In the past, the indigenous people of the Salish Sea relied on a variety of edible seaweeds as a food source. Some of these seaweeds contain biologically active compounds including fucoidans and alginates that have anti-oxidant, anti-inflammatory, anti-allergy, anti-tumor, anti-obesity, anti-coagulant, and anti-viral properties. This project will take advantage of seaweed samples harvested from the Salish Sea region that were used for measurements of toxin content. Although these seaweeds contain toxins they also have numerous health benefits that likely outweigh the detrimental effects of the toxins. Traits are commonly influenced by environmental and genetic factors, yet the contribution of each is often poorly understood. An important life history trait in baker’s yeast (Saccharomyces cerevisiae) is the rate at which cells prepare for mating in response to environmentally released pheromones. Wild strains of yeast differ in mating pheromone response and alleles of the G-Protein alpha subunit of the pheromone receptor have been shown to affect both pheromone response and fitness. Polymerized hemoglobin (Hb) molecules have been shown to decrease previously observed adverse events associated with the administration of cell-free hemoglobin. To create these polymers, a method will be developed which employs the site specific ligation reaction of the sortase A enzyme from S. aureus. An Hb mutant (“αcpβ“) previously developed in our lab has been further modified by adding either the sortase recognition sequence, LPXTG, to the C-terminus of the α-subunit (s-αcpβ), or a tetraglycine motif, GGGG, to the N-terminus (n-αcpβ). Bacterial sortases have been widely studied for their usefulness in protein modification, however, the variable substrate specificity and activity between homologs of these enzymes is not yet fully characterized. To attempt to further understand sorting signal recognition, we are working towards a substrate bound structure of sortase A from Streptococcus pneumoniae (SrtApneu). This enzyme displays a wide tolerance for alternate amino acids within the canonical LPXTG sorting motif. Reservoir drawdown following the removal of two hydroelectric dams on the Elwha River, Washington, exposed ~800 acres of sediments and left a barren landscape in the former reservoirs. The establishment of native vegetation within these areas is critical to meeting restoration objectives. Large woody debris (LWD) can play an important role in riparian ecosystems and may assist in the development of early plant communities. LWD helps create favorable microsites for seedlings by creating shade, which decreases temperatures and increases moisture levels in sediments. There are long-standing uncertainties about toxicity of chemical mixtures to populations. Laboratory toxicity tests have confirmed synergistic and antagonistic effects to individuals, but not to populations.We will conduct a regional scale ecological risk assessment by evaluating the effects chemical mixtures to populations with a new Bayesian Network- Relative Risk Model (BN-RRM) incorporating an Adverse Outcome Pathway (AOP). We started applying this new BN-RRM framework in a case study with organophosphate pesticide mixtures (diazinon, chlorpyrifos, and malathion). Between 1987 and 2000, nesting populations of Pelagic Cormorant (Phalacrocorax pelagicus; PECO) and Double-crested Cormorant (P. auritus; DCCO) declined in the Strait of Georgia, BC. This northern section of the Salish Sea is a rapidly urbanizing area, and piscivorous birds are important indicators of ecosystem health. To update population status, we conducted a complete survey of 35 PECO and 23 DCCO colonies in July 2014 and opportunistic surveys of some colonies between 2001 through 2015. The PECO population decreased from ~2100-2400 nests in 1959-1987 to ~1100 nests by about 2000, and then rose slightly …

The first fish tracking arrays were deployed in the Salish Sea over a decade ago. These arrays have yielded a rich data set which have provided the first direct estimates of early marine-survival and migratory behavior for acoustic-tagged juvenile sockeye, Chinook, Coho and steelhead >130 mm in fork length (FL). In spring of 2015, as part of the Salish Sea Marine Survival Project, the Pacific Salmon Foundation, the Ocean Tracking Network and Kintama Research deployed additional arrays in the Discovery Islands and Johnstone Strait (north of the Strait of Georgia) to provide higher resolution survival data.
