a multi-subunit RNA polymerase (RNAPs) that is essential for gene expression. Because of the centrality of these enzymes in cellular life, the structure and function of the various subunits is intensely studied. The primary sequence of the RNAP β’ subunit contains five cysteine residues that are highly conserved. Food waste is a pervasive global issue with many environmental and social repercussions. While about one-third of all food produced for human consumption goes to waste, many people in the United States and the world are affected by food insecurity. Food recovery, the process of rescuing edible food that would otherwise go to waste and delivering to hungry people, is an effective solution for both food waste and food insecurity.
