Frontotemporal dementia (FTD) with inclusion body myopathy and Paget disease of bone (IBMPFD) is a rare, autosomal-dominant disorder caused by mutations in the valosin-containing protein (VCP) gene, a member of the AAA-ATPase gene superfamily. The neuropathology associated with sporadic FTD is heterogeneous and includes tauopathies and frontotemporal lobar degeneration with ubiquitin-positive inclusions (FTLD-U). However, there is limited information on the neuropathology in IBMPFD. We performed a detailed, systematic analysis of the neuropathologic changes in 8 patients with VCP mutations. Muscle fatigue is a common clinical sign and possible precursor to musculoskeletal injury. Existing methods of detecting muscle fatigue qualify changes within muscle or across joints, but are expensive, invasive or difficult to use. A technique of mechanomyography utilising maximal percutaneous stimulation (known as the Muscle Displacement Laser {MDL}) is an easy non-invasive assessment tool, which may have considerable application in measuring muscle fatigue and determining recovery. This investigation aimed to determine the neuromuscular response of the bicep brachii during heavy (6RM) dynamic resistance exercise performed to volitional exhaustion. Electromyographic (EMG) activity was compared in twelve male subjects performing dominant-limb elbow flexion exercise during four conditions: i) Explosive concentric and eccentric (SSC); ii) Explosive concentric, control eccentric (EC); iii) control (C); and iv) control with limb occlusion (CO) (110mmHg). CO and SSC are reported to increase motor unit activity when compared to control exercise (2 second flexion, 2 second extension). The traditional in vitro correlate of immunological protection is the induction by a vaccine of neutralizing antibodies against the virus. It was formerly assumed that protection induced by a vaccine was solely due to neutralizing antibodies. Neutralizing antibodies are potent in the prevention of certain diseases, but virus-specific CD4+ T helper cells aid in the proliferation of specific antigen-triggered B cells to make antibodies. CD8+ T cells are responsible for eliminating virus-infected cells during viral illness, and may act as a second line of defence by becoming activated and eliminating any cells that become infected despite the presence of neutralizing …

Kainate receptors containing the GluK1 subunit have an impact on excitatory and inhibitory neurotransmission in brain regions, such as the amygdala and hippocampus, which are relevant to seizures and epilepsy. Here we used 2-amino-3-(3-hydroxy-5-tert-butylisoxazol-4-yl) propanoic acid (ATPA), a potent and selective agonist of kainate receptors that include the GluK1 subunit, in conjunction with mice deficient in GluK1 and GluK2 kainate receptor subunits to assess the role of GluK1 kainate receptors in provoking seizures and in kindling epileptogenesis.
