Sorghum Wrky Transcription Factor Sbwrky45 Enhanced Seed Germination Under Drought Stress In Transgenic Arabidopsis, Elamin Baillo, Muhammad Sajid Hanif, Ping Xu, Zhengbin Zhang

Turkish Journal of Botany

The WRKY transcription factors (TFs) family is an important family of plant-specific TFs, playing vital roles in various abiotic and biotic stress responses. WRKY TFs are gaining considerable attention due to their significant roles in stress responses. However, their functions in sorghum (Sorghum bicolor) are lagging behind. In this study, a WRKY gene designated as SbWRKY45 was isolated and characterized from sorghum. SbWRKY45, belonging to Group IIa, consists of one intron and two exons and encodes 430 amino acids. SbWRKY45 is located in chromosome 4. The cis-element prediction analysis showed that the promoter region of SbWRKY45 has several abiotic …

Rubisco And Abiotic Stresses In Plants: Current Assessment, Abdulbaki Shehu Abdulbaki, Hameed Alsamadany, Yahya Alzahrani, Bolaji Umar Olayinka

Turkish Journal of Botany

Abiotic stresses are serious environmental factors militating against the production of many crops around the world. The consequence of this, is the difficulty of meeting the demands of the increasing world population. Aside from other negative effects, reduction in photosynthesis is an important feature of abiotic stresses. Abiotic stresses limit photosynthesis in a number of ways. The reduction in ribulose 1, 5-bisphosphate carboxylase/oxygenase (Rubisco) content and activity is one of the paramount ways through which abiotic stresses affect photosynthesis. Rubisco is the CO2 fixing enzyme of photosynthesis and also catalyses the photo-respiratory carbon oxidation. The enzyme has low turnover and …

Synergism And Antagonism In Plant Acclimation To Abiotic Stress Combinations, Karl-Josef Dietz Jan 2021

Synergism And Antagonism In Plant Acclimation To Abiotic Stress Combinations, Karl-Josef Dietz

Turkish Journal of Botany

Acclimation of plants to their environment involves mechanisms at all molecular levels and only the complete temporal and spatial pattern adequately describes the acclimation state. The diversity of physiological states realized in plants increases by the essentially infinite combinations of abiotic and biotic stresses, considering duration, impact order, and strength. The interference between stressors involves phenomena of synergism, antagonism, and indifference. This review exemplarily sketches principles of interference considering posttranslational modifications, the formation of molecular assemblies in regulosomes, the regulation of gene expression, and the vast number of other posttranscriptional controls e.g., in RNA processing and recruitment to ribosomes. Small …

Mirnas Involved In Drought Stress In Italian Ryegrass (Lolium Multiflorum L.), Gürkan Demi̇rkol

Turkish Journal of Botany

miRNAs have been characterized as a regulator of main processes in plants by silencing genes. The functions of microRNAs have been studied in various crops, hovewer, no studies have been observed about miRNAs in Italian rygrass (Lolium multiflorum L.) against drought stress. This experiment aimed to reveal the involvement of miRNAs against drought stress in sensitive and tolerant Italian ryegrass genotypes. Four genotypes (G1 and G2 as drought sensitive - G3 and G4 as drought tolerant) were selected. The sensitivities of these genotypes against drought stress were verified by performing growth parameters, relative water and proline contents under normal and …

Competitiveness Of Generalist Plant Populations At A Mine Site, Justin S.H. Wan, Fati̇h Fazlioğlu, Stephen P. Bonser Jan 2019

Competitiveness Of Generalist Plant Populations At A Mine Site, Justin S.H. Wan, Fati̇h Fazlioğlu, Stephen P. Bonser

Turkish Journal of Botany

Plants inhabiting extremely stressful mine site environments tend to be specialized and localized, where they express lower performance than nonmine site plants from adjacent areas. However, such a cost may be concealed. In a previous study at a mine site, we found mine and adjacent nonmine plants of multiple species expressed similar performances in the absence of competition. However, a lower competitive ability may be a concealed driver. We aim to test whether costs under competition could explain specialization. In a glasshouse experiment, we measured the performance (i.e. vegetative and reproductive growth) of mine and nonmine potted plants under competition. …

Roles Of Plant Cbl?Cipk Systems In Abiotic Stress Responses, Shilian Huang, Shaofeng Jiang, Junsong Liang, Miao Chen

Turkish Journal of Botany

Plants evolved from long-term adaptation to form regulatory mechanisms of perception, transduction, and response to stresses. The CBL?CIPK signaling system is a basic calcium sensor that plays an important role in sensing adverse environmental stimuli. CBLs can perceive and bind Ca2+ under adversity and then specifically interact with CIPKs. The activated CBL?CIPK complex phosphorylates downstream target proteins (such as ion channels and transporters), or regulates transcription factors and stress responsive genes. Important progress has been made in the study of CBL?CIPK signaling pathways in response to high salt, low N, low K, high Mg, and high pH. This review summarizes …

