Positive associations among species have contributed to the maintenance of biodiversity. These interactions are typically studied in pairwise fashion where two interacting organisms are the focus. This dissertation uses network theory to analyze positive plant-ant associations in three Neotropical forests. Obligate plant-ant mutualisms were used, where plants host the ants in domatia and provide food bodies while the ants protect the plants and can feed them. A network approach was used to depict all obligate plant-ant interactions within a community. Spotted knapweed is an invasive, short-term-perennial plant that is native to Eurasia. It was accidentally introduced into North America in the early 1890's and has since spread across The United States and Canada. Spotted knapweed degrades rangelands and pastures by negatively impacting native plants, increasing soil surface runoff and stream sediment yields, and reducing soil infiltration. A biological control program for spotted knapweed using Larinus minutus (Coleoptera: Curculionidae), was initiated in Arkansas in 2008. Considerable confusion exists regarding the infection process and the disease cycle. Site-specific recombination systems are powerful tools for genetic modification. They have been used to integrate a transgene into a pre-defined locus and to remove marker genes from a transgene locus. Two of the most widely used site-specific recombination systems in plants are the Cre/lox system from the bacteriophage P1 and the FLP/FRT system from the yeast Saccharomyces cerevisiae. The Cre/lox system is well-characterized and is the first choice in application of site-specific recombination system. The genus Manfreda Salisb. of Asparagaceae is a genus of potential horticultural interest and is currently subject to breeding efforts at the University of Arkansas. A lack of taxonomic clarity however undermines the classification of potential inter - and intrageneric hybrids. The study aims to assess existing species delimitation within the genus Manfreda employing morphology while investigating the potential utility of Consortium for the Barcode of Life Plant (CBOL) DNA Barcodes for identification of specific taxa and an External Transcribed Spacer (ETS) - Internal Transcribed Spacer (ITS) DNA barcode for developed hybridized taxa.

Recent studies on the colonization time of Hawaiian plant and animal lineages have suggested certain lineages arrived to the archipelago before the formation of the extant islands. Kadua, a recently resurrected genus, is mostly endemic to the Hawaiian Archipelago, with many member taxa autochthonous to individual islands. This work investigates the colonization time of Kadua to the Hawaiian Islands. Sequence analyses of the quickly evolving nuclear regions ITS, ETS and 5s-NTS regions for 20 Hawaiian and 7 French Polynesian species of Kadua were used to create dated phylogeny calibrated with island ages. Here, I characterize a root-branching hypermorph of Lotus japonicus, called cluster root-like1 (crl1), which originated from a har1-1 suppressor screen. I have shown that the crl1 root phenotype is determined by two independently segregating recessive mutations, har1-1 and Ljamp1-1, with corresponding HAR1 and LjAMP1 encoding an LRR‑receptor-like kinase and a predicted homologue of the Arabidopsis ALTERED MERISTEM PROGRAM 1 protein, respectively. Unlike har1-1, the Ljamp1 mutation does not affect the symbiotic properties of L. japonicus Gifu but exerts a pleiotropic effect on shoot development. Weedy rice is a persistent weed of cultivated rice (Oryza sativa) fields worldwide, which competes with the crop and drastically reduces yields. Within the US, two main populations of genetically differentiated weedy rice exist, the straw-hulled (SH) group and the black-hulled awned (BHA) group. Current research suggests that both groups are derived from Asian cultivated rice. However, the weeds differ from the cultivated groups in various morphological traits. Involvement Of Hexokinase1 In Plant Growth Promotion As Mediated By Burkholderia Phytofimans, Jae Min Park

Burkholderia phytofirmans strain PsJN consistently enhanced the growth of potato plants in vitro. The role of hexokinase1 in glucose phosphorylation was investigated in plants with PsJN. Increased hexokinase1 activity only in roots of PsJN-treated plants cultivar Kennebec suggests that hexokinase1 is associated with plant root and stem growth.

