Cytokinesis, the process in which cytoplasm is apportioned between dividing daughter cells, requires coordination of myosin II function, membrane trafficking and central spindle organization. Most known regulators act during late cytokinesis; a few, including the myosin II-binding proteins anillin and supervillin, act earlier. Anillin's role in scaffolding the membrane cortex with the central spindle is well established, but the mechanism of supervillin action is relatively uncharacterized. We show here that two regions within supervillin affect cell division: residues 831-1281, which bind central spindle proteins, and residues 1-170, which bind the myosin II heavy chain (MHC) and the long form of …

Background: This study’s goal was to provide dose-response data for a dopamine agonist in the baboon using standard methods (replicate measurements at each dose, across a range of doses), as a standard against which to subsequently validate a novel pharmacological MRI (phMRI) method. Dependent variables were functional MRI (fMRI) data from brain regions selected a priori, and systemic prolactin release. Necessary first steps included estimating the magnitude and time course of prolactin response to anesthesia alone and to various doses of agonist. Kisspeptin (KP) and the KP receptor (Kiss1r) are integral to central regulation of the gonadotropic-axis. The demonstration that intravenous infusion of KP can stimulate gonadotropin secretion and ovulation in seasonally anestrous female sheep offers a means of manipulating the reproductive axis. However, KP may be of limited clinical use because of the short circulating half-life. Kiss1r agonists with increased half-life and similar efficacy to KP in vitro potentially may provide beneficial applications in breeding management of many species. The purpose of the study was to determine if preseason and postseason body fat percentages (BF%) change relative to playing time in Division I women’s basketball players. Subjects for the study included 11 National Collegiate Athletic Association (NCAA) Division I female collegiate basketball athletes over the age of 18 from a Midwest public university. Alcohol-induced neuroinflammation is mediated by proinflammatory cytokines, including IL-1beta. IL-1beta production requires caspase-1 activation by inflammasomes-multiprotein complexes that are assembled in response to danger signals. We hypothesized that alcohol-induced inflammasome activation contributes to increased IL-1beta in the brain. WT and TLR4-, NLRP3-, and ASC-deficient (KO) mice received an ethanol-containing or isocaloric control diet for 5 weeks, and some received the rIL-1ra, anakinra, or saline treatment. Inflammasome activation, proinflammatory cytokines, endotoxin, and HMGB1 were measured in the cerebellum. Expression of inflammasome components (NLRP1, NLRP3, ASC) and proinflammatory cytokines (TNF-alpha, MCP-1) was increased in brains of alcohol-fed compared with control mice. Increased …

Castration may detrimentally affect the health and performance of weaned calves, and painful procedures are increasingly becoming a public concern. The objective of this study was to determine the effects of castration (by banding) with or without administration of meloxicam, a non-steroid anti-inflammatory, on performance and inflammatory response in weaned beef calves. Mechanisms that govern anhydrobiosis involve the accumulation of highly hydrophilic macromolecules, such as late embryogenesis abundant (LEA) proteins. Group 1 LEA proteins comprised of 181 (AfLEA1.1) and 197 (AfLEA1.3) amino acids were cloned from embryos of Artemia franciscana and expressed in Drosophila melanogaster cells (Kc167). Confocal microscopy revealed a construct composed of green fluorescence protein (GFP) and AfLEA1.3 accumulates in the mitochondria (AfLEA1.3-GFP), while AfLEA1.1-GFP was found in the cytoplasm. In the presence of mixed substrates, oxygen consumption was statistically identical for permeabilized Kc167 control and Kc167-AfLEA1.3 cells. Rats' voluntary ethanol intake varies with dispositional factors and energy status. The joint influences of these were of interest here. We previously reported that rats selectively bred for high voluntary saccharin intake (HiS) consume more ethanol and express more robust conditioning of preference for flavors paired with voluntarily consumed ethanol than do low-saccharin consuming counterparts (LoS). Three new experiments examined the effect of refeeding after an episode of food restriction on ethanol intake and on preference for ethanol-paired flavors in HiS and LoS rats. Metabolic depression is typically correlated with extended survival of environmental challenge and energy-limitation in early life stages of various invertebrates and vertebrates. Diapause is an ontogenetically-programmed reduction of development and often metabolism seen in many invertebrates. When embryos of Artemia franciscana enter the state of diapause, the overall metabolic depression is estimated to be greater than 99%. These embryos also contain trehalose and express multiple isoforms of Late Embryogenesis Abundant (LEA) proteins, constituents often present in a number of such anhydrobiotic animals. Background: Hormones regulate the expression of multiple phenotypic traits. Therefore,

divergence in hormone concentrations may lead to evolutionary changes in the coordinated

physiological and behavioural traits that comprise an organism’s integrated phenotype. Adults

of marine ecotypes of threespine stickleback (Gasterosteus aculeatus) have higher concentrations

of the thyroid hormone thyroxine (T4) than adults of stream-resident ecotypes (Kitano

et al., 2010). Thyroid hormones are well-established mediators of osmoregulation and migratory

behaviours in fish, and the difference in T4 concentrations indicates that changes in thyroid

hormone signalling may underlie the evolutionary and ecological divergence of migratory and

non-migratory ecotypes.

It is generally accepted that taxa exhibit genetic variation in phenotypic plasticity, but many questions remain unanswered about how divergent plastic responses evolve under dissimilar ecological conditions. Hormones are signaling molecules that act as proximate mediators of phenotype expression by regulating a variety of cellular, physiological, and behavioral responses. Hormones not only change cellular and physiological states but also influence gene expression directly or indirectly, thereby linking environmental conditions to phenotypic development. Actinotroch larva (Phoronida) concentrate and collect food particles during their planktonic existance. Reported mechanisms of particle capture include both ciliary reversal and direct interception along the larval tentacles, and suction of particle-containing seawater into the larval vestibule. These diverse mechanisms explain collection of particles, but not subsequent ingestion. Particle delivery into the digestive system is accomplished, at least in part, by the activity of the esophageal cilia. If and how the beat frequency of esophageal cilia is modified by the presence or absence of particulate or dissolved organic materials remains unknown for actinotroch larvae.
