Assessing The Genetic Diversity Of The Genus Viburnum Using Simple Sequence Repeats, Deborah Dean

The genus Viburnum was established in 1753 by Linnaeus and is the largest genus in the Adoxaceae and consists of approximately 160 species. Viburnum species are small trees which grow throughout the temperate regions of the Northern Hemisphere (Hoch, 1995). While this genus shares little variety in fruit and flower morphology, it is diverse in many other traits. As molecular studies advance, this large genus continues to undergo reclassification. Here three SSR libraries were constructed to discern additional molecular insight into this vast genus.

Economically, legumes (Fabaceae) represent the second most important family of crop plants after the grass family, Poaceae. Grain legumes account for 27% of world crop production and provide 33% of the dietary protein consumed by humans, while pasture and forage legumes provide vital part of animal feed. Fabaceae, the third largest family of flowering plants, has traditionally been divided into the following three subfamilies: Caesalpinioideae, Mimosoideae, and Papilionoideae, all together with 800 genera and 20,000 species. Castanea Mill. (chestnut) is a multipurpose deciduous tree and member of the family Fagaceae, widely distributed throughout North America, Europe, and Asia. The evaluation of the genetic diversity of chestnut species is crucial for the effective conservation of this economically and ecologically valuable tree. In this study, we applied 4 DNA markers to detect the genetic variability among and within Castanea species and to compare the effectiveness of each system in estimating genetic variation. The main aim of the study was to increase the knowledge on the ecology and biology of the boreal relict Salix lapponum L., an endangered plant in Poland, in order to create an effective protection program. An important aspect was to determine the within- and among-population genetic diversity of the studied species in order to evaluate whether the basic stages of the generative reproduction cycle in the populations were correct. Diploid and tetraploid wild relatives of cultivated peanut are genetically diverse, providing rich sources of genetic resources for tapping resistance to various biotic and abiotic stresses. Arachis glabrata Benth. is a tetraploid (2n = 40) rhizomatous species that is reported to be tolerant to water deficit stress. Identification of A. glabrata accessions with high SPAD chlorophyll meter reading (SCMR) and low specific leaf area (SLA) values would be an option for improving water deficit stress tolerance in peanut. Twelve plantain ecotypes were characterized for molecular variability using random amplified polymorphic DNA (RAPD) and inter simple sequence repeat (ISSR) markers. Out of the 60 RAPD and 40 ISSR primers tested, 16 RAPD and 14 ISSR primers were found polymorphic, and were used for molecular profiling of the plantain ecotypes. The average number of bands per primer, average number of polymorphic bands per primer, mean polymorphic information content (PIC) value, resolving power (Rp), and marker indices (MI) for RAPD primers were 8.63, 3.25, 0.87, 15.49, and 2.86, whereas the respective values for the ISSR assay were 7.93, 4.14, 0.85, 12.49, …

Barbarea integrifolia DC. (Brassicaceae) is an endangered and endemic species located in Erzincan and Gümüşhane provinces of Turkey. In total, 27 individuals from 2 natural populations were assessed using the random amplified polymorphic DNA polymerase chain reaction (RAPD-PCR) method coupled with sequence analysis of the internal transcribed spacer (ITS1) rDNA region. Genetic accuracy of RAPD-PCR was tested with 25 RAPD primers, and it resulted in 115 clear and reproducible DNA fragments from 13 RAPD primers. Among these, 76 (66.1%) fragments were found to be polymorphic. According to the ITS1 analysis of B. integrifolia populations, the plants exhibited a relatively poor …
