Enhanced tumor delivery of plasmid DNA with electric pulses in vivo has been confirmed in many preclinical models. Intratumor electrotransfer of plasmids encoding therapeutic molecules has reached Phase II clinical trials. In multiple preclinical studies, a reduction in tumor growth, increased survival or complete tumor regression have been observed in control groups in which vector or backbone plasmid DNA electrotransfer was performed. This study explores factors that could produce this antitumor effect. The specific electrotransfer pulse protocol employed significantly potentiated the regression. Tumor regression was observed after delivery of single-stranded or double-stranded DNA with or without CpG motifs in both …

Streptococcus pyogenes, group A streptococci (GAS), causes diseases ranging from asymptomatic to life threatening. Some strains of S. pyogenes produce extracellular hyaluronate lyase (HylA), a potential virulence factor. HylA is an enzyme that degrades hyaluronic acid, which is found in the extracellular matrix of human tissues. The breakdown of the host tissue contributes to the spread of infection. The hylA gene is not constitutively expressed in vitro, which implies regulation.
A proposed regulator of hyaluronate lyase expression is RegR, a LacI/GalR like protein. A vector containing disrupted regR was electro-transformed into S. pyogenes M-type 22 strain 10403. Growth characteristics …

Seagrass habitats have long been known to serve as nursery habitats for juvenile fish by providing refuges from predation and areas of high forage abundance. However, comparatively less is known about other factors structuring fish communities that make extensive use of seagrass as nursery habitat. We examined both physical and biological factors that may structure the juvenile seagrass-associated fish communities across a synoptic-scale multiyear study in lower Chesapeake Bay. Across 3years of sampling, we collected 21,153 fish from 31 species. Silver Perch Bairdiella chrysoura made up over 86% of all individuals collected. Nine additional species made up at least 1% …

Malaria is among the most devastating human diseases, and a majority of lethal cases are caused by the protozoan parasite, Plasmodiumfalciparum. The emergence of multi-drug resistant P.falciparum is a major obstacle to malaria control and is highlighted by the abandonment of chloroquine (CQ) as a first-line treatment of P.falciparum infections worldwide. Chloroquine resistance (CQR) is associated primarily with mutations in the transmembrane digestive vacuole protein, PfCRT. However, CQR P.falciparum parasites harboring the same mutant pfcrt allele vary in their CQ response, suggesting the CQ response is multigenic in nature. No gene outside of pfcrt is completely associated with CQR. Thus, …

Exposure of cells to a pulsed electric field (PEF) is the basis of multiple techniques and treatments. Nanosecond pulsed electric field (nsPEF) poses unique characteristics to induce subtle cellular effects while preserving cell integrity. Improving understanding of the mechanisms triggered by nsPEF in cells inspires new applications for the nanosecond pulse technology. Although many effects of nsPEF remain unknown, they can be inferred from morpho-physiologic changes, or cell reshaping, that accompany nsPEF exposure. During the exposure cells undergo reshaping that is manifested in swelling and diffuse blebbing. Springtime Contribution Of Dinitrogen Fixation To Primary Production Across The Mediterranean Sea, E. Rahav, B. Herut, A. Levi, Margaret Mulholland, I. Berman-Frank

Dinitrogen (N-2) fixation rates were measured during early spring across the different provinces of Mediterranean Sea surface waters. N-2 fixation rates, measured using N-15(2) enriched seawater, were lowest in the eastern basin and increased westward with a maximum at the Strait of Gibraltar (0.10 to 2.35 nmol NL-1 d(-1), respectively). These rates were 3-7 fold higher than N-2 fixation rates measured previously in the Mediterranean Sea during summertime and we estimated that methodological differences alone did not account for the seasonal changes we observed. The pathogenic virus Panulirus argus virus 1 (PaV1) was first discovered in Caribbean spiny lobsters Panulirus argus from the Florida Keys (USA) in 1999 and has since been reported in Belize, Mexico, and Cuba; its distribution in the wider Caribbean is unknown. We collected tissue samples from adult spiny lobsters from 30 locations in 14 countries bordering the Caribbean Sea and used molecular diagnostics to assay for the presence of PaV1. PaV1 occurred primarily in the northern areas of the Caribbean, where its prevalence was highest. The virus was not found in lobsters from the southeastern Caribbean, and its prevalence …

Patellofemoral joint pain (PFP) is one of the most common afflictions of the active population. While etiological factors encompassing the entire lower extremity have been associated with PFP, participation in athletic activities while fatigued may further influence abnormal biomechanics in individuals with this condition. The aim of this study was to examine changes in postural and locomotor control under varying task demands. Three experiments were designed to address the impact that fast walking had on standing posture over time, slow walking had on gait dynamics over time, and the extent to which gait speed interacts with the ability to walk randomly.

