Depletion Of The Rna Helicase Skiv2l2 Impedes Mitotic Progression And Histone Mrna Turnover In Murine Cell Lines, Alexis Marie Onderak

RNA surveillance via the nuclear exosome requires cofactors such as the helicase SKIV2L2 to process and degrade certain noncoding RNAs. This dissertation aimed to characterize the phenotype associated with RNAi knockdown of SKIV2L2 in two murine cancer cell lines: Neuro2A and P19. Skiv2l2 knockdown in Neuro2A and P19 cells induced changes in gene expression indicative of cell differentiation and reduced cellular proliferation. Analysis of the cell cycle revealed defective progression through mitosis following SKIV2L2 depletion. These results indicate that SKIV2L2 enhances mitotic progression, thereby maintaining cancer cell proliferation and preventing differentiation. Mitochondrial respiratory complexes are critical components of cellular energy production that require tight regulation to ensure optimal function. Rcf1 and Rcf2 are mitochondrial proteins that can physically associate with the yeast respiratory complexes III and IV, and the higher-ordered III-IV respiratory supercomplex that also contains the ADP/ATP translocase, AAC. Rcf1 can physically associate with both complex III and IV independently, and can be chemically crosslinked to AAC, indicating a close physical proximity to a predominant regulator of energy flux within the cell. This dissertation describes two research projects on anaerobic digestion (AD) that investigated the relationship between microbial community structure and digester function. Both archaeal and bacterial communities were characterized using high-throughput (Illumina) sequencing technology with universal 16S rRNA gene primers. In the first project, bioaugmentation using a methanogenic, aerotolerant propionate enrichment culture was investigated as a possible method to increase digester methane production. Nine anaerobic digesters, seeded with different biomass, were operated identically and their quasi steady state function was compared. Explaining the mechanism behind a genetic disease involves two phases, collecting and analyzing data associated to the disease, then interpreting those data in the context of biological systems. The objective of this dissertation was to develop a method of integrating complementary datasets surrounding any single biological process, with the goal of presenting the response to a signal in terms of a set of downstream biological effects. Copper oxide nanoparticles (CuO NPs) are an up and coming technology increasingly being used in industrial and consumer applications and thus may pose risk to humans and the environment. In the present study, the toxic effects of CuO NPs were studied with two model organisms Saccharomyces cerevisiae and Caenorhabditis elegans. The role of released Cu ions during dissolution of CuO NPs in growth media were studied with freshly suspended, aged NPs, and the released Cu2+ fraction. Reinforcement learning (RL) has been widely used as a model of animal and human learning and decision-making. The neural networks underlying RL involve many of the same structures primarily affected by Alzheimer’s disease (AD) such as the hippocampus. Yet, RL and non-invasive evaluation of its neural underpinnings have been underutilized as a framework for understanding disease pathology and its pre-clinical states. The aim of this thesis is to better understand the regulation of the cystine/glutamate antiporter (system xc-) and its role in regulating neuronal survival and death. Expressed primarily on astrocytes, system xc- takes up cystine and releases glutamate in a 1:1 ratio. Cystine uptake is the rate-limiting step in glutathione synthesis, the brain’s main antioxidant. Glutamate released into the extrasynaptic space can regulate neuronal function; however excessive glutamate release can cause excitotoxicity. Antimicrobial chemicals in consumer personal care products have been found to increase antibiotic resistance in pure culture studies. Although many studies focus on antibiotic resistance development pertinent to medical scenarios, resistance developed in natural and engineered environments might be significant and has become an emerging concern for human health. This dissertation focuses on the antimicrobial chemicals triclosan and triclocarban. These compounds are distinctly different from antibiotics and are used in products like soaps that are labelled as “antibacterial”.
