Barbarea integrifolia DC. (Brassicaceae) is an endangered and endemic species located in Erzincan and Gümüşhane provinces of Turkey. In total, 27 individuals from 2 natural populations were assessed using the random amplified polymorphic DNA polymerase chain reaction (RAPD-PCR) method coupled with sequence analysis of the internal transcribed spacer (ITS1) rDNA region. Genetic accuracy of RAPD-PCR was tested with 25 RAPD primers, and it resulted in 115 clear and reproducible DNA fragments from 13 RAPD primers. Among these, 76 (66.1%) fragments were found to be polymorphic. According to the ITS1 analysis of B. integrifolia populations, the plants exhibited a relatively poor …

Identification Of Stable Sources For Surrogate Traits In Arachis Glabrata And Marker-Trait Association For Tolerance To Water Deficit Stress, Sandip Kumar Bera, Ajay Basapur Chandrashekar, Sneha Patel, Vrajlal Kanji Sojitra, Anil Maurya Jan 2014

Diploid and tetraploid wild relatives of cultivated peanut are genetically diverse, providing rich sources of genetic resources for tapping resistance to various biotic and abiotic stresses. Arachis glabrata Benth. is a tetraploid (2n = 40) rhizomatous species that is reported to be tolerant to water deficit stress. Identification of A. glabrata accessions with high SPAD chlorophyll meter reading (SCMR) and low specific leaf area (SLA) values would be an option for improving water deficit stress tolerance in peanut. An investigation was undertaken to study the genetic diversity of A. glabrata accessions using molecular markers as well as surrogate traits (SLA …

Molecular Variability Of Plantain Ecotypes From The Genus Musa (Musaceae), Rakeshkumar Choudhary, Raghunath Keshavachandran, Rema Menon, Ganesh Khalekar, Nirbhay Singh, Divya Maruthiyottu Jan 2014

Molecular Variability Of Plantain Ecotypes From The Genus Musa (Musaceae), Rakeshkumar Choudhary, Raghunath Keshavachandran, Rema Menon, Ganesh Khalekar, Nirbhay Singh, Divya Maruthiyottu

Turkish Journal of Botany

Twelve plantain ecotypes were characterized for molecular variability using random amplified polymorphic DNA (RAPD) and inter simple sequence repeat (ISSR) markers. Out of the 60 RAPD and 40 ISSR primers tested, 16 RAPD and 14 ISSR primers were found polymorphic, and were used for molecular profiling of the plantain ecotypes. The average number of bands per primer, average number of polymorphic bands per primer, mean polymorphic information content (PIC) value, resolving power (Rp), and marker indices (MI) for RAPD primers were 8.63, 3.25, 0.87, 15.49, and 2.86, whereas the respective values for the ISSR assay were 7.93, 4.14, 0.85, 12.49, …

Reproduction And Diversity Of The Endangered Salix Lapponum L. Populations In Eastern Poland, Magdalena Pogorzelec, Katarzyna Glebocka, Barbara Hawrylak-Nowak, Marzena Parzymies Jan 2014

Reproduction And Diversity Of The Endangered Salix Lapponum L. Populations In Eastern Poland, Magdalena Pogorzelec, Katarzyna Glebocka, Barbara Hawrylak-Nowak, Marzena Parzymies

Turkish Journal of Botany

The main aim of the study was to increase the knowledge on the ecology and biology of the boreal relict Salix lapponum L., an endangered plant in Poland, in order to create an effective protection program. An important aspect was to determine the within- and among-population genetic diversity of the studied species in order to evaluate whether the basic stages of the generative reproduction cycle in the populations were correct. The results confirmed that S. lapponum pollen viability and germinability, for both fresh and stored pollen, were high and so were the seed germination ability and dynamics. All individuals were …

The Assessment Of Genetic Diversity Of Castanea Species By Rapd, Aflp, Issr, And Ssr Markers, Sofiane Abdelhamid, Cong-Linh Lê, Marco Conedera, Philippe Küpfer Jan 2014

The Assessment Of Genetic Diversity Of Castanea Species By Rapd, Aflp, Issr, And Ssr Markers, Sofiane Abdelhamid, Cong-Linh Lê, Marco Conedera, Philippe Küpfer

Turkish Journal of Botany

Castanea Mill. (chestnut) is a multipurpose deciduous tree and member of the family Fagaceae, widely distributed throughout North America, Europe, and Asia. The evaluation of the genetic diversity of chestnut species is crucial for the effective conservation of this economically and ecologically valuable tree. In this study, we applied 4 DNA markers to detect the genetic variability among and within Castanea species and to compare the effectiveness of each system in estimating genetic variation. We amplified 106 random amplified polymorphic DNA (RAPD), 228 amplified fragment length polymorphism (AFLP), 42 intersimple sequence repeat (ISSR), and 36 simple sequence repeat (SSR) polymorphic …