Costal cartilage is a type of hyaline cartilage that forms rod-like structures that connect the ribs to the sternum. Deformation of costal cartilage is observed in the chest wall deformities, pectus excavatum and pectus carinatum. Pectus excavatum involves a sternal displacement causing a depression of the chest while pectus carinatum causes a protrusion of the chest. As costal cartilage is not a widely studied tissue, this leaves little knowledge into possible factors involved in the pathogenesis of pectus deformities. Relatively little is known about the relationship between the hispid cotton rat (Sigmodon hispidus) and the eastern harvest mouse (Reithrodontomys humulis). Literature on these two syntopic genera in other areas of the United States suggests that competition may exist; however, both species seem to be greatly affected by environmental changes, making it difficult to draw any definite conclusions. The Coral Triangle (CT) is a hotspot for marine species diversity as well as for intraspecific genetic diversity. Here, nuclear RAG2 and mitochondrial D-Loop genes were used to identify deep genetic divergence among Dascyllus aruanus (Linnaeus, 1758) populations across relatively short scales within the CT. Mitochondrial clades different by greater than 20 mutational steps were geographically isolated from one another across the distance between Java and the Lesser Sunda Islands, and also east and west across the Philippines. Evidence for population structure in the Sulu Sea and at the Lesser Sunda Islands is also identified. The results suggest that the …

This study investigated reproductive behaviours of the carrion beetles (Coleoptera: Silphidae) of southeastern Virginia. Carrion beetles have been known to have ranges of carcass masses used for reproductive behaviors. In this study, pitfall traps were baited with 25g, 50g, or 75g of a cat food/mackerel mixture and left to decay and attract beetles for one week to investigate if any Silphidae species prefer a single mass over another. Many questions remain regarding the origins of the well-known peak in marine biodiversity found in the Coral Triangle. Barriers to gene flow may promote lineage diversification and provide the potential for speciation contributing to the high biodiversity in this area. Population structure was examined in two species of Emperor Snappers (Lethrinidae), Lethrinus harak and Lethrinus lentjan. These species were selected because they share similar life-history traits and geographic distributions, but differ in their habitat preferences. A short-term longitudinal study was used to examine the relations of achievement motivation, general motivation, interest, and self-regulatory predictors to cognitive strategy use and academic performance. A total of 102 college undergraduates enrolled in their first college-level introductory psychology course completed an Achieving Tendency Scale of achievement motivation, the Motivated Strategies For Learning Questionnaire, and an Interest Profile in psychology during the first two weeks of the semester. At midterm they completed the Study Activity Survey of cognitive strategy use. At the end of the semester exam grades, total points earned in the course, and grade point average were obtained. …

Coastal eutrophication resulting from nutrient over-enrichment plagues coastal waterways worldwide. In addition to dissolved inorganic N, many marine microbes, including phytoplankton, have the capacity to use certain dissolved organic nitrogen (DON) compounds. However, while we know that many specific DON compounds are labile, the bulk of the DON pool in nature is uncharacterized. The bioavailability of DON to estuarine microbes currently remains unclear.

Cartilage deformities within the human chest wall, specifically pectus excavatum (PE) and pectus carinatum (PC) are common (1/400-1/1000) and yet, despite their clinical significance, are some of the least studied disorders pertaining to cartilage [1]. The costal cartilage connecting "false ribs" 8-10 to the sternum is often abnormally grown and can lead to formation of a severely sunken "funnel" chest (PE) or push outwards to form a "pigeon" chest (PC). In the nitrogen (N)-impoverished photic zones of many oceanic regions, prokaryotic organisms fixing atmospheric dinitrogen (N2; diazotrophs) supply an essential source of new nitrogen and fuel primary production. We measured dinitrogen fixation and primary productivity (PP) during the thermally stratified summer period in different water regimes of the oligotrophic eastern Mediterranean Sea, including the Cyprus Eddy and the Rhodes Gyre. Low N2 fixation rates were measured (0.8-3.2μmol N m-2 d-1) excluding 10-fold higher rates in the Rhodes Gyre and Cyprus Eddy (~20μmol N m-2 d-1). Objectives were to model fish species richness relative to natural and anthropogenic variables in Quantico Creek, a forested undisturbed stream environment, and Cameron Run, a highly disturbed urban stream environment in the lower Piedmont-Fall Line region of the Potomac River watershed. Species richness in all stream orders (e.g. avg. range=2.5-9.65 in 1st-3rd orders) of Quantico Creek were significantly higher than those (e.g. avg. range=2.1- 7.6 in 1st -4th orders) of Cameron Run. The communities of small mammals were evaluated for 13 months with capture-mark-recapture methods in two Spartina-Juncus marshes of the Atlantic coast in Northampton County, Virginia. Small mammals were trapped for three days each month using live traps placed on floats on two study grids. Two rodents were numerically dominant (~90% of small mammals) there: marsh rice rat, Oryzomys palustris, and meadow vole, Microtus pennsylvanicus. Monthly estimates of population density were greater for rice rats (peak: 45/ha) than for those of meadow voles (peak: 30/ha). Survival rates were generally low, especially for rice rats, indicating highly vagile populations. Both …
