Genetic Analysis Of Drought Tolerance In Rice (Oryza Sativa L.), Uttam Bhattarai

Drought is a major challenge in rice production system worldwide. We conducted a phenotypic screening of the USA rice genotypes for drought tolerance and assessed genetic diversity using SSR markers. Identification of QTLs for drought tolerance during both vegetative and reproductive stage was done using genotyping by sequencing (GBS) based saturated SNP linkage map. The rice genotypes Jes, Leah, Roy J, Jazzman, and Madison were found to be drought tolerant. Population structure analysis grouped the USA rice genotypes into California, Louisiana, and Arkansas types. Developing perennial grain crops is an effective way for sustainable agriculture and it can be a tool for the betterment of the overall ecosystem. To understand how perenniality works in perennial species, we made a cross between Zea diploperennis and maize (Z. mays) inbred line B73. Developing perennial grain crops is an effective and crucial way to prevent soil erosion caused by conventional agriculture of using annual crops while meeting the increasing need of global food demand. We hypothesized that the regrowth in Zea might be controlled by two dominant complementary genes. F1 hybrids were created by crossing Zea diploperennis Iltis, Doebley & R. Due to the threat of salinity stress to Louisiana rice production, an effort was made to understand the molecular genetics of salinity tolerance with the overall goal of developing salt tolerant varieties. The objectives of this study were to 1) determine if salinity tolerance exist in the US rice varieties, 2) map the additive and epistatic QTLs for traits related to seedling salinity tolerance in recombinant inbred lines (RILs) using genotyping-by-sequencing (GBS)-derived SNP markers, and 3) identify and validate stable QTLs and their effects in introgression lines (ILs) of Pokkali in Bengal background.