Phenotypic Plasticity And Specialization Along An Altitudinal Gradient In Trifolium Repens, Fati̇h Fazlioğlu, Justin S.H. Wan, Stephen P. Bonser Jan 2018

Phenotypic Plasticity And Specialization Along An Altitudinal Gradient In Trifolium Repens, Fati̇h Fazlioğlu, Justin S.H. Wan, Stephen P. Bonser

Turkish Journal of Botany

Phenotypic plasticity and specialization represent seemingly contrasting strategies to heterogeneous environments. Specialization is associated with the loss of phenotypic plasticity, particularly in functional traits. However, it is equivocal if this loss of plasticity is observed only in the specific habitat and stress type where the specialization occurs or a general loss of plasticity is seen across habitats. We examined populations of Trifolium repens L. following an expansion during the colonization of Australia from relatively good low altitude habitats to more stressful high altitude habitats in the Blue Mountains region, New South Wales, Australia. We examined if specialization to abiotic stress …

Metabolic And Molecular-Genetic Regulation Of Proline Signaling And Itscross-Talk With Major Effectors Mediates Abiotic Stress Tolerance In Plants, Aryadeep Roychoudhury, Aditya Banerjee, Vikramjit Lahiri Jan 2015

Metabolic And Molecular-Genetic Regulation Of Proline Signaling And Itscross-Talk With Major Effectors Mediates Abiotic Stress Tolerance In Plants, Aryadeep Roychoudhury, Aditya Banerjee, Vikramjit Lahiri

Turkish Journal of Botany

Proline (Pro) accumulation is a common response of several plant species to combat abiotic stresses. Under stress conditions, Pro acts as an excellent compatible solute in the plant system, participating in the alleviation of stress sensitivity. Though the metabolic pathways associated with Pro are well studied, parts of its regulatory cascades are still not properly known. It has also been conjectured that epigenetic modifications regulate Pro metabolism during abiotic stress. Apart from Pro, the plant abiotic stress responses are essentially mediated by multiple effectors. Hence, proper analysis of the cross-talks of Pro with the other components of the abiotic stress …

Screening Of Pvlea3 Gene Mrna Expression Levels With Qrt-Pcr In Differentbean Varieties (Phaseolus Vulgaris L.) Subjected To Salt And Drought Stress, İlker Büyük, Sümer Aras

Turkish Journal of Botany

Bean is an edible grain legume species with the greatest selectivity in terms of ecological conditions. Increasing the yield per unit area depends on identification and breeding of species resistant to different ecological conditions. In this regard, we aimed to evaluate mRNA levels of the PvLEA3 gene and lipid peroxidation levels in 7 different bean varieties subjected to NaCl and PEG stresses. Lipid peroxidation analysis (MDA) and qRT-PCR analysis were performed with root and leaf tissues sampled at 3 and 27 h of stress conditions. All results showed that the PvLEA3 gene plays a key role in the defense mechanism …

Turkish Journal of Botany

Cold stress is a major environmental factor in plant life cycles. Nicotiana benthamiana, which belongs to the family Solanaceae, is one of the most commonly used model species in plant-microbe interaction studies. In total, 5205 differentially expressed genes were identified under cold stress in N. Benthamiana. Of these, 5029 were upregulated and 176 were downregulated within four time periods (4 h, 12 h, 24 h, and 48 h). The common up- and downregulated genes were identified as 692 and 6, respectively. The functional annotations of these genes were studied and these common genes involved in protein, RNA, miscellaneous enzyme families, …

Turkish Journal of Botany

The family of AP2/ERF plant specific transcription factors, including dehydration-responsive element binding (DREB) proteins, has been reported to activate and bind with the dehydration responsive element/C repeat (DRE/CRT) of stress-inducible gene promoters. We amplified DREB1A from Oryza sativa var. IR6 (807 bp) and subjected it to protein homology modelling and phylogenetic analysis. BLAST sequence analysis indicated high similarity (99%) with the O. sativa japonica group. Phylogenetic analysis was carried out with other 12 DREB sequences from cereals/grasses using Clustal Omega. Protein secondary structure and 3D models were determined using SOPMA and MODELLER v9.10, respectively. PROSA was used to determine the …

Expression Of Sod Gene And Evaluating Its Role In Stress Tolerance In Nacl And Peg Stressed Lycopersicum Esculentum, Semra Aydin, İlker Büyük, Emi̇ne Sümer Aras

Turkish Journal of Botany

Antioxidant enzyme mechanisms are important for protecting crop productivity against stressful environmental conditions. Therefore, the current study was designed to evaluate lipid peroxidation (via malondialdehyde (MDA)) levels, superoxide dismutase (SOD) gene expression profiles, and SOD enzyme activities in tomato (Lycopersicum esculentum L.) plants subjected to different concentrations (100 and 150 mM) and time periods of NaCl and polyethylene glycol (PEG) stress (3 h, 6 h, 9 h, 12 h, and 48 h). Enhancement of lipid peroxidation in plants may be attributed to the increased accumulation of reactive oxygen species (ROS). Results indicated the antioxidant responses of tomato plants could be …