Plant growth with PsJN was determined when plants were grown with different sugars at various concentrations. PsJN-treated plants expressed diverse forms of growth promotion. When growth promotion did occur, hexokinase1 activity also increased. Growth promotion and hexokinase1 activity appear to be correlated to the enzyme to recognize the substrate for catalytic activity. …

Cadmium (Cd) is a non-essential trace element and its environmental concentrations are increasing due to human activities. Edible plants can accumulate high concentrations of Cd, which could be toxic to humans. Plants are recognized as an efficient and inexpensive system to produce valuable recombinant proteins. However, the use of plants still faces two main limitations: the low accumulation levels of some recombinant proteins and the lack of efficient protein purification methods. Two fusion partners, elastin-like polypeptides (ELP) and hydrophobin I (HFBI) were found to increase the accumulation of recombinant proteins and induce the formation of protein bodies (PBs) when targeted to the ER in transient expression assays. Jacalin-related lectins (JRLs) are proteins that bind carbohydrates. Functionally, JRLs are thought to be involved in a diverse array of functions including biotic stress defense, intracellular storage, abiotic stress response, and plant development. Recent Arabidopsis thaliana microarray data indicate that a Jacalin-like domain-containing protein (JLL1) exhibits highly root specific expression. Previous transcriptomic and proteomic studies indicate that JLL1 may have a dual role in planta as a biotic stress defense protein and in plant development. During the formation of plant roots these processes take place at a region of the root tip called the meristem. Cytokinin and auxin are two plant growth hormones that influence this process. Nuclear pore complexes (NPCs) are vital to nuclear-cytoplasmic communication in eukaryotes. The yeast Thp1-Sac3-Cdc31-Sus1 complex, also known as the TREX-2 complex, is anchored to the NPC via the nucleoporin Nup1, and is essential for mRNA export. In this study, the Arabidopsis homolog of the yeast TREX-2 complex was discovered. Physical and functional evidence support the identification of the Arabidopsis orthologs of the yeast Thp1 and Nup1. Of three Sac3 Arabidopsis homologs, two are putative TREX-2 components. Surprisingly, none are required for mRNA export as is the yeast Sac3. Wind is a dominant abiotic factor that a tree experiences throughout its lifetime and can cause severe tree damage, resulting in risk of injury to humans, and economic and ecological losses. It is thought that trees develop structural properties and architectures that help withstand loading conditions by dissipating wind energy through damping mechanisms. The historic effects of fire and climate on stand dynamics in the ponderosa pine-dominated forests of the American Southwest are of increasing concern to land management agencies. Using present forest stand structure, the mixed-conifer forests of the volcanic features in El Malpais National Monument were analyzed at three separate sites: a cinder cone, an ancient basalt flow, and an isolated “island” (kipuka) completely surrounded by basalt flows. Increment cores were collected from 632 trees in 19 plots within the monument to obtain dates of establishment. The plant hormone cytokinin plays essential roles in many aspects of growth and development. The cytokinin signal is transmitted by a multistep phosphorelay to the members of two functionally antagonistic classes of Arabidopsis response regulators (ARRs): the type-B ARRs (response activators) and type-A ARRs (negative-feedback regulators). Previous studies have shown that mutations in AXR1, encoding a subunit of the E1 enzyme in the related to ubiquitin (RUB) modification pathway, leads to decreased cytokinin sensitivity. This research shows that the cytokinin resistance of axr1 seedlings is suppressed by loss-of-function of type-A ARRs and that the cytokinin resistance caused by …

The vascular flora of Breaks Interstate Park was documented during two growing seasons, 2008 and 2009; with supplemental collections made in 2010 and 2011. The project area is located in eastern Kentucky and western Virginia at the northeastern terminus of Pine Mountain along the Appalachian Plateau. A total of 118 families, 341 genera, and 549 species, varieties, and subspecies were documented from Breaks Interstate Park. Six main vegetative communities were described: mesophytic forest, upper slopes and ridgetops, ponds/wetlands, river bottomland and floodplain/streamside, sandstone outcrops, and disturbed/open areas. Algae are being considered as a possible tool for carbon dioxide mitigation because they uptake carbon dioxide during photosynthesis. Using flue gas from a coal-fired power plant as a carbon source would allow the algae to remove CO2 from the flue gas before it is emitted into the atmosphere. This dissertation presents a comprehensive study on the origin and evolutionary relationships of the species of Bursera in Cuba, Hispaniola, Jamaica and the Bahamas. The goals of the first chapter were to test monophyly of the group, revisit a recent transfer of two species of Bursera to Commiphora, and place recently discovered mainland species using the reconstructed phylogenies. Additionally, divergence estimations using fossils were used as independent tests of several hypotheses regarding the arrival of the modern biota to the Greater Antilles and Bahamas (GAB).
